6.Effects of overexpression of NADH kinase gene on ethanol fermentation by Saccharomyces cerevisiae.

Glycerol is the main byproduct in ethanol production by Saccharomyces cerevisiae. In order to improve ethanol yield and the substrate conversion, a cassette about 4.5 kb for gene homologous recombination, gpd2Δ::PGK1(PT)-POS5-HyBR, was constructed and transformed into the haploid strain S. cerevisiae S1 (MATa) to replace the GPD2 gene by POS5 gene. The NADH kinase gene POS5 was successfully over expressed in the recombinant strain S. cerevisiae S3. Comparing with the parent strain, the recombinant strain S. cerevisiae S3 exhibited an 8% increase in ethanol production and a 33.64% decrease in glycerol production in the conical flask fermentation with an initiatory glucose concentration of 150 g/L. Overexpression of NADH kinase gene seems effective in reducing glycerol production and increasing ethanol yield.

7.Effects of Four-week Intermittent Fasting on Skeletal Muscle Mass and Autophagy in Rats

Objective To observe the changes of body weight,fat mass and skeletal muscle mass of rats after 4 weeks of intermittent fasting,and explore relationship with autophagy in skeletal muscle,so as to provide theoretical basis for intermittent fasting.Method Twenty male Sprague Dawley rats were randomly divided into a control group (Con) and an intermittent fasting group (IF),each of 10.The rats of IF group were forbidden to eat food every Wednesday and Friday,and the body weight of both groups was recorded weekly.After 4 weeks,Dual-Energy X-Ray Absorption (DEXA) was used to analyze the body fat mass,then the bilateral soleus was separated to record the wet weight and measure the cross-sectional area of the soleus fibers by testing laminin with immunofluorescence confocal laser scanning microscope.The form of autophagic vacuole of soleus was observed using a transmission electron microscopy.The expression of autophagy-related protein LC3,p62 and regulating protein AMPK,pAMPK and ULK1 were measured using Western blotting.Result After 4 weeks of intermittent fasting,the weight and fat mass of IF were significantly lower than those of Con (P<0.01),but there were no significant differences between them in wet weight and cross-sectional area of soleus (P>0.05).The expressions of AMPK,p-AMPK,ULK1 in IF were significantly higher than those in Con (P<0.01).Compared with Con,the expression of LC3-Ⅱ/LC3-Ⅰ of IF increased significantly,while the expression of p62 decreased significantly (P<0.01).Conclusion Four weeks of intermittent fasting decreases the fat mass significantly,and control the weight efficiently.Intermittent fasting can maintain the skeletal muscle mass by promoting moderate autophagy through the AMPK-ULK1 pathway.It should be a potential lose weight method for further research.
8.The suppressive effect of bone marrow stromal cells modified by CTLA4Ig-gene on lymphocyte activation

Objective To investigate the suppressive effect of bone marrow stromal cells modified by CTLA4Ig-gene on lymphocyte activation. Methods The expressions of CTLA4Ig-gene at transcription level and protein level were assessed by RT-PCR, immunohistochemical techniques and Dot-ELISA. The suppressive effect of CTLA4Ig on lymphocyte activation was investigated in MLR system through MTT and ELISA which quantified IL-2 level. Results The transcription of CTLA4Ig-gene could be detected by RT-PCR in the transfected group and in the passage number two. Immunohistochemistry indicated that CTLA4Ig expression was diffusely located in the cytoplasm of BMSCs. Dot-ELISA could detect the expressed CTA4Ig in the culture medium at 12h and even in culture medium of the passage number l after transfection. MLR system showed that CTLA4Ig had significant inhibition on lymphocyte proliferation and IL-2 secretion. Conclusion Ad-CTLA4Ig could transfect BMSCs efficiently. Morever, CTLA4Ig can suppress lymphocyte activation and IL-2 production.
9.Clinical analysis of cellular schwannoma in the gastric wall misdiagnosed as gastric stromal tumor

Cellular schwannoma is a rare benign tumor of the peripheral nerves.The tumor is consist of spindle cells which arranged in interwoven bundles,and lacking the mesh district and palisading structure.The tumor cells are abundant,with coarse chromatin,deeply stained nuclei and in multiforms.So the tumor may easily be misdiagnosed as various types of spindle cell sarcoma.Intraperitoneal cellular schwannoma may also be mistaken as gastrointestinal stromal tumors.In this article,the clinical characteristics,diagnosis and treatment of cellular schwannoma were analyzed based on the clinical data of a patient with cellular schwannoma in the gastric wall misdiagnosed as gastric stromal tumor.
10.Analysis on changes before and after treatment of adult angle class Ⅱ division Ⅰ malocclusion patients accompanied with temporomandibular joint disorders

Objective compare the changes in condyle positions, face type lateral position and facial soft tissue before and after clinical correction of adult ClassⅡ Division Ⅰ malocclusion patients accompanied with the temporomandibular disorder. Method there were 7 adult Class Ⅱ Division Ⅰmalocclusion patients with accompanied with temporomandibular disorder, they accepted orthodontic extraction treatment routinely with MBT straight wire appliance technology, by which the occlusion was opened, the front teeth was received within correction, the occlusion was finely adjusted to canine fossa staggered relationship and the overbite cover was proper. Results The face type facial soft tissue profile was changed from raised face before treatment to straight face type and 1/3 of height under the face was increased and the temporomandibular disorder was made better. Conclusion the molars occlusion relationship of adult Class Ⅱ Division Ⅰ malocclusion patients accompanied with temporomandibular disorder was adjusted by opening the occlusion and adducting the upright front teeth , the condylar process was moved forward , the posterior teeth alveolar bone height was increased and the temporomandibular disorder was substantially improved.
