1.Efficacy Observation of Ultrasonic Conductometric Acupoint Penetration of Wenhua Zhitong Recipe for Adenomyosis Patients.

OBJECTIVETo observe the efficacy of ultrasonic conductometric acupoint penetration of Wenhua Zhitong Recipe (WZR) in treating adenomyosis.
METHODSTotally 40 adenomyosis patients with Shen-deficiency blood stasis syndrome were randomly assigned to the treatment group and the control group, 20 in each group. Patients in the treatment group received ultrasonic conductometric acupoint penetration of WZR from the 2nd day before menstruation to the end of menstruation, while those in the control group took WZR. All patients received 3 consecutive menstrual cycles. Endometrial contents of cyclooxygenase-2 (COX-2) and oxytocin receptor (OTR) were determined using SP immunohistochemical staining method before and after treatment. Clinical effect, efficacy of Chinese medical syndrome, changes of uterus volume, and contents of carbohydrate antigen 125 (CA125) were observed before and after treatment.
RESULTSThere was no statistical difference in the total clinical effect between the two groups (P = 0.357). Efficacies for relieving abdominal pain and lumbar soreness during menstruation were better in the treatment group than in the control group (P < 0.05). Uterus volume, endometrial levels and integrals of COX-2 and OTR decreased in the two groups after treatment (P < 0.05, P < 0.01).
CONCLUSIONUltrasonic conductometric acupoint penetration of WZR and oral administration of WZR both could effectively improve adenomyosis patients' symptoms such as Shen deficiency, and blood stasis, reduce the foci of uterus, and relieve the pain.

2.Multicystic renal cell carcinoma:a clinicopathologic study with comparison of it with renal clear cell carcinoma undergoing cystic change

Objective Multicystic renal cell carcinoma(MCRCC)appears to be a distinct subtype of renal cell carcinoma with characteristic gross and microscopic features.To explore the clinical and pathological features of MCRCC in order to find out its difference from renal clear cell carcinoma undergoing cystic change(RCCCC),and to propose prognosis criteria for therapeutic modalities.Methods From 1999 to 2003 32 patients were identified to have renal carcinoma,and they received radical or partial nephrectomy in the authors' institution.The pathological diagnosis was either MCRCC(n=21)or renal cell carcinoma associated with features of cystic changes(n=11).All specimens were reviewed histologically by two independent pathologists.The clinicopathologic and follow-up prognosis data on MCRCC and RCCCC were collected for study.Results 21 patients with MCRCC were 17 men and 4 women.The mean age at the time of surgery was 49 years(range 37 to 67 years).15 MCRCC patients with follow-up data were still alive with a mean follow-up time of 61 months(ranged 31 to 73)and no evidence of recurrence and metastasis was discovered.The mean survival time for 7 of the 11 patients with RCC was 32 months(ranged 18 to 63).There was significant difference in survival time between the two groups(P
3.Study on in vitro Antimicrobial Activity of Humulus Water Extract

Objective:To study the in vitro bacteriostasis of water extract of humulus. Methods:By using double dilution method and 96-well plates trace broth dilution method, the inhibitory test of water extract of humulus was conducted on Staphylococcus aureus, Escherichia coli and Pseudomonas aeruginosa. The minimum inhibitory concentration ( MIC ) , minimum bactericidal concentration ( MBC) and the diameter of inhibition zone were determined. Results: The drug showed different sensitive degree on the test strains. MIC and MBC for Escherichia coli were 31. 25 mg·ml-1 , that for Staphylococcus aureus were 3. 91 mg·ml-1 , and that for Pseudo-monas aeruginosa was 125 and 250 mg·ml-1 . Conclusion:With the water extract of humulus as the research object, the study on the in vitro bacteriostasis effect of humulus scandens is closer to the clinical application form, which provides reference for the further re-search on the related preparation of humulus scandens.
4.Extraction Technology for Antitumor Active Components in Fructus Ligustri Lucidi

Objective:To optimize the extraction process of antitumor active ingredients in fructus ligustri lucidi. Methods:Using fructus ligustri lucidi medicinal herb as the raw material, the extraction process of luteolin and pelargidenon in the herb was screened by D-optimal design. The extraction yield of different extraction processes was compared to obtain the optimal extraction conditions. Re-sults:The best extraction conditions were as follows:adding 12-fold 95% ethanol, extracting 2 times with 60 minutes for each time. Conclusion:The optimal extraction technology can provide significant reference for the further pharmacodynamic effect development of fructus ligustri lucidi.
5.Relationship between Caspase-3 and Apoptosis in Aseptic Necrosis of Femoral Head

Objective To investigate the expression and role of Caspase-3 during the development of non-traumatic avascular/aseptic necrosis of femoral head(ANFH).Methods Tissue samples were obtained from 22 femoral heads of 21 patients with ANFH.The samples were divided into three groups:early stage(6 cases),middle stage(8 cases) and late stage(8 cases) groups according to the progress of the disease.Meanwhile,19 tissue samples of 19 femoral heads were obtained from 19 cases of acute femoral neck fracture and set as a control group.The level of apoptosis of each group was compared in terms of the percentages of TUNEL positive osteocytes and empty osteocyte lacuna,and then the activity of Caspase-3 was detected by colorimetric method.Results Both the pecentages of empty osteocyte lacunae(F=45.43,P=0.000) and the TUNEL positive osteocytes(F=120.86,P=0.000) increased significantly with the progress of ANFH.The two indexes in the early and middle stage groups were significantly higher than those in the control(q=18.899,P0.05).Caspase-3 activity was not significantly related to the capacity of osteogenesis(r=0.126,P=0.425).Conclusions Caspase-3 is not the major executor of apoptosis in ANFH.It is very likely that it does not play a nonapoptotic role in the progress of the diseases.
6.The relationship between expression of PIAS3 and malignancy of human brain meningioma

Objective To explore the relationship between PIAS3 expression and the degree of malignancy of meningiomas.Methods 72 cases of different degree of malignant meningioma and 13 cases of surgical specimens of normal brain tissue were extracted the total RNA,and detected the levels of PIAS3 in different degree of meningioma and normal brain tissue through comparative control study by using real-time fluorescence quantitative PCR.Results PIAS3 was low expressed in normal brain tissue,however,high expressed in the human brain meningioma with significant differences(P<0.01).There were also significant differences found in expression level of PIAS3 between meningiomas grade I and Ⅱ,or Ⅲ level(P<0.05),but the differences between meningiomas grade Ⅱ and Ⅲ were not obvious(P>0.05).And PIAS3 expression was not related with the depth of infiltration in invasive meningiomas(P>0.05).Conclusion PIAS3 was overexpression in meningiomas,and was be closely related to the occurrence and development of meningiomas.
7.Effect of Polysaccharides from Radix Saplshnikoviae on the Immune Factors in Rats with Allergic Rhinitis

Objective: To investigate the effects and underlying mechanism of polysaccharides from Radix Saplshnikoviae on the immune factors in rats with allergic rhinitis induced by OVA.Methods: Firstly, 0.3mg OVA, 30mg Al(OH)3 and 1ml saline were mixed and intraperitoneally injected for the initial immunization, and then 200μg OVA (4%) was given once a day by nasal dripping since the 15th day for the second immunization to establish the allergic rhinitis model.Fifty SD rats were randomly divided into the allergic rhinitis (AR) model group, the polysaccharides from Radix Saplshnikoviae (600 mg·kg-1, 300mg·kg-1, 150 mg·kg-1) groups, the positive (Biyankang, 400 mg·kg-1) group and the normal control group.After the 14-day intragastric administration, blood was collected from abdominal aorta and serum was obtained.The levels of immune factors and inflammatory factors in serum were examined by radioimmunoassay.Results: The positive group and the polysaccharide groups could inhibit the typical symptoms of allergic rhinitis including sneezing, nasal scratching and running nose in varying degrees, and alleviate the inflammatory reaction symptoms.After the drug intervention, the positive group and the polysaccharide groups could down-regulate the expression of immune cell factors IL-4, TNF-A, VCAM-1 and IL-5, enhance IL-12 and INF-γ expression levels, and suppress the generation and release of inflammation factors IgE, HA, LTC4 and PGD2, and the differences were significant when compared with the model group (P<0.05).The dose-effect relationship was not too obvious among the polysaccharide from Radix Saposhnikoviae groups respectively at high, medium and low dose, and the high dose group was best among them.Conclusion: The polysaccharides from Radix Saplshnikoviae exhibit curative effect for AR, which can down-regulate the serum levels of IL-4 and IL-5, up-regulate the serum levels of IFN-γ and IL-12, adjust Th1/Th2 lymphocyte subsets balance and body immune response and reduce nasal mucosa allergic inflammation, and the underlying mechanism may be related with reducing the generation and release of immune factors in AR induced by OVA.

Objective To investigate the relationship between the expression of Fas, FasL, Bcl-2 and Bax in transitional cell carcinoma of bladder and its clinical significance. Methods Immunohistochemistry method was used to detect the expression of Fas, FasL, Bcl-2 and Bax in 40 specimens of transitional cell carcinoma of bladder and 24 specimens of normal bladder mucosa tissue. 9.The clinicopathological analysis of 16 cases of sarcomatoid renal cell carcinoma

Objective To study the pathological features and clinical significance of sarcomatoid change of renal cell carcinoma. Methods The specimens of 16 cases of sarcomatoid renal cell carcinoma admitted to our hospital were reviewed, and stress of observation was put on the the morphology and the number of the sarcomatoid components, and then their relation with clinical course, staging, and prognosis was retrospectively analyzed. Results Among the 16 cases of renal cell carcinoma with sarcomatoid transformation, 9 cases were conventional clear cell carcinoma, 3 cases were papillary renal cell carcinoma, 2 cases were chromophobic cell carcinoma, and 2 cases were collecting tubule carcinoma. In 9 cases the sarcomatoid elements were over 50% of total tumor area, while in the other 7 cases the sarcomatoid elements were less than 50%. The TNM stage among the 9 cases with more than 50% sarcomatoid elements were T2 (1/9), T3 and T4 (8/9). The TNM stage among the 7 with less than 50% sarcomatoid elements were T2 (4/7), T3 and T4 (3/7). The sarcomatoid area resembled malignant fibrous histiocytoma or fibrosarcoma. The average survival period of the patients was 19 months (8-154). Cases with over 50% sarcomatoid elements survived for 14 months in average, while the cases with less than 50% sarcomatoid area survived for 27 months in average. The difference was significant (P

Objective: To investigate the effects of fibrin glue(FS) and glutamine(Gln) on the healing of colon anastomoses after immediate postoperative intraperitoneal administration of 5-fluorouracil.Methods: Thirty two female Wistar rats were randomized into four groups.After resection of a 1-cm segment of the transverse colon,an end-end sutured anastomosis was performed.Then the rats were divided into control group,5-FU group,FS group,and Gln group.All rats were killed on the 8th postoperative day and the anastomoses were examined microscopically.The bursting pressure measurements were recorded and anastomoses were graded histologically.Results: The adhesion formation score was significantly higer in the rats of the 5-FU group than in the other groups.Bursting pressures were also significantly lower in the 5-FU group than in the other groups.Conclusion: The immediate postoperative intraperitoneal administration of 5-FU inhibited wound healing.However,FS and Gln can improve early healing of colon anastomoses after immediate postoperative intraperitoneal administration of 5-fluorouracil.
