Harpagoside (HAR) is believed to be a main compound in Scrophularia ningpoensis which possess a broad of biological activities.Human serum albumin (HSA) has important physiological roles in transportation, distribution and metabolism of many endogenous and exogenous substances in body.It is great significance in pharmacology to investigate the interaction mechanism of HAR and HSA.In this work, the interaction between HAR and HSA was investigated by fluorescence and ultraviolet absorption spectroscopy at different pH (pH=4.0, 7.4, and 9.0) and temperatures (297, 310 and 323 K).The experimental results showed that the HAR could cause the fluorescence quenching of HSA through a static quenching procedure, showing that the HAR regularly quenched the intrinsic fluorescence of HSA, and a decrease in the quenching constant was observed with an increase in temperature.Under different conditions, all the magnitude of binding constants (KA) was larger than 105 L/mol and the number of binding sites (n) in the binary system were approximate to 1.Base on the magnitude of enthalpy and entropy changes, the negative values of ΔG, ΔH and ΔS revealed that the binding of HAR with HSA was spontaneous and exothermic process, and the main interaction forces of the HAR with HAR were van der Waals forces and/or hydrogen bonding interaction.The binding distance (r) between the HAR and HSA was calculated to be about 4.2 nm based on the theory of F(o)rster′s nonradiation energy transfer, which indicated that the energy transfer from HSA to HAR occurred with high possibility.What was more, the synchronous florescence spectroscopy confirmed the conformational changes of HSA during the binding reaction.
3.Clinical observation of comprehensive treatment for type Ⅲ A prostatitis

Objective To study the efficacy of comprehensive treatment for type ⅢA prostatitis.Methods One hundred and eighty-four patients with type Ⅲ A prostatitis, recruited to this study, were comprehensively treated for 8 - 12 weeks by oral antibiotics and α-1 receptor antagonist,indometacin suppository applied into rectal, prostate massage and psychological counseling. The clinical effects of the treatment were evaluated according to the NIH chronic prostatitis symptom index (NIH-CPSI) and leukocyte counts in the expressed prostatic secretions ( EPS ). Results Before and after the treatment, the NIH-CPSI scores were 28. 6 ± 6. 5 and 12. 9 ± 3. 8 ( t = 28. 3, P < 0. 05 ); the pain or discomfort scores were 14. 1 ± 3. 3 and 6. 4 ± 2.2( t = 26. 3, P < 0. 05 ), the urinary symptoms scores were 5.6 ± 1.8 and 2. 1 ± 0. 9 ( t = 23.6, P < 0. 05 ), the scores of life quality were 8.9 ± 3. 1 and 4. 4 ± 2.4 ( t = 15.6, P < 0. 05 ), the leukocyte counts were ( 24. 5 ±4. 4)/HP and ( 6. 2 ± 2. 7 )/HP ( t = 48.1, P < 0. 05 ) respectively, all comparisons showed significantly differences. Seventy-nine cases recovered completely, 57 cases recovered excellently, 36 cases recovered effectively and 12 cases did not recover, the overall effective rate was 93.5%. Conclusion Comprehensive treatment is an effective method for type Ⅲ A prostatitis.
4.Study on spatio-temporal pattern of mountainous Oncomelania hupensis snails at village scale in Eryuan County,Yunnan Province

Objective To develop a spatio-temporal model of mountainous Oncomelania hupensis snails based on the Bayes-ian model,and to analyze and identify the spatio-temporal pattern at a village scale. Methods The data including the intensity and spatial distribution of live and infected snails from 2000 to 2006 and the village boundary were collected. The independent and interactive spatio-temporal models were established,and then the best fitness model was selected to analyze the spatio-temporal pattern of live and infected snails. Results The interactive model of live snails and the independent model of infected snails were relative fitness models,and the models showed 95%CI(confidence interval)of the spatial and temporal coefficient included ze-ro,and indicated that the spatial and temporal correlation of live and infected snails was not significant at a village scale. Conclu-sion There is no significant spatial and temporal correlation of live and infected mountainous snails at a village scale,and the fur-thermore study should be carried out at a small scale.
5.Effect of sorafenib on lymphangiogenesis in subcutaneously transplanted human cholangiocarcinoma in nude mice.

OBJECTIVETo study the effects of sorafenib on lymphangiogenesis in transplanted human cholangiocarcinoma in nude mice.
METHODSThe model of transplanted human cholangiocarcinoma in nude mice was established by subcutaneous inoculation of cholangiocarcinoma cell line QBC 939 cells. Thirty-six nude mice were randomly divided into 3 groups after tumor formation: control group, sorafenib 30 mg × kg⁻¹ × d⁻¹ group and sorafenib 60 mg × kg⁻¹ × d⁻¹ group (n = 12 each), and then treated by gavage for 6 weeks. The tumor growth of the dose groups and control group was measured with calipers. Using immunohistochemical staining, the lymphatic microvessels at tumor edge were marked by LYVE-1 and counted. The expression of VEGFR-3 mRNA in paracancerous tissues was evaluated by RT-PCR.
RESULTSSorafenib significantly depressed the growth of cholangiocarcinoma. The inhibitory rate in the sorafenib 30 mg × kg⁻¹ × d⁻¹ group and 60 mg × kg⁻¹ × d⁻¹ group was 55.1% and 67.9%, respectively. The LMVDs of the control group, sorafenib 30 mg × kg⁻¹ × d⁻¹ group and 60 mg × kg⁻¹ × d⁻¹ group were 11.75 ± 3.19, 6.84 ± 2.18 and 5.03 ± 1.91, respectively. The LMVD of the control group was significantly higher than that in the dose groups (P < 0.01). The relative expressions of VEGFR-3 mRNA in the control group, sorafenib 30 mg × kg⁻¹ × d⁻¹ group and 60 mg × kg⁻¹ × d⁻¹ group were 2.158 ± 0.312, 1.027 ± 0.144 and 0.736 ± 0.149, respectively. The relative expression of VEGFR-3 mRNA in the control group was significantly higher than that in the dose groups (P < 0.05). No occurrence of lymph node metastasis was found in all groups.
CONCLUSIONSorafenib can significantly inhibit the growth of xenograft cholangiocarcinoma in nude mice. Sorafenib may reduce LMVD by down-regulation of the expression of VEGF-C/D and VEGFR-3 signaling axis.

In order to establish a method by which the recombinant suicide plasmids integrated on the chromosome could be recircled, A simple method of transconjugative cloning was established with the helper plasmids pMT999 or pRK2013 and fusion strains of Shigella flexneri which were obtained by screening with in vivo expression technology. And the cloning efficiency with this method is very high.

Objeetive To construct the cell DNA damage models for the human CML K562 cell line before or after imarlnib mesylate treatment and observe the repairing process dynamically for investigating the iniluence of imatinib mesylate on the repair function of K562 cells after cell DNA danlage.Methods The MTT assay was used to estimate the optimal pretreatment concentration of imatinib mesylate in K562 cells and Western blot was employed to evaluate the phosphorylation status in K562 cells after imatinib mesylate treatment to estimate BCR/ABL tyrosine kinase inhibition by imatinib mesylate.The comet assay was used to detect the DNA damage induced by hydrogen peroxide at various concentrations in K562 cells with or without the pretreatment of imatinib mesylate.A dynamic observation on the repairing process after cell DNA damage was made by the comet assay.Results The pretreatment by imatinib mesylate for K562 cells was optimized to be at a final concentration of 1 μmol/L for 24 h as revealed by the MTT assay additionly imatinib mesylate treatment at this concentration could effectively inhibit the phosphorylation of the BCR-ABL fusion protein at Tyr177(Deusityrate 0.100±0.018).When compared with the control group(Deusityrate 0.425±0.039),the BCR/ABL phosphorylation at Tyr177 was significantly decreased by (77. 11±5.59) % (t=4. 57,P<0. 05). The cell DNA damage models for both imatinib mesylate-nontreated and imatinib mesylate-pretreated K562 cell groups were constructed with hydrogen peroxide treatment at a final concentration of 10 μmol/L for 10 min at 4℃ as confirmed by the comet assay. When compared with the control imatinib mesylatenontreatod K562 cell group,the time duration required for the DNA repair in imatinib mesylate-pretreated K562 cell group was significantly prolonged (F= 97.79,P<0. 05 ). Conclusions The cell DNA damage models for the leukemic K562 cell groups before or after imatinib mesylate treatment were successfully constructed and the tyrosine kinase inhibitor imatinib mesylate for BCR/ABL fusion protein was revealed to attenuate the DNA repair capacity of the K562 cells after DNA damage.
