1.Preventive effects of soybean isoflavones on chronic allograft nephropathy in rats

Objective To identify whether soybean isoflavones plays a role in the prevention of chronicallograft nephropathy(CAN)in rats.Methods Forty-five male Lewis rats as receptors were randomly divided into 3 groups,receiving a diet with protein from high isoflavone soy protein fraction (HIS),low isoflavone soy protein fraction(LIS)or casein(CAS)starting one week before renal transplants from fisher donors,and continuing throughout the study.Serial serum and urine samples were collected at the beginning of the study,and at 4th,12th and 24th week.All rats were killed and the grafts were harvested in the 24th week.The functional and histopathogical changes of renal grafts were evaluated.Results In HIS group,arteria caudilis systolic blood pressure(SBP)value,24 h uri- nary protein(Upro)and serum creatitine(Cr)concentrations were significantly lower than those in LIS and CAS groups(all P
2.Clinicopathologic features of 9 cases of adult rhabdomyosarcoma

Objective·To investigate the clinicopathologic characteristics and prognosis of adult rhabdomyosarcoma (RMS).Methods·The clinical and pathological data,immunohistochemical test results of 9 RMS were analyzed retrospectively.Results·The information of 6 males and 3 females were collected in this research,whose average age was (46.89+20.09) years old.The tumors had a wide range distribution.The subtypes of histology included 6 cases of embryonal RMS,1 case of alveolar RMS,1 case of spindle cell RMS and 1 case of pleomorphic RMS.Immunohistochemically study showed that 6 cases were myogenin positive,4 cases were MyoDl positive and 8 cases were desmin positive.During the follow-up period of the 7 cases,only 2 patients survived free of tumors,2 patients presented with tumor recurrence and metastasis after the operations,and 3 patients died of tumor.Conclusion·In this study,embryonaI RMS is the most common type,and immunohistochemistry test is helpful to diagnosis of RMS.
3.Thoracolumbar disc herniation and Scheuermann's disease

Objective To explore the relationship between thoracolumbar disc herniation (TLDH) and Scheuermann1's disease (SD),as well as the role of SD in the etiology of TLDH.Methods From June 2006 to June 2010,45 patients with TLDH (T10-11-L2-3) underwent surgery in our department.Forty-five patients with lower lumbar disc herniation (LDH,L3-4-L5S1) acted as controls.The incidence of SD and Scheuermann's signs of these patients were examined by reviewing CT,MRI and Ⅹ-ray films.The thoracolumbar kyphotic angles of the two groups were compared.Furthermore,in TLDH group,the incidence of disk herniation within segments with the Scheuermann's signs was compared to that within segments without Scheuermann's signs.Results All except one patient in TLDH group(97.8%) had been associated SD while the incidence of SD in LDH group was only 26.7%.The incidence of all Scheuennann's signs was higher in TLDH group than that in LDH group.The average thoracolumbar kyphotic angle of TLDH group was 15.8°±6.9° while that of LDH group was 4.8°±4.0°.In TLDH group,the incidence of disc herniation within segments with Scheuermann's signs was all higher than that within segments without Scheuermann's signs.Conclusion There is a close relationship between TLDH and SD,suggesting that TLDH is probably a manifestation of SD.Schmorl's node,irregular end plate,wedge-shaped vertebra and especially,posterior bony edge separation,are associated with disc herniation.
4.The Applied Study of Postprocessing for ~(31)P MRS of Human Liver in Vivo

Objective To investigate the best protocol of liver 31 P MR spectroscopy . Methods The 31 P MRS was performed using 1.5T MR scanner,heart/liver surface coil and prone position with Resp/Trigger gate in the normal volunteer and the cases with liver diseases including hepatic cirrhosis, hepatocellular carcinoma and benige hepatic tumour. The age ranged From 15 to 58 years old. The related data were processed and compared using several kinds of protocol methods.Results By using the edit skills with Filter, Zero-filling and Curve fitting under the type of J-coupling peak, the curves corresponding with the request and precise data had been obtained for 31 P MRS of the liver. By the simple protocol methods, the 31 P MRS, concentration and ratio of the materials in the liver could be obtained in all cases.Conclusion The process of MR spectroscopy is easy to apply. The requested curves and the accurate data will be obtained by using the editing skill of edit.
5.Clinicopathological observation of 5 cases of primary liver adenosquamous carcinoma

Primary liver adenosquamous carcinoma is a rare kind of liver malignant tumor with very poor prognosis. Clinical data of 5 cases were analyzed retrospectively, four male and one female, whose average age was 60.2 years (50-67 years) old. Adenosquamous carcinoma contains both adenocarcinoma and squamous carcinoma histologically, and squamous carcinoma cells are dominant (>80%). Four cases underwent surgical resection and one received liver transplantation. The postoperative follow-up period ranged from 6 to 35 months and the mean survival time of these cases was about 15 months. Three patients died during the follow-up period. The survival rates 1 and 2 years after operation were 60% and 20%, respectively.
6.Influence of sirtinol in cell cycle of prostate cancer DU145 cells and its mechanism

Objective To observe the influence of sirtinol,a silent information regulator 1(SIRT1)inhibitor,in the cell proliferation, cell cycle progression and the expression levels of positive regulator proteins of the cell cycle including Cyclin D1,CDK4 and pRb in prostate cancer DU145 cells,and to explore the possible mechanism of SIRT1 in occurrence of prostagte cancer.Methods The DU145 cells at logarithmic growth phase were cultured in vitro and divided into control group(DMSO)and different doses (10,25,50μmol·L-1 )of sirtinol groups.The inhibitory rate of growth of DU145 cells was detected with MTT method,the SIRT1 mRNA and protein expression levels were determined by RT-PCR and Western blotting method, and the cell cycle was measured by flow cytometry.The Cyclin D1,CDK4 and pRb protein expression levels were examined by Western blotting method. Results Compared with control group, the inhibitory rates of growth of the DU145 cells in different doses of sirtinol groups were increased markedly in a dose-dependent manner(P<0.01),and the flow cytometry analysis result showed the DU145 cells at G1 phase were increased (P<0.01 ). Compared with control group, the expression levels of SIRT1 mRNA and protein in DU145 cells in different doses of sirtinol groups were decreased significantly(P<0.01);the expression levels of Cyclin D1 and pRb proteins were decreased(P<0.01),whereas the expression levels of CDK4 had no change(P>0.05).Conclusion SIRT1 inhibition by sirtinol can inhibit the cell growth of prostate cancer DU145 cells in a dose-dependent manner and arrest the cell cycle progression,and its mechanism may be related to decreasing the CyclinD1 and pRb protein expressions.
7.Effects of Paeonol on Aortic NF-κB Activity and Blood Lipid Levels in Rats with Atherosclerosis

Objective:To investigate the effects of paeonol on aortic nuclear factor-kappaB ( NF-κB) activity and blood lipid levels in the rats with atherosclerosis ( AS) . Methods:The AS rat model was established by administration of high-fat diet and vitamin D3 ( i. p. ). The rats were divided into 5 groups (n=8) randomly, namely the normal group, model group, positive control group(simvasta-tin, 10 mg·kg-1 ) and paeonol groups with the dose of 20 and 10 mg·kg-1 , respectively. After the 4-week treatment, the serum of rats was collected to determine the lipid levels. The aortic pathologic changes of AS rats were observed under a microscope, and then graded, and aortic NF-κB activity was detected by an immunohistochemistry method. Results:Administration of paeonol improved the severity of aortic pathology in AS rats. The pathologic grade in the paeonol group (20 mg·kg-1 ) showed a significant difference com-pared with that in the model group (P<0. 05). Paeonol (20 and 10 mg·kg-1) markedly decreased the TG, TC and LDL-C levels in the serum of AS rats (P<0. 05 or P<0. 01). Meanwhile, paeonol significantly inhibited the aortic NF-κB activity(IOD value) with (1. 96 ± 0. 55) and (2. 50 ± 0. 80) in 20 and 10 mg·kg-1 group, respectively, which showed significant differences compared with that in the model group (P<0. 05 or P<0. 01). Conclusion:Paeonol shows certain therapeutic effect in AS rats, and the mechanisms may be related with the regulation of lipid mobilization and the inhibition of aortic NF-κB activity.
8.Preliminarily analysis on traditional Chinese medicine advices in Treatise on Febrile Diseases.

To make a systematic analysis on literatures concerning traditional Chinese medicine (TCM) advices in Treatise on Febrile Diseases, and summarize the main connotations of traditional Chinese medicine advices, relevant TCM advices in Treatise on Febrile Diseases were collected, screened, compared, summarized and analyzed according to TCM dosage form preparation methods, TCM administration methods, medication contraindications and nursing after TCM administration. The literatures concerning medications in Treatise on Febrile Diseases were consulted, summarized and compared to standardize medicine advices and facilitate rational clinical application of TCMs. The standard medicine advices were as follows. The boiling water for TCMs shall be tap water and well water. The decoctions that have effects in promoting blood and meridians can be boiled with wine. The decoctions containing toxic components can be boiled with honey. Some TCMs shall be boiled with special methods, e. g. Herba Ephedra that could be boiled before other medicine and skimmed. Japonica rice could be added in decoctions to measure the duration of decoctions. Different dosages were required for different forms (litre, pill, medicine spoon). Administration times, temperature and frequency shall be adjusted according to target positions, functions and stage of illness. As for dietary contraindications during medication, thick porridges are recommended, where foods impacting medicine efficacy are prohibited. Regarding nursing after medication is important to recover physical functions, particularly warm porridges can go with diaphoretic recipes, while thick porridges can go with purgative recipes. And drug efficacies shall be defined by observing urine and excrements, and blood form. In conclusion, Treatise on Febrile Diseases is the first book that discusses TCM advices and records them in details. In this study, new standard medicine advices were proposed to provide important basis for improving clinical advices of TCMs and supports for developing the TCM dispensing technology.

10.Effect of Electro-acupuncture on Matrix Metalloproteinase in Degenerated Cervical Intervertebral Disc of Rats

To observe the effect of electro-acupuncture on matrix metalloproteinase in degenerated cervical intervertebral disc rats.Methods:The rat model of cervical intervertebral disc degeneration was induced by unbalanced dynamic and static forces,then the rats were randomly allocated to model group,electro-acupuncture groupⅠ(acupoints Dazhu(BL 11) and Tianzhu(BL 10) were electro-acupunctured),and electro-acupuncture group Ⅱ(acupoints Dazhu (BL 11),Tianzhu(BL 10)and Shenshu(BL 23) were electro-acupunctured),with the normal rats as control.then the HE staining method was adopted to observe the morphological change of cervical intervertebral disc and the immunohistochemical staining method was used to detect the marx metalloproteinases-1(MMP-1) and matrix metalloproteinases-3(MMP-3) in cervical intervertebral disc.Results:The morphological observation showed that after electro-acupuncture treatment,the degrees of cervical intervertebral disc degeneration in electro-acupuncture groupⅠand electro-acupuncture group Ⅱ were alleviated,and the electro-acupuncture group Ⅱ was superior to electro-acupuncture group Ⅰ.The expressions of MMP-1 and MMP-3 in degenerated cervical intervertebral disc were increased(P<0.01),and after electro-acupuncture treatment,the expressions of MMP-1 and MMP-3 in degenerated cervical intervertebral disc were decreased,especially the electro-acupuncture group Ⅱpresented the better effect as compared with electro-acupuncture group Ⅰ(P<0.05).Conclusions:Electro-acupuncture at acupoints Dazhu (BL 11) and Tianzhu (BL 10) had a certain therapeutic effect on cervical intervertebral disc degeneration,and in combination with acupoint Shenshu(BL 23),this therapeutic effect could be enhanced,its action mechanism might be that electro-acupuncture can reduce the expressions of MMP-l and MMP-3 in degenerated cervical intervertebral disc,inhibit the degradation of matrix in intervertebral disc.so as to relieve cervical intervertebral disc degeneration.
