2.Expression and significance of Cathepsin D and Laminin receptors in gastric cancers

Objective To investigate the elationship between the expression of Cathepsin D and Laminin receptors with the invasion and metastasis of gastric cancers.Methods Cathepsin D and Laminin receptors immunoreactivities were evaluated in 96 patients with gastric cancer using streptavaclin pemxidase method.Results Over expression of Cathepsin D in gastric cancer tissue was significantly related with the depth of invasion,lymphnode metastasis and grade of differentiation (P<0.01,P<0.01 and P<0.05, respectively).Moreover,the high expression of Lamninin receptors in gastric cancer had significant correlations with the invasion depth and grade of differentiation(P<0.05 and P<0.01);The positive expression of Laminin receptors were not significantly related with lymph node metastasis(P>0.05).By statistics analysis Cathepsin D had a significant positive correlation with Laminin receptors (P<0.05). Conclusion Cathepsin D and Laminin Receptors play all important role in the invasion and metastasis of gastric Cancers.
3.Bacteria Carried by Infected Abdominal Surgical Wound: An Investigation

OBJECTIVE To study drug resistance status of infected bacteria in abdominal surgical wound for their countermeasures. METHODS The abdominal surgical infection of a hospital was investigated from Jan 2000 to Dec 2004.Bacterial isolation and identification were carried out complying with National Operation Procedure of The Clinical Laboratory.Drug sensitivity test was performed with KB method. RESULTS The infection rate was 4.1%.The species were predominated by Gram-negative bacilli(71.1%),especially Escherichia coli(17.2%),Pseudomonas aeruginosa(16.7%) and Klebsiella pneumoniae(12.7%),and they all beared high antibiotics resistance. CONCLUSIONS Special attention should be paid to abdominal surgical wound infection.
4.Recent advances in DNA vaccines against allergic airway disease: a review.

DNA vaccine is used in infectious diseases initially, and later is applied in neoplastic diseases, allergic diseases and other fields with the further understanding of DNA vaccine and the development of genetic engineering. DNA vaccine transfers the genes encoding exogenous antigens to plasmid vector and then is introduced into organism. It controls the antigen proteins synthesis, thus induces specific humoral and cellular immune responses. So it has a broad application prospect in allergic diseases. Compared with the traditional protein vaccines used in specific immunotherapy, DNA vaccine has many advantages, including high purity and specificity, and improvement of patients' compliance etc. However, there are still two unsolved problems. First, the transfection rate of unmodified naked DNA plasmid is not high, Second, it's difficult to induce ideal immune response. In this study, we will review the progress of DNA vaccine applications in respiratory allergic diseases and its various optimization strategies.

5.Effects of Different Extraction Methods on Chemical Component Contents and Pharmacological Actions of Yangxue Ruanjian Prescription

Objective To establish a proper extraction method for Yangxue Ruanjian Prescription (YRP) by comparing the chemical component contents and analgesic and anti - inflammatory effect of extracts extracted from YRP by different methods. Methods Water - decocting, alcohol - refluxing and alcohol - percolating methods were adopted and component indexes of paeoniflorin and gentiopicrin were determined by HPLC. The anti - inflammatory effect of the extracts were observed on mice with xylol - induced auricular swelling and analgesic effect by hot - plate method. Results The content of gentiopicrin is similar in the extracts extracted by three methods and the content of paeoniflorin is higher in alcohol - percolating extract than in alcohol - refluxing extract, lowest in water - extraction. The analgesic effect of water - decocting extract is better than that of alcohol - refluxing extract, and that of alcohol - percolating extract is weakest; the anti - inflammatory effect of the three extracts is almost the same. Conclusion Water - decocting extraction is suitable to YRP, and this is determined by the characteristics of Chinese compound prescription.
6.Establishment of a hepatic carcinoma cell line with stable ClC-3 silencing by lentivirus-mediated RNA interference and its influence on invasion and migration

Aim To establish a hepatic carcinoma cell line with stable voltage-gated chloride channel 3 ( ClC-3 ) gene silencing through the lentivirus-mediated short-hairpin RNA ( shRNA ) method and investigate the effects of gene silencing on invasion and migration. Methods Three lentiviral vectors coding shRNA tar-geting ClC-3 gene were constructed, the recombinant plasmids were packaged into mature lentivirus by 293FT cells, and then the lentiviruses were harvested, concentrated and titrated. MHCC97H cells were infec-ted with the recombinant lentiviruses and then were se-lected to obtain cell lines stably expressing ClC-3 shR-NA. The efficiency of ClC-3 mRNA and protein ex-pression interference were determined by real-time flu-orescence quantitative PCR and Western blot, respec-tively. The effects of ClC-3 gene interference on inva-sion and migration of MHCC97 H cells were performed by Transwell chamber assays with or without Matrigel and cell scratch assay. Results The recombinant lentiviral vectors were successfully constructed and four lentiviruses were acquired after packaged by 293 FT cells. One negative control cell line and three cell lines with ClC-3 gene interference ( MHCC97 H/shClC-3-1 , shClC-3-2 and shClC-3-3 ) were successfully construc-ted after MHCC97 H cells were infected with lentivirus-es. The expression level of ClC-3 mRNA and protein in three ClC-3-silenced cells were obviously lower than the negative control cells ( P <0. 01 ) , MHCC97 H/shClC-3-2 cells showed the greatest inhibition of ClC-3 mRNA and protein expressions. As compared with the negative control cells, the ClC-3 gene interference sig-nificantly decreased invasion and migration of MH-CC97 H cells in vitro ( P <0. 01 ) . Conclusion The hepatic carcinoma cell lines with stable ClC-3 gene si-lencing were successfully established and the ClC-3 gene interference could significantly inhibit invasion and migration of MHCC97H cells.
7.Development in New Dosage Forms of Active Components from Ginkgo Biloba Extracts

The main active components in Ginkgo biloba extracts were Ginkgo biloba flavonoids and lactone compounds. This pa-per reviewed on the kinds and pharmacological effects of the active ingredients in Ginkgo biloba extracts, and focused on four aspects including controlled-release preparations, solubilized solid preparations, nanoparticle formulations and time- and site-specific formula-tions to introduce the development in the new dosage forms of Ginkgo biloba flavonoids and lactone compounds.
8.The messenger management on asthma control and pulmonary function in young and middle-aged asthma patients

Objective To investigate instant messenger management on moderate asthma control in young and middle-aged patients.Methods Sixty young and middle-aged asthma patients enrolled were divided into 2 groups by random number table,traditional management group (group A) and messenger management group (group B),and received the asthma treatment with traditional management or messenger management for 1 year.The compliance,control condition,pulmonary function and Saint George respiratory questionnaire(SGRQ) were compared between two groups.Results After one year management,there were 20 patients in proceeding of the standard treatment,15 total control,10 partial control and 5 uncontrolled in group A,while there were 26 patients in proceeding of the standard treatment,21 total control,7 partial control and 2 uncontrolled in group B.Compared with group A,the treatment adherence(x2 =6.331,P < 0.05),satisfaction degree (t =6.051,P < 0.01),asthma control rate (x2 =6.490,P < 0.05),efficacy satisfaction (t =6.051,P < 0.01) were all increased in group B.Forced expiratory volume in 1 second was elevated more significantly in group B than that in group A [(0.132 ± 0.175) L vs.(0.326 ± 0.125) L] (t =5.099,P< 0.01).SGRQ scores were decreased more obviously in group B than that in group A (t =4.316,P < 0.01).Conclusions The asthma control,respiratory and pulmonary function can be improved by messenger management in young and middle-aged patients.So promoting the messenger management model actively is advantageous to improve the level of prevention and treatment of asthma in China.
9.Effects of combination of laryngeal mask airway and epidural anesthesia on hemodynamics for hypertensive patients

10.Application of MR Imaging Technology in Articular Cartilage of Knee Joint

Objective To study the MR imaging technology and 3D reconstruction of the articular cartilage of knee.MethodsConventional,2D,3D and multiparameter MR imaging scans of knee were performed in ninety patients with trauma(n=34)andosteoarthritis(n=56) of knee and 10 healthy volunteers.The articular cartilages on 3D images were reconstructed using maximum intensity projection(MIP).The mean thickness of articular cartilage was measured and analysed statistically.Results Articular cartilages of knee were clearly depicted by MR imaging technique and 3D.On 2D FSE sequence,about 50% of articular cartilage appeared as three laminar apearance.On 3D imaging,the three-dimensional morphology of articular cartilage was showed clearly.Conclusion MR imaging and 3D reconstruction of the articular cartilages of knee is helpful in early diagnosis and treatment of disordered cartilages.
