1.Effect of hepatocyte cytochrome oxidase Ⅰ on nonalcoholic fatty liver in rats

Objective To study the expression and effect of hepatocyte mitochondrial DNA cytochrome c oxidase subunitsⅠ (COXⅠ) gene in rat nonalcoholic fatty liver model. Methods Forty Wistar rats were randomly divided into control group and high-fat diet group, and the high-fat diet group was subsequently divided into 4 subgroups (2, 4, 8 and 12 week) with 8 rats in each. Cytochrome c oxidase activity was determined by ultraviolet spectrophotometer. Meanwhile,the expression of hepatocyte COXⅠ antigen in rat nonalcoholic steatosis model induced by high fat diet were detected by Western blotting. Mitochondrial DNA COXⅠ mRNA were assayed by reversetranscription polymerase chain reaction(RT-PCR). Results The levels of activity in nonalcoholic steatosis rats induced by high fat diet at 2, 4, 8, and 12 weeks were (0.88?0.32), (0.76?0.37), (0.48?0.26), (0.39?0.21) nmol?mg~ -1 ?min~ -1 , respectively, significantly lower than (0.98?0.37) nmol?mg~ -1 ?min~ -1 in control group (P

RNA interference is a very important mechanism for the regulation of gene expression, participates in defense against viral infections, and keeps jumping genes under control. RNA interference has been taken as a powerful "gene silence" method widely used in basic science to study the function of genes, and it may lead to novel therapies in the future.
3.Meta-analysis of entecavir in lamivudine-refractory patients with chronic hepatitis B

Objective To evaluate the efficacy of entecavir in lamivudine-refractory patients with chronic hepatitis B.Methods Literature from Medline,Embase and CBM between January 2001 to December 2008 were reviewed.Acccording to the including and excwding criteria,four randomized controlled trials (RCT) were enrolled.The results of the trials were reviewed and analysed in software Revman 4.2.Results Patients in the entecavir group enjoyed significantly higher negative conversion ratios of HBV-DNA and ALT,and positive seroconversion of HBeAg:RR were 13.90(95% CI 6.39~30.24,P<0.00001),2.49(95% CI 1.40~4.45,P=0.002),and 3.53 (95% CI 2.85~4.38,P<0.000 01) respectively.There was a trend for the improvement of negative conversion ratio of HBeAg (RR=2.27,95% CI 1.00~5.15,P=0.05),but the incidence of side-effects of the drugs has no difference (RR=1.05,95% CI 0.97~1.14,P=0.21).Conclusions Compared with lamivudine or placebo,entecavir can significantly decrease HBV-DNA,improve liver function and positive seroconversion ratio of HBeAg without raising the incidence of drug side-effects.

Objective To discuss the application of visualization software of CiteSpaceⅢ to the treatment and research of transfusion medicine.Methods The software of CiteSpaceⅢ and the function of the reference database ISI Web of Science itself were used to the study.Results In the past 16 years,paper quantity and cited frequency on transfusion medicine re-search had the wave-like increasing tendency year by year.The research forces of the field were mainly distributed in Europe and the United States,the research hot spot and frontier around the blood safety,change over time in a dynamic develop-ment.Conclusion The study reveals the progress and development tendency of transfusion medicine,which could provide ef-fective reference for related research.
5.Knowledge requested by patients with coronary artery disease after stent implantation

Objective To survey the knowledge requested by patients with coronary artery disease (CAD) after stent implantation.Methods A survey on CAD and stent-related knowledge requested by CAD patients with stent implantation was conducted with a self-designed questionnaire from September 2013 to November 2013.Total 362 patients (235 males and 127 females) aged (67 ± 12) years participated in the survey in Yuetan Community Health Service Center and Fuxing Hospital in Beijing.Open questions and heuristic questions were used together during the survey.Results The knowledge requested by patients were:diet management(255,70.4%),side effects of medication(241,66.6%),management of physical activity (221,61.0%),prevention of CAD(181,50.0%),withdrawal of medication(176,48.6%),the life of stents (168,46.4%),post-operation check-up items (162,44.8%),the causes of CAD (156,43.1%)and drug mechanisms(134,37.0%).Patients with stent implantation less than one year requested more knowledge about the cause of CAD (x2 =37.4,P < 0.001),life-time of stent (x2 =39.9,P <0.001),prevention of CAD (x2 =13.4,P =0.001),post-operation check-up items (x2 =21.2,P =0.001) and requested less knowledge about withdrawal of medication (x2 =9.56,P =0.008),diet management (x2 =6.49,P =0.04) than those with more than one year.Patients aged < 70 years requested more knowledge about the cause of CAD (x2 =31.6,P < 0.001),prevention of CAD (x2 =9.99,P =0.007),life-time of stent (x2 =79.1,P < 0.001),side effects of medication (x2 =7.84,P =0.02) than patients aged > 70 years.Patients with education higher than junior high school requested more knowledge about the cause of CAD (x2 =26.6,P < 0.001),prevention of CAD (x2 =14.4,P =0.001) and life-time of stents (x2 =7.82,P =0.02) than those with education lower than junior high school.Patients with education higher than junior college requested more knowledge about the cause of CAD (x2 =26.6,P < 0.001) and post-operation check-up items (x2 =10.7,P =0.005) than those with education lower than junior college.Conclusion Some factors affect the knowledge requested by patients with coronary artery disease (CAD) after stent implantation,which should be considered in health education for CAD patients.
6.In vitro study on increase in radio sensitivity of renal cell carcinoma induced by ?-elemenen

Objective To study the effect and mechanism of increase in radio sensitivity of kidney cancer cells(GRC-1) induced by ?-elemenen in vitro. Methods GRC-1 cells were divided into 3 groups, blank group (added with 2 mL culture medium), emulsion group (added with 2 mL blank emulsion culture medium) and drug group (added with 2 mL 50 mg?L -1 ?-elemenen culture medium). After been cultivated for 24 hours, the cells were irradiated using 6MeV X-linear accelerator in different doses at the rate of 400cGy per minute. Number of cell clones was counted, and radiation-survival curve of GRC-1 cells was drawn. Flow cytometry (FCM) was used to measure cell cycle and apoptosis. Cells of climbing flake were dyed by immunocytochemical method, the gene expression of bcl-2 and PCNA was measured by imaging system. Results The cell cycle showed that the G 2M blocking caused by 50 mg?L -1 ?-elemenen was enhanced with time increase. It reached peak at 24 hours. FCM showed that the level of apoptosis increased with increase in drug dose and action time. The gene expression of bcl-2 was decreased by 20% in drug group than that in blank group, but there was no expression of PCNA in the two groups. Conclusion The radiosensitivity of GRC-1 cells can be enhanced by ?-elemenen. The mechanism of effect may be associated with the cell cycle blocking, inducing cell apoptosis and down-regulating expression of bcl-2 gene.
8.Effect of clinical nursing pathway on the mechanical ventilation effect of ARDS patients

Objective To investigate the effect of clinical nursing pathway on mechanical ventilation effect of ARDS patients. Methods 59 ARDS patients with mechanical ventilation from June 2008 to December 2010 were randomly divided into the observation group (30 cases)and the control group (29 cases). The control group used the traditional care model, conventional mechanical ventilation monitoring,treatment and care according to routine measures; the observation group used clinical nursing pathway. The mechanical ventilation time, complication rate of mechanical ventilation, and the gratification level of patients in the two groups of patients were observed. Results The mechanical ventilation time in the observation group was less than the control group, and incidence of complications was lower, and satisfaction degree of patients and their families was better, the difference was statistically significant. Conclusions The clinical nursing pathway can shorten the time of mechanical ventilation, lower incidence of complications of mechanical ventilation, and it improves satisfaction degree of patients and their families.
