1.Study on the Content Change of Glucose and Fructose in Different Solvents Extracts of Leaves of Lonicera Hypoglauca Miq

[Objective]To establish a method to determine the contents of glucase and fructose in extracts of leaves of Lonicera hypoglauca Miq. using HPLC-ELSD, and apply this method to study the content change of glucose and fructose in different solvents extracts in leaves of Lonicera hypoglauca Miq. [Methods] The chromatographic separation was achieved on Lichrospher NH2 column using a mobile phase composed of a mixture of acetonitrile and water(90:10) at a flow rate of 0.8mL·min-1 and the column temperature was 25℃.The ELSD drift tube temperature was set at 70℃ and the carrying gas was N2 and the flow rate of the gas was 1.6L·min-1. [Results] Glucose and fructose from extracts of the leaves of Lonicera hypoglauca Miq. could be wel separated, and the linearity ranges of glucose and fructose were shown at the concentration of 0.5840~1.168mg·mL-1 and 0.8256~1.8576mg·mL-1, the average recovery rate of glucose and fructose were 96.88%(RSD=1.11%) and 97.55%(RSD=1.15%), the stability of glucose and fructose was 2.12%and 0.38%, and the repeatability was 1.31% and 0.24%. Among the water extract,50% alcohol extract and 95% alcohol extract,the content of the glucose and fructose in the water extract was highest,which was 2.19mg·mL and 4.10mg·mL-1, the 50% alcohol extract was second, which was 1.13mg·mL-1 and 2.12mg·mL-1, and the 95% alcohol extract was lowest, which was 0.45mg·mL-1 and 0.65mg·mL-1.[Conclusion] The method is simple, accurate and has good repeatability, and can be used to determine the contents of glucose and fructose in the extract of leaves of Lonicera hypoglauca Miq. In this batch of leaves of Lonicera hypoglauca Miq., the contents of fructose and glucose were of water extract>50%ethanol extract>95%ethanol extract.
2.The spatial analysis of myocardium ultrastructure and observation of activity of mitochondrial enzymes

Objective To perform a spatial analysis of myocardium uhrastructure and the activity of mitochondrial succinate dehydrogenase in sub-acute Keshan disease. Methods Myocardium samples were collected from the cases with sub-acute Keshan disease and non-myocarditis(control), and their ultrastructure was observed under electron microscope. The density of mitochondrion volume and cristal membrane and its volume were measured by a point-counting method, while mitochondrion volume was estimated by water displacement method, succinate dehydrogenase activity of mitochondrion by iron-copper method in sub-acute Keshan disease and non-myocarditis cases. Results The volume ratio of mitochondrion to the cell on myoeardium [(47.79±6.20)%], the area ratio of mitochondrion to sarcoplasm [(55.06±6.50) %], mitochondrion to myofibrils [(1.43±0,41)%], mitochondrion section area[(0.78±0.15)μm2], and ratio of the lesion of cristal membrane area to the matrix area and mitochondrion volume[(67.14±13.96)%, (44.62±13.44)%]in sub-acute Keshan disease group were obviously higher than those in control [(33.20±7.62)%, (38.07±9.43)%, (0.71±0.33)%, (0.44±0.07)μm2, (14.11± 12.51)%, (9.34±11.28)%; t = 3.75,7.93,6.61,36.40,52.65,37.51, all P < 0.05]. The volume ratio of myofibrils to cell[(34.52±5.12)%]and the area ratio of cristae mitochondria to matrix[(32.43±14.42)%]in sub-acute Keshan disease group was obviously less than those in control [(48.51±4.30)%, (86.04±12.37)%; t = 9.85, 53.46, both P < 0.05)]. Succinate dehydrogenase activity was negative in sub-acute Keshan disease group. Conclusions Myocardium ultrastructure changes in sub-acute Keshan disease including the increase of volume and areas of mitochondria and the damage of the cristal membrane in mitochondria. Succinate dehydrogenase activity is decreased or even disappeared.

4.Follow-up study of discharged patients infected with novel recombinant avian-origin influenza A H7N9

Objective To analyze the laboratory results and chest CT appearances of novel recombinant avian-origin influenza A (H7N9) in discharged patients for understanding the imaging changes.Methods The clinical and imaging data of 4 patients with novel recombinant avian-origin influenza A (H7N9) were collected.The imaging changes and laboratory results were analyzed.Results All patients underwent chest CT examination 2 to 5 times from admission to discharge.The lesions primarily presented as ground-glass opacity (GGO) and mainly located in the inferior lobe of the left lung in 3 cases,in the inferior lobe of the right lung in 1 case,alternating between absorption and progress.GGO was absorbed obviously in about 3 days after treatment and consolidation was progressed after treatment.Consolidation absorption was observed with continue treatment in 1 case.The pulmonary lesions were absorbed obviously in 1 case 7 days after discharge.Synchronous reduction of the absolute values of CD3,CD8,CD4,CD45 were observed in 3 cases and the ratio of CD4/CD8 was normal.Conclusions (1) The areas of pulmonary involvement are large and variable in H7N9.The lesion area and imaging patterns are important for disease prognosis.(2) H7N9 virus infection may causes immunosuppression,decrease of CRP value can predict the improvement of the disease.

Objective:To investigate the expression of chemokine receptor in eupotic and ectopic endometrial tissues of women with endometriosis, and in endometrium of women without endometriosis.Methods:Normal endometrium, eutopic endometrium and endometriotic tissues were obtained from patients with endometriosis at laparoscopy. 7.Pathologic study on the perineural invasion in 513 cases of pancreatic cancer

Objectives To investigate the characteristics of neural invasion of pancreatic cancer as well as its relationship with other clinicopathological factors. Methods The neural invasion situation of 491 cases of ductal adenocarcinoma and other 22 pancreatic malignancies, 41 cases of benign tumor of pancreas and 21 cases of chronic pancreatitis was observed under light microscope, and its relationship with other clinicopathological factors was analyzed. Results The rate of neural invasion in ductal adenocarcinoma (74%) was much higher than in other types of pancreatic neoplasm (23% ,P < 0.01). Pancreatic ductal adenocarcinoma cell often invaded through peripheral nerve membrane into inner nerve fiber bundle, sometimes even invaded the whole cross-sectional nerve fiber. But neural invasion was not associated with differentiation of the tumor. The occurrence of chronic inflammation in the para-tumoral pancreas (52%) was also higher than that in other types of malignant (14%) or benign lesions (15% ,P <0.01). Lymphocytes neural invasion rate in pancreatic ductal adenocarcinoma was 65%, which were significantly higher than those in other types of malignant (36%) or benign lesions (22%, P < 0.01). Neural invasion rate in pancreatic ductal adenocarcinoma was associated with paratumoral chronic pancreatic inflammation and lymphocytes neural invasion, but not with lymph node metastasis. Conclusions Neural invasion was characteristic biological behavior in pancreatic ductal adenocarcinoma.

9.A retrospective study comparing endoscopic self-expandable metallic stents with surgery in the treatment of malignant obstructive jaundice

Objective To compare the efficacy and survival of patients with malignant obstructive jaundice using either endoscopic self-expandable metallic stents or surgery,and to evaluate the compounding factors influencing prognosis.Methods 56 patients with malignant obstructive jaundice treated with endoscopic self-expandable metallic stents (the endoscopic group) were compared with 90 patients who received surgery (the surgery group) during the same study period.Clinical data and survival of the 2 groups of patients were retrospectively analyzed.Results The success rate was 100% in the endoscopic group.The serum bilirubin,alkaline phosphatase (ALP) and γ-glutamyl transferase (γ-GT) decreased significantly by using either therapeutic endoscopy or surgery (P<0.01).There was no significant difference between the two groups in the reduction of serum total bilirubin.The mean survival of the endoscopic and surgery groups were 340 d and 795 d respectively.The accumulative survivals of the endoscopic group at 3,6 and 12 months as evaluated by the Kaplan-Meier method were 82.6 %,61.1% and 46.6 %,respectively,and for the surgery group were 97.0%,90.9 % and 65.4% respectively. There was a significant difference in survival between the two groups (P<0.01).Survival after therapeutic endoscopy was similar to surgery for patients with metastasis and hilar biliary obstruction.Conclusions Self-expandable metallic stents gave similar palliation in the relief of jaundice in patients with malignant biliary obstruction.The stents had no effect on the primary tumor.Therapeutic endoscopy with self-expandable metallic stents is a safe and effective method for the relief of jaundice in patients with obstructive jaundice caused by non-resectable malignant tumors.
10.CT features of pulmonary mycobacterial disease in patients with acquired immunodeficiency syndrome

Objective To study the CT features of pulmonary non-tuberculous mycobacteria (NTM) disease in patients with acquired immunodeficiency syndrome (AIDS) and explore the different CT appearances between AIDS-NTM and AIDS-TB.Methods CT findings of pulmonary NTM disease in 27 AIDS patients (NTM group) were retrospectively analyzed and compared with that of tuberculosis in 30 AIDS patients (TB group).The results were statistically analyzed using Fisher's exact test.Results CT findings of NTM appeared significantly more than that of TB as follows:high-density nodules (n =18 vs 1,P < 0.01),ground-glass opacities (n =10 vs 0,P < 0.01),fibrotic band (n =17 vs 3,P < 0.01),bronchiectasis (9 vs 2,P =0.012).CT findings of NTM appeared significantly less than that of TB as follows:miliary nodules (0 vs 6,P =0.016),air space consolidations (n =2 vs 11,P < 0.01),pleural effusion (n =1 vs 9,P < 0.01).Conclusion Nodule and fibrotic band companied with bronchiectasis were the main CT manifestations of pulmonary NTM disease in AIDS patients,while air space consolidation companied with pleural effusion and miliary nodules were the predominate CT findings of pulmonary tuberculosis in AIDS patients.
