2.Dot Immuno-Gold Filtration Assay in the Diagnosis of Suspected Paragonimiasis and Evaluation of Chemotherapeutic Effect

Objectiv To evaluate the usefulness of dot immuno-gold filtration assay(DIGFA) for the diagnosis of Paragonimus infection.Methods During 2003 to 2006,72 cases suspected of paragonimiasis in Zhejiang Province were examined with DIGFA for rapid detection of specific antibodies against Paragonimus(Pw-DIGFA).The diagnosis was primarily established with the presence of antibodies,experience of ingesting raw freshwater crabs or crayfishes and clinical presentations.The cases were treated with praziquantel and followed-up at 3 and/or 6 months post-treatment.Antibody level in patients(pre-and post-treatment) were detected in parallel and analyzed comparatively by Pw-DIGFA and ELISA.Results The result of detection by Pw-DIGFA was in agreement with that of ELISA.28 of 72 cases were antibody positive and 44 cases were negative.Among the 28 positives,26 cases had a history of eating raw freshwater crab or crayfishes and the other 2 cases drank freshwater from brook before.21 cases showed paragonimiasis-related clinical symptoms such as low-grade fever,cough,or changes in image examination,while the other 7 cases showed only eosinophilia in peripheral blood(15%-70%).The mean absorbance values(A450) of positive sera,negative sera and normal sera tested by ELISA were 1.7361,0.2973 and 0.2657 respectively.There was significant difference between the positive cases and the negative cases(t=12.047,P0.05).At 3 month post-treatment,serum antibody in 5 cases whose clinical symptoms and physical signs relieved or disappeared decreased 2-5 titers and that of one case who relapsed with new signs increased by one titer.In Pw-DIGFA,the dot color of 5 cured cases showed a little weaker than that of pre-treatment and the relapsed case displayed similar response.At 6 month post-treatment,7 sera of clinically cured cases showed significantly weaker response than that of pre-treatment.The antibodies of those sera dropped 3-6 titers.Conclusion Pw-DIGFA is of supplementary value for clinical diagnosis of paragonimiasis.Antibody detection by pre-and post-treatment using Pw-DIGFA shows potential for the evaluation of therapeutic effect.

Although the promulgation of the law is important,the task of carrying out the law is more difficult. On the basis of concluding the problems in carrying out the food sanitation law and the analysis of its causes,it is significant to discuss the Food Safety Law from the viewpoint of Food Sanitation Law enforcement.
4.Effects ofEGbon expression of CREBandpCREBd in cortex of aging rats

Objective To investigate effects of Ginkgo biloba extract(EGb) on expression of CREB and pCREB in cortex of aging rats.Methods Male Wistar rats were divided into three groups:young control group,old control group and EGb group.Rats in EGb group were treated with intragastric administration of EGb,while rats in the other two groups were treated with distilled water.The spatial learning and memory were evaluated by Morris water maze,and the expression of CREB,pCREB were detected by western blot.Results( 1 ) Compared with young control group (9.6 ± 2.88,41.55 ± 6.30),the swimming time and times through platform in the target quardrant of rats in old control group(6.8 ± 2.49,34.92 ± 4.56) were reduced (P < 0.05 ).The times passing through the platform and the time exploring the target quadrant were more and longer in EGb group(9.4 ± 2.63,41.0 ± 6.68 ) than those in old control group(P < 0.05 ).(2)Compared with rats in young control group( 1.07 ±0.33,0.26 ± 0.04),relative contents of CREB and p-CREB proteins in cortex (0.70 ± 0.21,0.13 ± 0.05 ) weredecreased in old control group(P<0.05 ).CREB and p-CREB Levels were higher in EGb group ( 1.02 ±0.18,0.18 ±0.02)than those in old control group(P < 0.05 ).Conclusion EGb can ameliorate spatial learning and memory of rats by increasing the expression of CREB and p-CREB in cortex.
5.UPLC/Q-TOF-MS(E) based analysis of chemical composition of banxia xiexin decoction.

In order to clarify the chemical composition and source of Banxia Xiexin decoction quickly and comprehensively, whole and individual herbs of Banxia Xiexin decoction were analyzed by ultra-performance liquid chromatography with quadrupole-time-of-flight mass spectrometry (UPLC/Q-TOF-MS(E)). Under identical experiment conditions, chromatography results were compared between experiment groups. Based on the Q-TOF-MS(E) analysis, 74 peaks were identified on line. The herbal sources of these peaks were assigned. The results implied that flavonoids, triterpenoid saponins, alkaloids and glycosides were the main components in effective part of Banxia Xiexin decoction. The method established is simple and rapid for elucidation the constituents of Banxia Xiexin decoction and the results could be used for the quality control of Banxia Xiexin decoction.
6.The dynamic observations on the development of IgG/IgM antibodies before and after treatment of rabbit infection with Schistosoma japonicumusing different kinds of schistosomal antigens

To determine the diagnostic value as well as the evaluation value in therapy, the specific IgM and IgG anti-bodies of the infected and treated rabbits were detected using soluble egg antigen(SEA) and adult worm antigen(AWA) by ELISA. By using SEA to detect the IgM antibodies, the serum antibody level rapidly dropped 7 weeks after infection even without treatment, and those of IgM in sera detected by AWA rose early than IgG detected by SEA. After 5 month treatment, IgM detected by AWA and IgG detected by SEA were still positive. From these observations, it is evident that SEA is a better antigen to detect specific IgG in the diagnosis of schistosomiasis, AWA is a better one for early diagnosis. The use of AWA as antigen to detect IgM showed high diagnostic value both in acute and chronic schistosomiasis. However, detection of the IgM/ IgG in ELISA using SEA and AWA could not evaluate the therapeutic effect well.
7.Optimization of Extraction Process of Resveratrol from Polygonum Cuspidatum Based on Central Composite Design and Response Surface Method

This study was aimed to optimize the extraction process of resveratrol by central composite design (CCD) and response surface method (RSM), based on the result of single factor experiment. Resveratrol was extracted from Polygonum cuspidatum by water bath extraction of organic solvent. And content of resveratrol was taken as index in the investigation of solvent concentration effect, extraction time and solvent content on extraction process. The results showed that the optimum condition was 12.7 times amount of 72.0% ethanol and extract for 2 times under the temperature of 50℃, 1.1 h for each time; the average deviation between the maximum theoretical value and measured value was 0.83%. It was concluded that this extraction process was highly predictive, which provided experimental evidence for the industrial production of P. cuspidatum extraction.
8.Prosapogenin A inhibits cell growth of MCF7 via downregulating STAT3 and glycometabolism-related gene.

This study is to investigate the inhibitory effect and mechanism of prosapogenin A (PSA) on MCF7. MTT assay was performed to determine the inhibitory effect of PSA on MCF7 cells. PI/Hoechst 33342 double staining was used to detect cell apoptosis. RT-PCR was used to test the mRNA levels of STAT3, GLUT1, HK and PFKL. Western blotting was performed to determine the expression of STAT3 and pSTAT3 protein in MCF7 cells. The results showed that PSA could dose-dependently inhibit cell growth of MCF7 followed by IC50 of 9.65 micrmol x L(-1) and promote cell apoptosis of MCF7. Reduced mRNA levels of STAT3, HK and PFKL were observed in MCF7 cells treated with 5 micromol x L(-1) of PSA. PSA also decreased the level of pSTAT3 protein. STAT3 siRNA caused decrease of mRNA of GLUT1, HK and PFKL which indicated STAT3 could regulate the expressions of GLUT1, HK and PFKL. The results suggested that PSA could inhibit cell growth and promote cell apoptosis of MCF7 via inhibition of STAT3 and glycometabolism-related gene.
