The paper reports the systematic study on felodipine and its impurities in tablets, to improve its quality standards for the control of the related substances. HPLC-DAD, UPLC-MS, IR and NMR methods were used for the isolation of felodipine and its impurities in tablets, their identification and the zebrafish animal model was used for the analysis of the toxic impurities. In felodipine material and its tablets, three impurities are isolated and identified. They are impurity 1 [dimethyl 4-(2, 3-dichlorophenyl)-2, 6-dimethyl-1, 4-dihydropyridine-3, 5-dicarboxylate], impurity 2 [ethyl methyl 4-(2, 3-dichlorophenyl)-2, 6-dimethylpyridine-3, 5-dicarboxylate] and impurity 3 [diethyl 4-(2, 3-dichlorophenyl)-2, 6-dimethyl-1, 4-dihydropyridine-3, 5-dicarboxylate], separately. The result of zebrafish animal model analysis showed that the teratogenic effects of four compounds were: impurity 3 > or = felodipine > impurity 1 > impurity 2, lethal effects were as follows: impurity 2 = impurity 3 > felodipine > or = impurity 1. This study confirmed the toxicity of three impurities in felodipine. According to the results, the paper suggested the amendments to the standard of the medicine and provided the support to the control of impurities in the manufacturing process.

5 fold) were noted.Six significant differential proteins in gel were identified by LTQ-ESI,e.g.endoplasmin precursor, acidic leucin-rich nuclear phosphoprotein 32 family member A,serotransferrin precursor,stress-70 protein precursor, fibronectin precursor, complement C3 precursor, fibrinogen gamma polypeptide.CONCLUSION: The protein profile of apoE-/-/LDLR-/-mouse liver exhibits significant difference compared to that of WT mice. The results imply that lipid metabolism relative polygenetic mutation contributes to the alteration of mouse liver protein expression profile, especially that lipid metabolism related perhaps participates in dysfunction in lipid metabolism during atherogenesis.

Objective To observe the protein degredation of empty puparium cases ofC. megacephala after weathering and to explore its practical significance on postmortem interval estimation in forensic science. MethodsThe standardized feeding was used inC. megacephala, and the empty puparium cases were collected and put into the forest. They were taken back 5 days, and 10 days later, respectively. The spectra were collected and preprocessed, and then the absorption peaks of amides were read, and curve fitting was performed in the average spectra of each group. The statistical analysis was performed by SPSS 19.0.Results Compared to control group, ifve days post weathering, the position of amideⅠabsorption peak showed blue shift, but the amideⅡabsorption peak showed no shift, moreover, none of the amide absorption peaks showed changes inpeak intensity. For the secondary structures, α-helix decreased and β-sheet increased slightly, however, β-turn did not change; Ten days post weathering, the positions of both amideⅠandⅡabsorption peaks showed blue shift, and an obviousshoulder peak appeared in amide I absorption peak. Meanwhile, both of amideⅠandⅡabsorption peaks showed decreased in peak intensity. Moreover, for the secondary structures, α-helix and β-sheet showed the same tendency as in the 5th day group, except for the β-turn increased dramaticlly. Conclusion The spectra of the empty puparium cases of C. megacephala showed that the absorption peaks of amideⅠandⅡpresented certain characteristic features within ten days post weathering, and estimate relative long-time postmortem interval.

Objective To detect the molecular characteristics of the serum samples of rats suffered from sudden unexpected death of acute myocardial ischemia (experimental group) and death caused by air embolism (control group) applied by ATR-FTIR, and to discuss the forensic signiifcance of identiifcation of causes of death by sudden unexpected death of acute myocardial ischemia by two-dimension infrared spectroscopy. Methods Adult male SD rats were randomly divided into SCD group and DCAE group. The serum samples were analyzed by ATR-FTIR. The similarity coefifcient of spectra between two groups was analyzed by OMNIC software. The spectra with higher signal-noise ratio were obtained and used to build two-dimension spectra model by Matlab 2010a software. Results The peak shapes, positions and intensities of the spectra were extremely similar between two groups, and the similarity coefifcient reached 99.78%. Meanwhile, the similarity coefifcient of second derivative spectra decreased to 98.62% between two groups. Moreover, on the two-dimension spectra model, the component differences were present at about 1 625cm-1, 1 550cm-1, 1 080cm-1 and 860cm-1, and the number of auto-peaks was different, which suggested that the evident differences were related to the molecular contents of protein, glycogen, nuclear acid, and so on. Conclusion It is hard to differentiate the molecular differences of serum samples of rats between SCD and DACE based on one-dimension spectra. However, two-dimension infrared spectra could intuitively and effectively relfect the molecular differences, and it would apply to determining the cause of death in forensic science.

OBJECTIVETo explore the effect of arteriosclerosis obliterans (ASO) blood stasis syndrome (BSS) serum on vascular endothelial cell injury and to study the regulation of Taohong Siwu Decoction (TSD) on it.
METHODSUmbilical vein endothelial cell culture system was established. The serum endothelial cell injury model with ASO BSS was prepared. Low, medium, and high concentrations TSD containing serums were respectively added. The endothelial cell proliferation activity was observed by MTT method. Ultrastructures of endothelial cells were observed under transmission electron microscope. Changes of intracellular calcium ion concentration and the cytoskeleton were observed under laser confocal microscope. Contents of ET, NO, and transforming growth factor beta1 (TGF-beta1) in endothelial cell culture supernatant were detected by ELISA.
RESULTSIn ASO BSS serum group endothelial cell proliferation activities decreased, the cell structure was obviously destroyed, calcium ion concentration increased, contents of ET, NO and TGF-beta1 increased significantly (P < 0.01), and ET/NO ratio was imbalanced. After incubating with TSD drug containing serum, endothelial cell proliferation activities and injured cell structures were obviously improved; ET, NO and TGF-beta1 levels decreased (P < 0.05, P < 0.01), ET/NO ratios approximated to the normal level.
CONCLUSIONThe main mechanism of TSD for treating ASO ASS lied in improving injured vascular endothelial cells and endocrine disorder.

China Occupational Medicine 2021;48(03):340-346
Silica dust is one of the main occupational hazards in various industries. In addition to causing occupational silicosis, silica dust can also stimulate innate and acquired immunity and induce autoimmune diseases(AID) such as systemic lupus erythematosus, rheumatoid arthritis, systemic sclerosis, and antineutrophil cytoplasmic antibody(ANCA)-associated vasculitis. Silica dust exposure can induce changes in the levels of autoantibodies in vivo, such as rheumatoid factor, antinuclear antibody, ANCA, anti-centromere antibody, anti-topoisomerase Ⅰ antibody, anti-desmoglein antibody, anti-centromere antigens-B antibody, anti-carbamylated protein antibody, anti-smooth muscle antibody and anti-glomerular basement membrane antibody. At present, the mechanism of autoimmune disorders induced by silica dust has not yet been fully elucidated. The current research suggests that it is related to cell apoptosis of alveolar macrophages, the disorder of the Fas/Fas ligand system, the imbalance of T cell proportion, and the dysregulation of T helper cell(Th) 1 and Th2 type cytokines homeostasis. Understanding the pathogenesis of autoimmune disorders induced by silica dust and exploring possible therapeutic targets will provide new ideas for the treatment of silicosis.
