2.Effect of Telmisartan on Renal Expression of Adiponectin in Insulin-resistant Rats

Objective To investigate the influences of telmiaartan on renal expression of adiponectin (APN) in insulin resistance rats. Methods Insulin resistance rat model was induced by long-term high-fat feeding and was assessed by euglycemic-hyperinsulinemia clamp technique. The morphological changes of the kidney lissue were observed by the light and electronic microscopy.Glomerular APN mRNA ex-pression and protein synthesis were evaluated by the reveree transcription polymerase chain reaction and immunohistochemical staining re-spectively. Results Both APN mRNA and APN protein were lower in insulin resistance rats than those in control rats (P < 0.05). More-over, the main features of the renal parenchyma were observed in insulin resistance rats. The expression levels of APN mRNA and APN pro-tein were increased after the treatment with lelmisartan (P <0.05). The renal damage was significantly reduced as well. Conclusion The up-regulation of APN expression induced by telmisartan might prevent the renal damage of insulin resistance.
4.Effect of telmisartan on levels of serum adiponectin and expression of cardiac adiponectin receptor 1 in spontaneously hypertensive rats

ObjectiveTo observe the effect of telmisartan on the level of serum adiponectin and the expression of cardiac adiponectin (APN) receptor 1 in spontaneously hypertensive rats (SHR) in order to explore the role of APN in ventricular remodeling caused by hypertension and the new possible mechanism that telmisartan protects the heart against ventricular remodeling.Methods The sixteen 12-week-old SHR were randomly divided into SHR control group (SHR group, n=8) and SHR+telmisartan group (TEL group, n=8), and eight age-matched male Wistar Kyoto rats were regarded as control group (WKY group). The TEL group was treated with telmisartan 5 mg-1 · kg-1 ·d-1 in a small amount of distilled water and the other two groups were given equal amount of distilled water by gavage.After 8 weeks, the left ventricular end diastolic diameters (LVEDD),thickness of interventricular septal thickness (IVST), left ventricular post wall thickness (LVPWT) and mitral valve blood flow spectrums were recorded by echoeardiograph.The E/A ratio was calculated by velocity of E peak divided by A peak. The left ventricular weight index (LVWI) was calculated. HE staining and Masson staining were used to detect the pathology changes of cardiac tissue and collagen volume fraction (CVF).Serum APN concentration of SHR was detected by enzyme-linked immunosorbent assay (ELISA). The myocardial protein expression of AdipoR1 was detected by in munohistochemical staining.Reverse transcription polymerase chain reactions (RTPCR) method was used to detect the mRNA expression of AdipoRl in myocardium.ResultsThe values of IVST, LVPWT and LVWI were higher, LVEDD and E/A ratio were lower in SHR group than in WKY group.The disordered arrangement of myocardialcells,interstitialfibroblast hypertrophy and hyperplasia were observed in SHR group. The CVF increased in SHR group than in WKY group [(6.22±0.16)% vs. (3. 18±0. 17)%, P<0.05]. The concentration of serum APN was significantly lower in SHR group [( 10.96±2. 15)μg/ml vs. (24.32±2. 20)μg/ml, P<0. 01].The expressions of myocardial AdipoR1 mRNA and protein were significantly lower in SHR group.The values of IVST, LVPWT and LVWI were lower, LVEDD and E/A ratio were higher in TEL group than in SHR group. The arrangement of myocardial cells were relatively ordered and interstitial fibroblasts hypertrophy and hyperplasia were not significant, CVF was decreased in TEL group versus SHR group. The concentration of serum APN was significantly higher in TEL group [(17.71 ± 5.82)μg/ml, P<0. 01]. The expressions of myocardial AdipoR1 mRNA and protein were significantly higher in TEL group than in SHR group. Conclusions When SHR undergoes ventricular remodeling, the level of serum APN is lower, the mRNA and protein expressions of myocardial AdipoRl are significantly reduced. Telmisartan can reverse the ventricular remodeling and play the role in protecting heart in SHR, which may be due to the serum APN levels increasing, cardiac APN receptor 1 and its mRNA up-regulation.
6.Effects of gauzes with composite lysozyme on chemotherapeutic phlebitis

Objective To study the effect of gauzes with composite lysozyme on chemotherapeutic phlebitis. Methods One hundred and twenty patients with chemotherapeutic phlebitis were equally randomized into the experiment group and control group with radom digit tade. The experiment group was treated by hydropathic compress with gauzes with composite lysozyme on the affected parts, while the control group was treated by hydropathci compress with 50%magnesium sulfate solution. The therapeutic effects after 1 week were compared between the two groups . Results The effective rate of the experiment group was significantly higher than that of the control group (P < 0.05). The average time for the treatment was significantly shorter than the control group (P < 0.05). Conclusion The effect of composite lysozyme for hydropathic compress in the treatment of chemotherapeutic phlebitis is better than that of 50%magnesium sulfate. It is worthy of clinical popularization and application.
8.Progresses of interventional treatment in biliary stenosis

Interventional therapy is an important treatment for biliary stenosis.The treatment methods are different ac cording to the different causes.Conventional interventional therapy include biliary drainage,balloon dilatation and stent implantation.There are some new treatment methods such as radiofrequency catheter ablation and biliary stent loaded with 125I seeds.The applications of interventional therapy in biliary stenosis were reviewed in this article.

10.Effects of high fat/high cholesterol diet on differentially expressed gene of the liver in treble genes mutant mice

AIM: To study the effects of dietary factor on liver gene expression profile in apoE~(-/-)/LDLR~(-/-)/Lepr~(db/db) mice and atherogenesis. METHODS: The gene expression profile was determined by using cDNA microarray. Total plasma TC and TG level were measured with COD-PAP and GPO-PAP methods respectively. The morphological characters of the aorta were also observed. RESULTS: Among the 4000 target genes, 78 and 114 genes were up-and down-regulated, respectively, in treble genes mutants fed with high fat high cholesterol diet compared with that of regular chow littermates, including lipid metabolism, carbohydrate, cell skeleton and immune related genes. Atherogenic diet induced severe plasma hyperlipidemia even in the early stage and also accelerated atherogenesis in the treble genes mutant mice. CONCLUSION: The data suggest that atherogenic diet aggravates abnormal plasma lipid metabolism and induces severe hyperlipidemia, and atherogenesis in the treble genes mutant mice as well.
