Cancer is still one of the major diseases threatening human life and health. At present, how to achieve precise diagnosis and treatment of tumors is the biggest challenge in cancer treatment. Prodrugs use the tumor specificity of targeting molecules to deliver anticancer drugs to tumor sites, which can effectively improve drug bioavailability, therapeutic efficacy and safety, and are currently a hot spot in the research and development of anticancer drugs. The targeting molecules of prodrugs mainly include nucleic acid aptamers, polymers, antibodies, polypeptides, etc. Among them, polypeptides have the advantages of good biocompatibility, controllable degradation performance, high

Aim To study the effect of novel AChE inhibitors, 2-phenoxy-indan-1-one derivatives (YKY-1~7), against glutamatic acid-induced neurotoxicity in PC12 cells and on learning & memory impairment in dementia model mice induced by A?25~35 icv Methods The PC12 cells were preincubated with different concentrations of YKY-1~7 for 24 h and subsequently treated by glutamatic acid, at the high concentration of 2 mmol?L-1 for 15 min to induce cytotoxicity. The cell viability was assessed with MTT method.. Dementia model mice were made by intracerebroventricular injection (icv) of aggregated A?25~35. From the next day, the model mice were administered YKY-7 (2.5, 5, 10 mg?kg-1, ig) for 10 consecutive days and sham control mice or A? model control mice received daily ig saline. After the final treatment, the passive avoidance learning was tested, regional cerebral blood flow at cerebral cortex was assessed, and the activity of AChE in the cerebral cortex, hippocampus and blood serum were determined. Results Six out of the seven YKY compounds appeared to be effective against glutamatic acid-induced neurotoxicity in PC12 cells, with YKY-7 demonstrating the most activity. YKY-7 significantly ameliorated the learning and memory ability in dementia model mice induced by A?25-35 icv, slightly and selectively inhibited the cortical and hippocampal AChE, and gently increased the blood flow at cerebral cortex. Conclusion Some of 2-phenoxy-indan-1-one derivatives reported here have protective effects against glutamatic acid induced neurotoxicity in PC12 cells, and improve the learning and memory impairment induced by A?25-35, which may be partly attributable to its selective inhibition of AChE activity in the cerebral cortex and hippocampus.

Objective:To evaluate the effect of shikonin on the expression of RANTES and chemotactic activity of monocyte in endometriosis. Methods:Established SCID endometriosis models and cultured U937 cells were treated by a series of concentration of shikonin.RANTES transcriptive expression was determined by Real-time PCR,and RANTES secretion was determined by ELISA.Furthermore,Chemotaxis assay in vitro was conducted to elucidate the effect of shikonin on chemotaxis of U937 cells by RANTES.Results:Shikonin improved the RANTES transcription of human endometrium transplanted to SCID mouse(P

Objective To investigate the effects of rapamycin on p-JAK2,STAT3,p-STAT3 and PCNA in ranal tissue of IgAN rats. Methods The animal models of IgA nephropathy were devided into control group,IgAN model group, losartan group and rapamycin group. The clinical efficacy of rapamycin, and its influences on the protein and mRNA expressions of STAT3,p-STAT3,p-JAK2 and PCNA were determined by western blot,RT-PCR and immunohistochemical techniques respectively. Results The protein and mRNA expressions of p-STAT3,STAT3,p-JAK2 and PCNA were significantly increased in kidney of IgAN rats(P

Objective To investigate the association betw een systemic lupus erythematosus(SLE)and polymorphismof Fc gamma receptor ty peⅢin Han patients fromHunan province. Methods Genotypes of Fc?RⅢa-158V/F were determined by polymerase chain reaction(PCR)and restriction fragment length polymorphism(RFLP)analysis in 65patients with SLE and 60normal controls.Results①It was found that the frequency of homozygous Fc?RⅢa-158F /F genotype was significantly h igher in patients with SLE than that i n controls(OR=2.23,? 2 =4.69,P=0.03).②The frequencies of both homozygous Fc?RⅢa-158F /F genotype and Fc?RⅢa-158F allele were significantly high er in patients with lupus nephritis c ompared with those in controls(OR=2.67,? 2 =5.36,P=0.02;OR=2.00,? 2 =4.91,P=0.03).Conclusions These results suggest that an abnorm al distribution of Fc?RⅢa-158V/F polymorphism is associated with SLE in the Hans of Hunan province, and the presence of Fc?RⅢa-158F allele is a risk factor for lupus nephritis. These findings support t he hypothesis of a genetic mechanism in the pathogenesis of SLE.[

Neurology Asia 2017;22(3):235-241
Background & Objective: Sleep quality in neuromyelitis optica spectrum disorders (NMOSD) were
investigated in two recent studies. However, factors affecting sleep quality have not been studied in
NMOSD. This study aimed to investigate the prevalence of sleep disorders in Chinese outpatient clinics
with NMOSD and its clinical correlates. Methods: We administered Chinese validated self-questionnaires
on HRQOL (MSQOL-54), sleep (PSQI), pain (SF-MPQ-2), anxiety (HARS) and depression (HDRS)
to 42 patients followed up in our outpatient department. We assessed the relationships between sleep
quality with pain, anxiety, depression, gender, age, disability, disease duration, NMO-antibody status
and explored the determinants of poor sleep quality. Results: Sixty four percent of NMOSD patients
were poor sleepers. Significant correlations were found between duration, disability, pain, anxiety,
depression and sleep quality. Disability, depression and the domain of affective descriptors of pain
were the three main predictors of poor sleep in NMOSD.
Conclusion: This study reveals that poor sleep in NMOSD is common and it decreases physical function
of quality of life. It is worthwhile considering exploring adjuvant strategies aimed at controlling pain
associated affect, and treatment of depression may help to improve sleep quality in NMOSD.
