1.Effect of active fractions from modified Simiao Wan on hyperuricemia and its mechanism

OBJECTIVE Tostudytheeffectofeffectivefractions(EFC)frommodifiedSimiaoWan (MSW)onthelevelofuricacidinhyperuricemicratsandinvestigatethemechanism.METHODS Two types of hyperurice mic models were established.A persistant hyperurice mic model was prepared by giving rats oxonic acid 200 mg·kg -1 and feeding the m with hypoxanthine.The models were ig given with modified Simiaowan (MSW)50 g·kg -1 or EFC 1 2.5,25 and 50 g·kg -1 consecutively for 5 d.The models were treated with MSW or EFC 50 g·kg -1 for 3 d.After the final treatment,the uric acid concen-trations in seru m and urine were determined by an auto matic bioche mistry analyzer.The activity of xan-thine oxidase (XOD )in the serum and liver was determined by enzymic colorimetric method.The activity of purine nucleoside phosphorylase (PNP)and uricase was detected by spectrophotometry. RESULTS Comparedwithnormalcontrolgroup,theserumlevelofuricacidinbothmodelgroupswas remarkably increased(P<0.01 ).Compared to model control group,MSW 50 g·kg -1 and EFC 12.5, 25 and 50 g·kg -1 significantly reduced the serum level of uric acid(P<0.05,P<0.01 ),but increased the activity of erythrocyte PNP(P<0.01 )in the oxonic acid potassium-induced hyperuricemia rats. MSW 50 g·kg -1 and EFC 50 g·kg -1 elevated the activity of liver uricase in the nicotinic acid-induced hyperuricemia rats(P<0.05).EFC 50 g·kg -1 also significantly decreased the serum XOD activity of hyperuricemicrats.CONCLUSION EFCsignificantlyinhibitstheserumlevelofuricacidinhyperurice-mic rats,which might involve down-regulation of protein levels of serum XOD to inhibit the production of uric acid and activation of uricase to pro mote the deco mposition of uric acid.
2.Establishment and Application of Zebrafish Auditory Evoked Potential(AEP) Detection System

Objective To establish an auditory evoked potential (AEP) detection system in zerbafish.Methods The AEP detection tank was designed and made, and then verified for its quality and reliability via four experiments: anesthesia experiment, swim bladder deflation, noise exposure and goldfish AEP test.Finally, zebrafish (total length form 10 mm to 46 mm) were determined using this system for AEP.Zebrafish randomly were divided into five groups according to total length (TL=12~15 mm, n=6;TL=17~20 mm,n=4;TL=22~26 mm, n=4;TL=32~37 mm, n=9;and TL=42~46 mm,n=12).Goldfish, as control group, were purchased for local petshop (TL=38~54 mm,n=8).Results The results of these four verifying experiments confirmed the biological, rather than artefactual, nature of the responses represented by the recorded waveforms.The AEPs were detected up to a much higher frequency limit (12 kHz) than previously reported.In this study, all fish demonstrated a range of hearing frequency from 100 Hz to 12 kHz without frequency expansion during development.The best hearing was observed at 600 Hz~1 kHz.The mean values of the frequency-averaged thresholds (mean SEM) were 141.7±1.32, 124.8±1.31, 121.8±1.49, 117.8 ±1.09 and 124.4±1.87 dB w, respectively, for the 5 TL groups.The AEP thresholds demonstrated both developmental improvement and age-related loss of hearing sensitivity.Conclusion An auditory evoked potential detection system of zerbafish has been established with stable performance and can be used for AEP detection of zebrafish.
3.Expression of Periostin in Craniopharyngioma Involving Third Ventricular Floor and its Relationship to Hypothalamus Functions

Objective To evaluate the expression and clinical significance of Periostin (POSTN) in craniopharyngi?oma. Methods Forty-two patients with pathologically diagnosed adamantinomatous craniopharyngioma involving the floor of 3rd ventricle were retrospectively analyzed. The correlation between expression of POSTN and outcome was studied. Results POSTN was expressed in stroma of craniopharyngioma tissues. The expression of POSTN was not associated with the postoperative recurrence (Z=-0.236,P=0.813) while was associated with the degree of the adhesion between tumor and floor of 3rd ventricle (r=0.685, P<0.001), postoperative Hypothalamic Status Scale (r=0.569, P<0.001) and Body Mass Index (r=0.594, P<0.001). Conclusions POSTN may predict the degree of the adhesion between craniopharyngioma and 3rd ventricle floor. The high expression of POSTN may indicate the worse hypothalamus function.

Objective:

Methods 52 patients with DEACMP were randomly divided into the observation group and the control group, 27 cases in the observation group and 25 cases in the control group. The 2 groups were adopted the treatment to improve the cerebral microcirculation and other symptomatic, the observation group on the basis of treatment for the treatment of oxiracetam Injection. Quantitative electroencephalogram (QEEG) and event-related potential P300 were used to evaluate the therapeutic effects of the 2 groups before and after treatment.

OBJECTIVETo identify methods in reducing the prevalence of lactose intolerance in children.
METHODSA hydrogen respiration test (HRT) method was used in screening lactose intolerance (LI) subjects after taking 25 g of lactose among 106 children aged from 10 to 11 years old in a primary school located in the suburban area of Beijing. A cross-design was used to detect the effects of low lactose milk, yogurt and cereal-effect among 68 selected LI children.
RESULTSThe incidence of LI was 80.2% after the children took 25 g of lactose, and after taking a 250 ml of full milk, lactase-fermented milk, coinfected milk, yogurt, or milk with meal, the LI incidences were 21.1% (12/57), 0% (0/25), 6.1% (2/33), 8.6% (3/35) and 13.6% (3/22) respectively.
CONCLUSIONLow lactose milks and yogurt could reduce the LI incidence among LI children significantly.

Objective:To discuss the diagnosis, treatment and outcome of patients with gliomatosis cerebri (GC). Methods:Retrospectively reviewed the clinical manifestations and radiological appearances of 6 patients with GC, which were diagnosed in our hospital between 1993 and 2003. We employed surgical treatment in three patients, stereotactic biopsy in two, and the other one received both biopsy and surgery. Results: The lesions of GC infiltrated more than two lobes in brain. CT studies showed diffuse hypodensity changes and enhancement was absent in four patients. MRI examinations revealed isointense or hypointense regions on T1WI, and uniformly high signal on T2WI. MRI also disclosed structural enlargement. All patients received radiotherapy after surgery and three patients underwent chemotherapy additionally. Five patients died during follow up with average course of 16.4 months. Conclusion:MRI examination is valuable in the diagnosis and the prognosis of GC is poor.
7.Mechanism of lncRNA CTD-3252C9.4 on invasion and migration of pancreatic cancer Panc-1 cells in vitro

The aim of this study is to investigate the inhibitory effect of long non-coding RNA(lncRNA)CTD-3252C9. 4 on migration and invasion of human pancreatic cancer cell Panc-1 in vitro and its mechanism. Panc-1 cells were stimulated by epidermal growth factor(EGF)in three-dimensional semi-solid system of cultured pancreatic cancer spheres. RT-qPCR was used to detected the transfection efficiency of lncRNA CTD-3252C9. 4. The effects of lncRNA CTD-3252C9. 4 and bone morphogenetix protein 7(BMP7)on the invasion and migration of Panc-1 cells were detected by scratch healing method and Transwell chamber method. The changes of target gene BMP7 and epithelial-mesenchymal transition(EMT)related proteins were verified by Western blot. EGF could significantly inhibit the expression of lncRNA CTD-3252C9. 4 in Panc-1 cells. The lncRNA can affect cells invasion and migration by inhibiting the transcription of the oncogene BMP7, then inhibit the process of EMT of tumors.
8.A three-year follow-up study on sera specific antibody in severe acute respiratory syndrome cases after the onset of illness.

OBJECTIVETo study the variation of specific antibody among convalescent of severe acute respiratory syndrome (SARS) patients through a three-year program.
METHODSSera samples were collected from SARS cases in the 5th, 20th and 35th month after onset of the illness. The SARS-CoV specific antibody was detected for all of them by ELISA and neutralized test simultaneously. The titer of neutralizing antibodies was calculated using Reed-Muench method, and the comparison between different time groups was analyzed regarding the variance of data on repeated measures after logarithm conversion.
RESULTS13, 17 and 13 sera samples were collected in the 5th, 20th and 35th month after onset. Results showed that despite the fact that the positive rates of ELISA antibody were 100%, 82.4% and 84.6% respectively,the neutralizing antibody was still positive for all the samples. The average neutralizing antibody titers were 1:43 (1:16-1:203), 1:36 (1:17-1:59) and 1:21 (1:10-1:39) on the 5th, 20th and 35th month after onset, and the differences were statistically significant (F = 60.419, P < 0.001). On the 35th month after the onset, 30.8% (4/13) of the patients were still having the neutralizing antibody level of above 1:36, but the neutralizing antibody level in another 30.8% (4/13) of the patients had decreased to as low as 1:10, when the cut-off level was set as 1:8.
CONCLUSIONResults of the study indicated that the neutralizing antibody of SARS cases could last for at least three years, but the sera specific antibody in SARS cases decreased gradually when time went by. However, neutralizing antibody in some of the cases decreased to a lower level on the 35th month. Further follow-up study was worthwhile to observe the long-lasting profile of antibody existence on SARS cases.

9.Effects of nutritional education on improvement of nutritional knowledge of infant's mothers in rural area in China.

OBJECTIVETo investigate the knowledge, attitude and practice (KAP) of young children's mothers on infant feeding and to evaluate the effects of nutritional education in the rural areas.
METHODSA cluster sampling method was used to select the local health station. Five hundred and fifteen mothers, who had infants with age of 4 - 6 months, were recruited for the questionnaire survey on the nutritional knowledge in rural areas of Tianjin municipality. The mothers were randomly divided into intervention group I (160), intervention group II (180) and control group (175). The mothers in the intervention group I were educated with feeding guideline on infants and young children and had had Group lectures and advisory from experts about maternal and child nutrition for teaching them how to feed their children; while, the mothers in the intervention group II were trained with feeding guideline on infants and young children by themselves; and the mothers in the control group received routine guidance at the local health station. The follow-up evaluation on nutritional knowledge of the mothers in each group was carried out after 3 and 6 months intervention, respectively.
RESULTSThe educational background had significant effect on KAP scores: KAP scores of the mothers with primary education or less (8.3 +/- 2.2) were significantly lower than that of the mothers educated with high school (9.4 +/- 1.6) and university (9.6 +/- 1.8) (LSD t = 3.70, P < 0.001). After being educated with feeding guideline on infants and young children, the knowledge of infant's mothers was greatly improved and KAP scores of the mothers after intervention were higher than that of the baseline (F = 183.556, P = 0.006); the percentage of correct answer on nutrition knowledge in the intervention groups was significantly higher than that of the control group. At six months of intervention, the KAP scores of intervention group I (12.0) and intervention group II (11.6) were higher than that of the control group (10.5) (LSD t = 5.96, P < 0.001; LSD t = 4.25, P < 0.001).
CONCLUSIONProviding nutritional and health education to the infant's mothers should be helpful for improving infant's feeding pattern and ensuring the adequate growth and development of infants.

10.Pathological study of cerebral amyloid angiopathy in rat models of Alzheimer's disease.

OBJECTIVETo examine cerebral pathologies in cerebral amyloid angiopathy in a rat model of Alzheimer's disease.
METHODSRat models of Alzheimer's disease was established by stereotactic Aβ1-42 fiber injection in the bilateral hippocampus. The cognitive function of the rats was evaluated with water maze test. HE staining, Congo red staining and double-labeling indirect immunofluorescence were used to examine the dynamic distribution of Aβ fiber deposit in the brain.
RESULTSThe model rats showed significant differences from the control rats in the escape latency and the times of crossing platform in waster maze test. HE staining revealed a decreased number and degeneration of the granular cells with increased glial cells in the model rats. Congo Red staining showed that the Aβ fiber was deposited gradually in the small vessels in the brain parenchyma to cause thickening, stenosis or occlusion of the small vessels. Immunofluorescence staining detected Aβ fiber migration from the parenchyma to the walls of the small arteries in the rat models.
CONCLUSIONCerebral amyloid angiopathy is a major pathological feature in Alzheimer's disease.
