1.Normal Values of Auditory Brainstem Responses for Infants of Different Ages

Objective To find the normal range and features for auditory brainstem responses(ABR)of infants within six months,and the basic parameters for identifying the hearing impaired infants.Methods May 2006 to May 2008,60 normal infants and 20 normal adults received the ABR tests.The infants were divided into three groups:group A(6 weeks),group B(3 months)and group C(6 months).Each group consisted of 20 infants(40 ears)and the ABR data were statistically analyzed.Results At 100 dB nHL,the waveⅠ,Ⅲ,Ⅴ latencies of ABR for 6-week infants respectively were 1.49?0.08 ms,4.42?0.16 ms and 6.61?0.25 ms,for 3-month infants,1.47?0.07 ms,4.35?0.20 ms and 6.50?0.25 ms,for 6-month infants,1.45?0.07 ms,4.17?0.15 ms and 6.32?0.22 ms.At 80 dB nHL,the waveⅠ,Ⅲ,Ⅴ latencies of ABR for 6-week infants were 1.63?0.08 ms,4.52?0.17 ms and 6.74?0.26 ms,respectively,for 3-month infants,1.64?0.11 ms,4.44?0.20 ms and 6.67?0.26 ms,for 6-month infants,1.60?0.11 ms,4.27?0.16 ms and 6.43?0.24 ms.As the intensity of stimulating signals decreased,the peak latency of ABR became more delayed.The peak latencies of Ⅲ,Ⅴ waves and interval peak latencies of Ⅰ-Ⅲ and-Ⅴ waves for the infants were more delayed than those for adults,and became shorter with the increase of the infants' ages,but they failed to reach adults' levels even for 6-month infants.However,the length of wave Ⅰ of ABR for infants of different months was close to that for adults.The thresholds of ABR for the infants of different ages were not significantly different,all being close to the values for adults.Conclusion This study has analyzed the ABR wave features for the infants of three ages and this can help to define the reference ranges for the infants and to provide the parameters for early diagnosis of hearing loss among the infants.

Objective To design and develop a Information System for Medical Center of Clinical Audiology , (MCCAIS301 ) .Methods The system framework was established by developing software ,constructing user platform and creating database .An implication procedure was also established for clinical use for the MCCA IS301 .The MC-CAIS301 was connected with the Hospital Information System (HIS) in order to connect the equipment in the auditory clinical center and other clinical database system .Results The MCCAIS301 was a new database system for hospital in-formation management specifically designed for audiological tests .It provided an extra functions of the existing HIS system .The MCCAIS301 could store the testing results from more than ten different hearing instruments made from five different companies .The data from the MCCAIS301 could be transferred to the HIS system .The results of the MCCAIS301 could be retrieved and analyzed using the HIS system .MCCAIS301 system had nine sets of standardized hearing testing results ,five output formats and three statistical analyzing functions .Conclusion The MCCAIS301 is an effective information management system which has a strong practical use to improve the efficiency of daily audiology data analysis .The MCCAIS301 using digital technology moves the audiology data analysis from a manual low efficient stage to an effective and intelligent level .
3.Effect of Hemoperfusion on Inflammation Factors in Patients with Sepsis

Objective To observe the changes of inflammatory factors and clinical parameters on septic patients with hemoperfusion,and to discuss the application of hemoperfusion on sepsis. Methods 43 patients with sepsis were divided into treatment group and control group randomly. In the treatment group,the patients received conventional treatment and hemoperfusion together,which performed every 24 hours,continuously for 3 times when they arrived in ICU in the first hour. The concentrations of TNF-α,IL-6,IL-10 and PAF were dynamically detected before hemoperfusion,after 24 hours,48 hours and 72 hours in treatment group. The concentrations of TNF-α,IL-6,IL-10 and PAF were compared between the two groups after 72 hours. So did the clinical parameters as WBC count,CRP,PCT and blood lactate acid. Results The concentrations of TNF-α,IL-6, IL-10 and PAF were increased significantly in the early stage of sepsis,and were decreased obviously after hemoperfusion(P < 0.01). After 72 hours treatment,the concentrations of TNF-α,IL-6 and PAF were decreased rapidly,so did the level of CRP,PCT and blood lactate acid. There were significant differences between the two groups(P < 0.05). ConclusionHemoperfusion could remove the inflammatory factor of septic patients and improve the clinical symptoms of them.
4.Efficacy and safety of alizarin combined with standard anti-tuberculosis therapy in treatment of multidrug-resistant pulmonary tuberculosis

Objective To evaluate the clinical efficacy and adverse reactions of alizarin combined with anti-tuberculosis therapy for multidrug resistant pulmonary tuberculosis (MDR-PTB).Methods A total of 200 confirmed MDR-PTB patients admitted in the Affiliated Hospital of Hangzhou Normal University during June 2013 and June 2015 were enrolled in the study.Patients were randomly divided into study group and control group (100 in each).Both groups were given standard anti -tuberculosis treatment for 8 months, and additional alizarin was given to study group .Chi-square test was used to assess the differences in clinical efficacy, sputum negative conversion rate, cavity closure and lesion absorption rate , as well as the incidence of adverse reactions between two groups ( including patients categorized according to TCM syndrome ). Results There were 39 markedly effective cases, 51 improved cases, 10 ineffective cases in study group, and 22 markedly effective cases, 35 improved cases, 43 ineffective cases in the control group.The total effective rate in study group was significantly higher than that in control group (90% vs.57%, χ2 =28.262, P <0.01).For patients with TCM syndrome differentiation as phlegm -heat stagnating lung and those with qi-stagnation induced blood-stasis, alizarin combination therapy had significantly higher total effective rate than standard anti -tuberculosis treatment (78.78% vs.63.33%, χ2 =7.187, P <0.05;95.74% vs.42.31%, χ2 =73.997, P <0.01), but the difference was not observed in patients with TCM syndrome differentiation as deficiency of qi and blood (95.00% vs.88.89%, χ2 =5.025, P >0.05). There was no significant difference in sputum negative conversion rate between two groups (76% vs.55%,χ2 =2.190, P >0.05).The cavity closure and lesion absorption rate in study group ( 91%) was significantly higher than that in the control group (54%,χ2 =38.294, P <0.01).The adverse reaction rate in study group was 27%, which was significantly lower than that in control group (66%, χ2 =30.570, P <0.01).Conclusion Alizarin in combination with standard anti -tuberculosis therapy can improve the clinical efficacy and reduce adverse reactions in treatment of MDR -PTB.
5.Cytotoxic effects of cytokine-induced killer cells transfected with the interleukin-2 gene on malignant melanoma cells

Objective To evaluate cytotoxic effects of cytokine-induced killer cells (CIK cells) transfected with the interleukin-2 (IL-2) gene on malignant melanoma cells.Methods Mouse spleen cells were extracted,lymphocyte cells were separated,and CIK cells were prepared from these lymphocyte cells.PEGF-N1 plasmids containing IL-2 gene (PEGF-NI-IL-2) were transfected into CIK cells.Fluorescence microscopy was used to observe transfection products,and reverse transcriptase-polymerase chain reaction (RT-PCR) was conducted to determine the IL-2 mRNA expression.Then,effector cells such as CIK cells and IL-2-transfected CIK cells were separately co-cultured with target cells (B16 melanoma cells) at effector-target ratios of 10∶ 1,20∶1 and 40∶1,then 4-hour lactate dehydrogenase release assay was performed to evaluate cytotoxic effects of the two kinds of CIK cells on B 16 cells.After effector cells were cocultured with target cells at the effector-target ratio of 40∶1 for 48 hours,enzyme-linked immunosorbent assay (ELISA) was conducted to detect levels of IL-2,interferon-γ (IFN-γ) and tumor necrosis factor-α (TNF-α) in the supernatant of the two kinds of CIK cells.Finally,mouse models of melanoma were established,and a total of 28 melanoma-bearing mice were divided into 4 groups to be peritumorally injected with 0.2 ml sodium chloride physiological solution (control group),100 IU IL-2 solution (IL-2 group),CIK cell suspension at a cell density of 1 × 106 cells per milliliter (CIK group) and IL-2-transfected CIK cell suspension at a cell density of 1 × 106 cells per milliliter (IL-2-transfected CIK group) respectively.Tumor morphology,tumor inhibition rate and cell apoptosis rate were used to evaluate tumor growth in the above groups.If data were normally distributed,t-test was used for comparing means between two groups,and analysis of variance and least significant difference (LSD)-t test were used for comparing means among multiple groups.Results Fluorescence microscopy and RT-PCR both showed that IL-2 was successfully transfected into CIK cells.The cytotoxic effect of IL-2-transfected CIK cells on B16 cells was strongest at the effector-target ratio of 40:1.Levels of IL-2,IFN-γ and TNF-α were also significantly higher in the supernatant of IL-2-transfected CIK cells [(1107.26 ± 6.49) pg/ml,(50.01 ± 3.35) pg/ml,(39.86 ± 3.25) pg/ml] than those in that of CIK cells [(51.09 ± 3.85) pg/ml,(32.71 ± 2.43) pg/ml,(30.11 ± 3.08) pg/ml,t =442.60,14.93,6.89,all P < 0.01].Animal experiments showed that the tumor volume obviously increased in the control group (P < 0.05),but significantly decreased in the IL-2 group,CIK group and IL-2-transfected CIK group (all P < 0.001) after intervention compared with those before intervention.Furthermore,the tumor volume in the IL-2-transfected CIK group was significantly less than that in the other three groups (all P < 0.01),but no significant difference was observed between the IL-2 group and CIK group (P > 0.05).In addition,the apoptosis rate was significantly higher in the IL-2 group,CIK group,and IL-2-transfeeted CIK group than that in the control group (all P < 0.01).The apoptosis rate and tumor inhibition rate were significantly higher in the IL-2-transfected CIK group than those in the IL-2 group and CIK group (all P < 0.01),but insignificantly different between the IL-2 group and CIK group (P > 0.05).Conclusion IL-2-transfected CIK cells had stronger killing effects on malignant melanoma.

7.Analysis on the prevention and treatment of drinking water fluorosis Guide county, in Qinghai province

Objective To observe the state of endemic flurosis, construction and running status of water improvement projects in order to provide a scientific basis for prevention and treatment fluorosis. Methods Water samples of the diseased and nondiseased villeges were collected from east, west, south, north and centre of each villege in 2005, and fluoride concentration was determined for each surveyed village with unimproved-water. At the same time, all the tap water and source water samples were collected to determine fluoride concentration in each water-improved village surveyed. In 2008, all the endemic fluorosis villages in Guied county were divided into slight, medium and heavy types according to the water fluoride content before water improved, and 1,1,3 survey villages were chosen from each type. In all of the village children aged 8 to 12 years were tested for dental fluorosis by Dean method. Six copies of the urinary fluoride were sampled in different age groups. The fluorine content in water and urine was determined by F-ion selective electrode. The situation of clinical skeletal fluorosis of adults over 16 years of age was investigated, 20 adults (evenly divided between men and women) in the villages of medium and heavy types were examined by X-ray for skeletal fluorosis. Results In 3 village fluoride content of drinking water exceeded the national drinking water standards ( <1.0 mg/L) of 85 surveyed villages with improved-water. Among the 16 projects, 8 were intermittently running and 3 were retired, leaving only 31.25% of the projects active. Theprevalence of enamel fluorosis was 41.13%( 116/282), that of skeletal flurosis was 47.95%(969/2021) and that of X-ray checked was 20.73% (17/82). The median of urine fluoride was 1.06 mg/L and the scope was 0.20 - 9.44 mg/L.Conclusions Most of the improved-water projects do not normally supply water in the disease ward of Guide county. Therefore, there is an increasing trend of the disease, so further control measures are needed.

9.Porphyromonas gingivalis induced interleukin-6 expression by Nod/Rip2-mediated signaling pathway.

OBJECTIVETo investigate into the signaling pathway of Porphyromonas gingivalis (P. gingivalis) on cytokine expression in human dental pulp cells (HDPC).
METHODSAnaerobic method was employed to culture P. gingivalis, and then HDPC were intracellularly infected by P. gingivalis. The extraction of total RNA, real-time quantitative polymerase chain reaction (qPCR) and enzyme linked immunosorbent assay (ELISA) was used for mRNA expression of Nods and Rip2, protein secretion of interleukin-6 (IL-6).
RESULTSHDPC expressed Nods, Rip2 mRNA and IL-6. The up-regulation of Nods and Rip2 mRNA started after P. gingivalis infection, reached maximal level at 2 h, and then decreased at 6 h; whereas elevated IL-6 was found when P. gingivalis infected.
CONCLUSIONP. gingivalis activate host innate immune responses in HDPC, and induce IL-6 production through Nod/Rip2-mediated signaling pathway.

10.Screening differentially expressed genes in the process of vascular smooth muscle cell calcification in vitro.

AIMThe process of vascular calcification involves various genetic alterations which may play a very important role in the vascular calcification. Vascular smooth muscle cells undoubtedly composed the main part of vascular cells, and are involved in vascular calcification. So bovine artery smooth muscle cell (BASMC) was used to investigate the gene changes during BASMC's calcification.
METHODSBovine artery smooth muscle cells cultured in vitro was induced calcified by beta-Glycerophosphate (beta-GP). Using DD-PCR technique to screening differentially expressed genes and those differentially expressed bands were reexamined by reverse Northern blot. All the ESTs were sequenced and BLAST with GenBank.
RESULTSTotal 65 cDNAs were isolated as differentially expressed genes and 40 of them were successfully reamplified. Using reverse-Northern blot, seven of these 40 cDNAs were reproducibly expressed differentially between the two cells. Three of them are new bands and have not been reported before.
CONCLUSIONThis is the first time using DD-PCR to screen differentially expressed genes of BASMC calcification. Seven related ESTs were identified relating to BASMC calcification.
