1.Nutlin-3 promotes pyroptosis in SMMC-7721 cells

Objective:To evaluate the effect of Nutlin-3,a MDM2 antagonist,on the pyroptosis in SMMC-7721 cells.Methods: The expression of actived caspase-1(p20) and IL-1β was detected using Western blot analysis.Pyroptosis was investigated by a standard lactate dehydrogenase release assay(LDH).IL-1β content in the cell culture supernatant was quantified by ELISA.Results: Nutlin-3 up-regulated the expression level of actived caspase-1 and IL-1β in SMMC-7721 cells.Meanwhile,Nutlin-3 increased significantly the content of LDH and IL-1β in the cell culture supernatant(P<0.05).Conclusion: Nutlin-3 activated pro-caspase-1,promoted pyroptosis and IL-1β release in SMMC-7721 cells.
2.Determination of amino acids in Galli Gigerii endothelium corneum by HPLC with pre-column derivatization.

Hydrolytic amino acids were extracted by acid hydrolysis method, then derivatized with phenyl isothiocyanate (PITC). And the samples were analysed by HPLC on an Ultimate Prime C18 (4.6 mm x 250 mm, 5 microm) column with gradient elution of 0.1 mol x L(-1) sodium acetate buffer solution (adjusted to pH 6. 5)-acetonitrile (93:7) (A) and acetonitrile-water (8:2) (B) at a flow rate of 1.0 mL x min(-1). Column temperature was 40 degrees C and the detected wavelength was 254 nm. Amino acids derivative solution remained stable in 36 hours. The response was linear for 16 amino acids with a correlation coefficient r > 0.999 5. The average recoveries were 98.01% -101.8%. The method is reliable with good accuracy and repeatability, which is useful for the determination of amino acids in Galli Gigerii Endothelium Corneum.

3.Preparation and evaluation of novel mesoporous molecular sieve of baicalin surface molecularly imprinted polymers.

Taking mesoporous molecular sieve MCM-41 as a substrate, baicalin (BA) as template, acrylamide (AM) as the functional monomer, ethylene glycol dimethacrylate (EGDMA) as a cross-linking agent, ethanol as solvent, under thermal polymerization initiator of azobis isobutyronitrilo (AIBN) , a kind of selective recognition of baicalin surface molecularly imprinted polymer was synthesized. The surface morphologies and characteristics of the MIPs were characterized by infrared spectroscopy (IR) and transmission electron microscope (TEM). The adsorption properties of polymer microsphere for the template were tested by the dynamic adsorption equilibrium experiments and static adsorption equilibrium experiments. The experiment showed that the imprinting process was successfully and the well-ordered one-dimensional pore structure of MCM-41 was still preserved. Furthermore, molecularly imprinted polymers had higher selective ability for BA, then provided a new method for the efficient separation and enrichment of baicalin active ingredients from medicinal plants Scutellaria baicalensis.

4.Clinical analysis of renal artery branch lesions caused by fibromuscular dysplasia in children

Objective To explore the lesion types of renal branches and evaluate the clinical results of endovascular treatment for renal artery branch lesions associated with fibromuscular dysplasia (FMD).Methods Eight cases with renal artery branch lesions due to FMD from June 2014 to December 2015 in Department of Vascular and Endovascnlar Surgery,Henan Provincial People's Hospital were retrospectively reviewed.The computed tomography angiography was routinely performed to ascertain the property of renal artery lesion,and digital-subtraction angiography was further performed to confirm FMD diagnosis and analyze lesion characteristics.The renal artery branch lesion types,blood pressure (BP),renin activity,glomerular filtration rate (GFR) and restenosis after operation were observed,and the outcomes were recorded.Results In 8 patients,6 patients were found with the second grade renal branch lesions and 2 patients were found with tertiary branch lesions,involving 2 and more branch arteries for 4 cases and 1 branch for 4 cases.Five cases with branch-type cystic dilatation or aneurysms with coexisting stenosis including 1 case with dissecting aneurysm and renal local infarction,and 1 case with aneurysm located in the bifurcations of branches,and 3 cases with only branch stenosis were found.The angiography showed the small branch lesions of renal artery indicating FMD-mediated focal (5 cases) and multiple stenosis (3 cases).The branch stenosis was not operated in 1 case with dissecting aneurysm and renal local infarction,as BP could be controlled by antihypertensive treatment.Other 7 cases underwent endovascular angioplasties and the technical successful rate was 100% without complications.After 1 week of operation,BP was improved,renal function remained normal,plasma renin activity was reduced and GFR was recovered for all patients.During the follow-up period (3 to 12 months),BP improvement was observed in 3 cases,and cure were found in 4 cases,and patient's GFR and plasma renin activity were improved in all patients.There were 2 cases with re-intervention during the follow-up period due to restenosis,and the involved branches were unobstructed without obvious stenosis after 1-year follow-up.Conclusions FMD can affect the renal branch artery with different lesions types and endovascular treatment is the first choice for the renal branch lesion of FMD.

In order to establish a method by which the recombinant suicide plasmids integrated on the chromosome could be recircled, A simple method of transconjugative cloning was established with the helper plasmids pMT999 or pRK2013 and fusion strains of Shigella flexneri which were obtained by screening with in vivo expression technology. And the cloning efficiency with this method is very high.
6.Study on Water-Soluble Constituents from Tibetan Medicine Terminalia billirica (Gaert.) Roxb

This paper aimed at investigating the water-soluble constituents of Terminalia billirica (Gaert.) Roxb.Compounds were isolated and purified by Diaion HP-20, Sephadex LH-20, Toyopearl HW-40, MCI gel CHP 20 P and ODS column chromatographies. The chemical structures were elucidated according to spectral data and physicochemical properties. The results showed that ten compounds were isolated and identified as corilagin, salicoside, methyl-α-Darabinofuranoside, 1, 4-dimethyl-β-d-fructopyranose, β-fructopyranose, β-fructofuranose gallic acid, ellagic acid, 1, 2, 3, 4, 6-penta-O-galloyl-β-D-glucopyranonside, Chebulagic acid. Except compound 7 and 8, others compounds are separated from Terminalia billirica (Gaert.) Roxb for the first time, and except corilgin, the other compounds are separated from Terminalia for the first time.
7.Clinical Observation of Moxibustion on Acupoint Feishu for Treatment of Acute Asthma in Children (Cold-Asthma)

OBJECTIVE To observe the clinical effect of moxibustion on acupoint Feishu (BL13) in the treatment of acute asthma in children (cold-asthma),and to provide clinical evidence for treatment of asthma with acupuncture and moxibustion.METHODS 87 children with asthma (cold-asthma) who meet the inclusion criteria were selected and randomly divided into 2 groups.The control group was treated with conventional western medicine,and the observation group was treated with moxibustion on bilateral Feihu Points(BL13) on the basis of routine western medicine.CRP,IgE,IL1,IL6,EOS,pulmonary function and symptom score were selected as observation indexes for the clinical effects comparison of patients in two groups.RESULTS Indexes levels such as CRP,IgE,IL6 and EOS in the observation group were significantly lower than that in the control group after treatment (P<0.05~0.01).FEV1 and PEF levels of patients in the observation group were significantly better than that in the control group after treatment (P<0.05).Symptom score in the observation group was significantly lower than that in the control group after treatment (P<0.05).Apparent rate (73.81%) of patients in the observation group was significantly higher than that in the control group (53.33%) (P<0.05).CONCLUSION Moxibustion on acupoint Feishu in the treatment of acute asthma in children (cold-asthma) can markedly decrease the levels of CRP,IgE,IL6 and EOS in patients,improve pulmonary function,and can achieve remarkable clinical curative effect.
8.A prospective randomized controlled trial of Sapylin combined with chemotherapy for advanced colon cancer following radical operation.

OBJECTIVETo evaluate the effect of Sapylin combined with intraperitoneal and systemic chemotherapy for advanced colon cancer following radical operation on local recurrence, hepatic metastasis, and overall survival rate.
METHODSFrom Jan. 2004 to Dec. 2005,132 patients with stage II or III colon carcinoma after radical operation were randomly divided into two groups: Sapylin combined with chemotherapy(Sapylin) group and the control group. Toxic reaction, local recurrence, hepatic metastasis, and overall survival rate between two groups were compared.
RESULTSBoth groups successfully completed the trial. There was no significant difference in toxic reaction between two groups, the recurrence and hepatic metastasis rate in Sapylin group were lower than those in the control group(9/60, 15.0% vs. 22/72, 30.6%; 11/60,18.3% vs. 22/72, 34.7%, respectively), which were statistically significant (P<0.05 respectively). The 3- year survival rate in Sapylin group was higher than that in control group(73.3% vs. 54.2%), which was statistically significant (P<0.05).
CONCLUSIONSapylin combined with intraperitoneal and systemic chemotherapy can effectively decrease local recurrence and hepatic metastasis,and improve the survival in patients with advanced colon cancer following radical operation.

9.Effect of adenovirus-mediated TK/GCV gene therapy in combination with TNF-alpha against murine bladder cancer cells in vitro.

OBJECTIVETo investigate the cell-killing effect of adenovirus-mediated TK-ganciclovir (GCV) gene therapy in combination with tumor necrosis factor-alpha (TNF-alpha) against murine bladder carcinoma cells in vitro.
METHODSMurine bladder carcinoma MB49 cells were transfected with the adenoviral vector containing TK gene and green fluorescent protein (GFP) gene. The transfection efficiency was observed and the TK gene expression in the transfected cells was detected by RT-PCR. The survival rate of MB49 cells in response to TNF-alpha treatment and that of the TK gene-transfected cells after treatment with GCV and GCV+TNF-alpha were determined by MTT assay. The apoptosis of the cells after the treatments was analyzed by flow cytometry.
RESULTSIn cells transfected with TK gene, the cell inhibition rate increased gradually with the increment of GCV and TNF-alpha concentration. GCV in combination with TNF-alpha resulted in significantly increased killing efficiency of the cells as compared with GCV or TNF-alpha treatment alone, and the effect of the combined treatment was enhanced as the TNF-alpha concentration increased. GCV treatment (50 microg/ml) alone produced a cell killing rate of (24.39-/+1.10)%, and when combined with 5 microg/ml TNF-alpha, the rate was increased to (40.05-/+0.97) %, and further to (65.47-/+0.67) % when TNF-alpha concentration increased to 20 microg/ml. Flow cytometry revealed obvious apoptosis of the cells 8 h after treatments with TK/GCV, TNF-alpha, or TK/GCV+TNF-alpha, and the combined treatment resulted in the highest cell apoptotic rate.
CONCLUSIONTK/GCV in combination with TNF-alpha can enhance the effect of suicide gene therapy against murine bladder carcinoma cells and effectively induce apoptosis of the cells.

10.The fabrication and clinical application of semi-fixed mandibular lingual arch expansion appliance.

Semi-fixed mandibular lingual arch expansion appliance is composed of a mandibular molar band, a keyway and arch expansion spring. The arch expansion spring are used to expand maxillary arch symmetrically or asymmetrically when bolts of the two ends are inset into keyways. Dental arch expansion appliance for 25 patients with mandibular arch stenosis showed that semi-fixed mandibular lingual arch expansion had good effect and could be used to expand mandibular arch.
