Objective:To explore the role and mechanisms ofω-3 polyunsaturated fatty acids (ω-3PUFAs) alone or in combination with dexamethasone (DEX) in inducing cell apoptosis and reversing drug resistance in multiple myeloma (MM). Methods:DEX-resistant MM cell line MM1R was treated with different concentrations of eicosapentaenoic acid (EPA) or docosahexaenoic acid (DHA) alone or in combination with DEX for 24 or 48 h. Cel proliferation was detected by MTT assay. Cel cycle and apoptosis were measured by flow cytometry. Expression levels of apoptosis-related proteins were analyzed by Western blot. Two-tailed, unpaired Student's t-test was used to compare the two treatment groups. A value of P<0.05 was considered statistically significant. Results:MM1R proliferation was inhibited by different concentrations (10, 20, 50, and 100μM) of EPA or DHA alone or in combination with 10μM DEX in a dose-and time-dependent manner. The inhibition effect was significantly higher in combinative groups than in single EPA or DHA treatment group (P=0.014, P=0.032). The percentage of G0/G1 phase and cell apoptosis rate in MM1R treated with different concentrations of EPA or DHA alone increased in a dose-dependent manner. This percentage was also significantly higher in the combinative groups than in the single EPA or DHA treatment group (P=0.015, P=0.004). The expression levels of cleaved caspase-3 and Bax were upregulated, whereas those of pro-caspase-3 and BCL-2 were downregulated in a dose-dependent manner. Drug resistance gradually decreased in MM1R cells at different concentrations of EPA or DHA with the increase of drug concentration. The reversal fold also increased gradual y, whereas the cel s decreased in the two drug-combination groups compared with the single-drug group. Moreover, the drug-resistance reversal index increased significantly. Conclusion:ω-3PUFAs can inhibit DEX-resistant MM cell proliferation, arrest cell cycle, and induce cell apoptosis.ω-3PUFAs also exhibit a synergistic anti-resistanteffect in combination with DEX. Furthermore,ω-3PUFAs can serve as novel effective drugs for MM treatment.

Objective To investigate the position sense in chronic non-specific low back pain (CNLBP) patients, and the correlation to pain and endurance of psoas muscles. Methods From January, 2014 to May, 2015, 30 patients with CNLBP and other 30 healthy subjects were measured spine flexion and extension position sense absolute error (AE) and psoas endurance with Biodex System 3 synchronously, and the patients were self-assessed the pain in lumbar with Numerical Rating Scale (NRS). The Pearson correlation coefficient of AE to pso-as endurance and NRS was calculated. Results AE of flexion and extension was both more in the patients than in the healthy (t>1.862, P<0.05), and the psoas muscle endurance was less (t=2.307, P<0.05). AE of flexion (r=-0.71) and extension (r=-0.47) negatively correlated with the psoas muscle endurance in the patients, and positively correlated with NRS score (r=0.78 for flexion, and r=0.49 for extension). AE mildly negative correlated with the psoas muscle endurance (r=-0.38 for flexion, and r=-0.29 for extension). Conclusion Spine sensation has been impaired in CNLBP patients, and may associate with pain and psoas muscles endurance loss.

AIM: To assess the impact of weaver gene on neuronal development, protein expression and vitality. METHODS: DNA encoding the wild-type and mutant ion channel was introduced into immortalized tyrosine hydroxylase-positive CNS-derived neurons named CAD(Cath.a-differentiated,a variant of Cath.a. Cath.a was established by targeted oncogenesis in transgenic mice) cells. DNA clone, immunostaining and Western blotting were used. Three different concentrations (0 25 mg/L, 0 5 mg/L, or 1 0 mg/L) of Girk2 and wv Girk2 expression plasmids were transfected into the CAD cells. The number of transfected cycling cells, protein synthesis and neurites growth were observed between two groups. RESULTS: The number of transfected cycling CAD cells with high concentration of wv Girk2 reduced to about 60%, compared to Girk2-transfected cells. Low concentration of wv Girk2 did not cause cell death but reduced the protein production of transfected genes. Neurite growth was also affected by wv Girk2. MK-801 and Kir2.3 altered the effect of wv Girk2. CONCLUSION: The results indicate that wv Girk2 functions as a blocker in weaver animals, which blockes the wv Girk2 channel to rescue the cells from death. Our data also suggest that the presence of channels and the level of wv Girk2 may have a significant impact on the fate of cells containing wv Girk2.

OBJECTIVE:
To investigate the value of indirect CT lymphography (CT-LG) combined with nanocarbon tracer suspension staining in the sentinel lymph node (SLN) detection of rabbit tongue (cancer).
METHOD:
Eight rabbits were divided into two groups, i. e. normal control group (6 rabbits) and VX2 tongue carcinoma group (2 rabbits). Pre-contrast CT images were taken for all rabbits. The mixture of iohexol and nanocarbon tracer suspension (iohexol 0.25 ml and nanocarbon tracer 0.25 ml, respectively, volume ratio = 1:1) was used as contrast agent of indirect CT-LG. Indirect CT-LG was performed as the contrast agent was injection into right ventrolateral submucosa of the tongues. CT images were acquired at 1, 5, 10 and 15 min after injection.
RESULT:
There were no cervical lymph nodes clearly identified in any group on the pre-contrast CT images. After the contrast agent injection, indirect CT-LG showed an ipsilateral SLN with rapid enhancement which was the only enhanced SLN, located lateral to the larynx-trachea region in all rabbits of VX2 control group and carcinoma group. The CT attenuation values of SLN were (58.80 ± 5.45) HU, (134.80 ± 28.92) HU, (91.00 ± 13.08) HU and (63.80 ± 4.32) HU at 1, 5, 10 and 15 minutes after indirect CT-LG. After the cervical anatomy guided by CT scan, all of the right deep SLN showed a distinct enrichment of black dye highly consistent with the locations on the indirect CT-LG images.
CONCLUSION
It is more accurate for sentinel lymph node detection by indirect CT lymphography combined with nanocarbon tracer staining.
