2.Diagnosis and treatment of iliac vein compression syndrome

The occurrence of iliac vein compression syndrome(IVCS) has the anatomic factor.IVCS has no specific symptoms and signs.The diagnosis of IVCS is mainly made by venography, intravascular pressure measurement, intravascular ultrasound, Doppler ultrasound,magnetic resonance venography, and CT.Before the occurrence of acute iliofemoral thrombosis,the treatment of IVCS is conservative therapy.The purpose of surgical intervention is to resolve the obstruction and keep the blood flow. The surgery of occluded iliac veins secondary to IVCS is now to be replaced by endovascular reconstruction. IVCS can be treated correctly before the occurrence of iliofemoral thrombosis and its sequelae can be reduced greatly, if the diagnosis of IVCS can be made as early as possible,and the degree of the stenosis of the iliac vein,the characteristics of its hemorheology and hemodynamics can be understood in time.
3.Inhibitory Effect of Co-Transfection of tPA Gene and PCNA-ASODN on Restenosis of Autograft Artery in Rabbits

Objective To observe the inhibitory effects of local co-transfection of tissue-type plasminogen activator(tPA) gene and proliferating cell nuclear antigen antisense oligodeoxynucleotides(PCNA-ASODN) on the intima proliferation and restenosis of autograft artery in rabbits. Methods One hundred and twenty male Zelanian rabbits were randomly divided into four groups(n=30, in each group): control group, PCNA-ASODN group, tPA group and tPA+PCNA-ASODN group. The left and right external iliac arteries (length 1.0 cm) were transplanted reciprocally. The transplanted arteries were respectively soaked in lipofection, PCNA-ASODN, pBudCE4.1/tPA and pBudCE4.1/tPA+PCNA-ASODN solution about 15 minutes. The transplanted arteries were sutured with 9-0 sutures soaked in PCNA-ASODN and pBudCE4.1/tPA solution. Each group were divided into five subgroups(n=6, in each subgroup) according to the sacrifice time (3 d, 7 d, 14 d, 28 d and 56 d after operation). On every sacrifice time point, the vascular specimens were harvested. The thrombocyte assembling and thrombus forming lining vessel wall were observed by scanning electron microscope. The pathological morphology of transplanted arteries were observed under microscope(HE). The intimal areas and stenosis ratio(%) of transplanted arteries were calculate and analyzed statistically among groups by computer system. The mRNA expression of tPA gene in transplanted ressel wall was detected with vevere transcription-PCR(RT-PCR). The number of PCNA positive cells in transplanted vessel wall was counted by SP immunochemisty. 4.The Effects of Spirulina on Intestinal Microflora of Diarrhea Model Mice

The effects of Spirulina on intestinal microflora of diarrhea model mice were observed.Diarrhea model mice were geted by gastric perfusion ampicillin,then these mice were given different doses spirulina by gavage.Selective culture media were used to culture bifidobacteria,bacterium lacticum,enteric bacilli,enterococci,bacteroid,clostridium perfringens in feces at different times.In order to find the difference of collecting samples from feces and intestine,the quantity of detection flora in different segment of intestine were compared too.The result showed that in middle dose group and high dose group spirulina can regulate and improve intestinal flora of diarrhea model mice,and effectively shorten time from disturbance to balance.Fecal sample and intestinal contents sample both manifest a same trend,that is the quantity of bifidobacteria,bacterium lacticum in treatment recovery group is significantly higher than saline group,and enteric bacilli,enterococci,bacteroid,clostridium perfringens is significantly lower than saline group,but each index flora in intestinal contents sample has a significant difference in quantity.
5.Inhibitory effects of local transfection of vascular endothelial growth factor 165 gene on intimal hyperplasia of artery in rabbits after operation injury

AIM: To observe the effects of local transfection of vascular endothelial growth factor 165 (VEGF165) gene on inhibiting intimal hyperplasia and restenosis of artery in rabbits after operation injury, and its possible mechanisms. METHODS: Microsurgery injury was used to establish the intimal injury model of right external iliac artery in rabbits. 105 male New Zealand rabbits were randomly divided into 3 groups (35 rabbits in each group). Group A was physiological saline control group, group B was pBudCE4.1-transfected group, group C was pBudCE4.1/VEGF165-transfected group. The physiological saline, pBudCE4.1 and pBudCE4.1/VEGF165 transfection solutions were injected into injured vessel walls of above-mentioned groups. The injured vascular specimen was harvested for pathologic examination, electric microscope observation, RT-PCR examining and immunohistochemical staining. 6.Establishment and Evaluation on Experimental Newborn Rats Model of Periventricular Leukomalacia

Objective To establish the 2 day-old SD rats model of periventricular leukomalacia(PVL).Methods Sixty-five healthy 2 day-old SD rats were randomly divided into 3 experimental PVL groups and 3 control groups.The experimental PVL groups were subjected to right carotid ligation(RCL),and then they were suffered from hypoxia by 6% oxygen and 94% nitrogen for 4 hours.Meanwhile sham surgeries were performed on control groups without exposed to hypoxia.Light and electronic microscopy were used to observe brain pathological changes.Immunohistochemistry methods were used to detect the distribution and expression of glial fibrillary acidic protein(GFAP),?-amyloid precursor protein(?-APP),myelin basie protein(MBP) and O4 of brain at 72 hours post-operation.Han-(ging) test,inclined plane test,open field test and cylinder test were performed on the rats 28 days post-operation.Results 1.In the PVL groups,light and electronic microscopy showed that tissue necrosis was observed in the periventricular white matter area at early stage,and at later stage right ventricular dilation,decrease of the corpus callosum area and loss of medullary sheath were detected.The morphometrical analysis showed that GFAP and ?-APP integrated optical density(A) of PVL group was increased,and mean diameter of GFAP-immunoreactive cells was also increased in PVL group,while MBP A of PVL group was decreased compared with contral group.The density of O4-immunoreactive abnormal cells was dramatically increased in PVL group.The outcomes of neurobehavioral tests of PVL group were greatly abnormal compared with control group.Conclusion The changes in 2-day-old SD rats,RCL-hypoxia model are accor-(ded) with the pathology and behavior characeriatis of PVL.
7.Effects of nitric oxide,endothelin-1, and liver sieve on biliary fibrosis in rats

Objective To study the effects of nitric oxide (NO) and endothelin-1(ET-1) on cholestasis liver fibrosis. Methods Forty-eight Wistar rats were randomily divided into 3 groups:the control group,the common bile dute ligation(CBDL)group,and the CBDL plus lactulose group.The animals were killed on the 3rd,7th,14th, and 21th days respectively to determine the contents of plasma endotoxin,NO,ET-1, and transaminases(ALT,AST).The degree of hepatic fibrosis was observed by microscopy and the ultrastructural changes of liver were observed by electron microscope. Results After common bile dute ligation,the contents of plasma ET-1 and NO increased significahtly,the plasma endotoxin level also increased and there were positive correlation between the levels of ET-1,and NO and the level of endotoxin.Plasma transaminases also increased greatly compared with those of the control group.The hepatic sinusoidal steonsis and widen Disse space were found.The diameter and number of fenestrate on hepatic sinusoid greatly decreased.Treatment with lactulose could partially alleviate all the pathological changes noted above. Conclusions NO and ET-1 may damage the "liver sieve"to cause the cholestasis liver fibrosis.
8.Diagnostic Value of X-Ray Computed Tomography on Neck Masses in Children

Objective To evaluate diagnostic value of X-ray computed tomography(CT) neck masses in children.Methods Clinic,pathologic diagnosis and CT scans of 26 patients from Jan.2004 to Jun.2006 with neck masses which location,density,edge and near construction were reviewed retrospectively.Results Seven cases in neck anterior area,13 cases in neck lateral area,6 cases in other areas or unconfirmed.Eight cases with lower density than cervical muscles,2 cases with same density,2 cases with higher density,14 cases with mixed density.Inflammatory masses were observed in 13 cases,congenital malformation in 11 cases,thyroiditis in 1 case,pharyngeal tumor-like proliferation in 1 case.Conclusion CT scan is the best choice of diagnosis of neck masses in children,and is valuable to diagnosis associated with history and physical examination.
9.CLONING OF NATTOKINASE GENE AND EXPRESSION IN E. COLI

In this study, nattokinase gene was amplified by PCR using bacillus subtilis chromosomal DNA as template and cloned into expressed vector pBV220. After transforming recombinant plasmid into E.coli HB101, the recombinant strain was yielded. It was proved that expression products was secretive and expression protein was 12% of total cell protein by SDS-PAGE. Optimum culture time and inducing time was determined as 6h and 5h respectively. The plasmid stability studies showed that recombinant plasmid has excellent segregational stability but the structural stability was not good in the host cell.
10.Research progress of growth factors in the pathogenesis and developments of myopia

Myopia is one of the most popular eye diseases all over the world. The development of the current understanding of its mechanism is still limited. Many studies indicated that the growth factors closely related to eye development and myopia. Some growth factors with biological activity, such as transforming growth factor ( TGF ), fibroblast growth factor ( FGF ) and epidermal growth factor ( EGF ), have an impact on scleral thickness variation, the regulation of the development of myopia and so on, which plays a non-negligible role in the pathogenesis of myopia. In this paper, the function of various growth factors in myopia will be reviewed.
