1.Establishment of a radioresistant human lung cancer cell subline and its mechanism of radioresistance

Objective To establish a radioresistant cell subline from a human A549 lung cancer cell line and investigate the mechanism of radioresistance. Methods Two proposals were applied for the non-small cell lung cancer A549 cells irradiated with X-rays:A group of A549 cell line was irradiated five times, the fractionated dose was 600 cGy, and the other group was exposed 15 times, the fractionated dose was 200 cGy. After the completion of irradiation, two monoclones were obtained from the survival of cells and named the subline A549-S1 and A549-S2. The radiosensitivity and cell cycle distribution of these two clones,together with its parental A549 cells were measured by clone formation assay and flow cytometry.The mRNA and protein levels of Notch1 in A549 cell line and the sublines were determined by RT-PCR and Western-blots. Results Compared with the parental A549 cells, A549-S1 cells showed significant resistance to radiation with D0, Dq and N values increased, and a broader initial shoulder as well as 1.38-fold increased value of SF2. The A549-S1 subline also showed higher percentage of cells in S phase and G2/M phase, but lower percentages in G0/G1 phase (P<0.05). The expression of Notch1 in A549-S1 was enhanced obviously than in A549 cells. But for A549-S2 the radioseasitivity was slightly increased compared with the parental cells with D0, Dq and N values decreased and a curve initial shoulder. The ratio of cells in S and G0/G1 phase ratio was lower than that in parental A549 cells, but that in G2/M phase ratio was higher significantly (P<0.05) .The expression of Notch1 had no marked change compared to A549 cell. Conclusions The radioresistance of the A549 cell subline is correlated with the irradiation program. The cell subline shows a different cell cycle distribution from their parental line. The cell cycle distribution has a close correlation with the expression of Notch1.
2.Optimization of Extraction Technology of Paeonol from Paeonia Suffruticosa

This article was aimed to study the optimum extraction process of paeonol. The extraction yield of paeonol was taken as investigation index. And the best extraction process was screened by orthogonal experimental design. The results showed that the optimum condition of extraction process was to soak coarse powder of Paeonia suffruti-cosa into 15-fold water for 0.5 h, and then the distillation lasted for 2.5 h. The distillate was collected and cooled to room temperature. The crystallization lasted for 24 h at 4℃, and then filtered and dried for 48 h at room tempera-ture. It was concluded that the selected technology was stable, reasonable and feasible. The extraction yield of paeonol is over 80%.

The recommended design utilizes the virtual instruments concept and high-speed open bus standard. The data processing methods involve qualitative and quantitative methods. Dynamic and static mapping techniques are introduced here as the qualitative methods while statistical methods, inverse calculation and principle component analysis are introduced as the quantitative methods.

4.Clinical research about level Ⅵ-1 lymph nodes and the lymph nodes posterior to the right recurrent laryngeal nerve of the papillary thyroid carcinoma

OBJECTIVE The objective of this study is to discuss when to dissect the lymph nodes behind the right recurrent laryngeal nerve (LN-prRLN) from the standpoint of the right cervical level Ⅵ-1 (superficial layer to the recurrent laryngeal nerve) lymph nodes in papillary thyroid carcinoma (PTC) patients.METHODS The clinical data of 306 bilateral or right PTC patients from the Hangzhou First People's Hospital who underwent dissection of level Ⅵ-1 lymph nodes and LN-prRLN between March 2014 and September 2015 were analyzed. We measured the number of level Ⅵ-1 metastatic lymph nodes and size of level Ⅵ-1 lymph nodes metastasis loci to predict the metastasis of LN-prRLN.RESULTS The number of level Ⅵ-1 metastatic lymph nodes and size of level Ⅵ-1 lymph nodes metastasis loci were risk factors of LN-prRLN metastasis(P<0.05). When the number of the level Ⅵ-1 metastatic lymph nodes was greater than 1.5, the AUC was 0.813 (the sensitivity was 78.43%, the specificity was 76.65%). The ROC showed that when the size of level Ⅵ-1 lymph nodes metastasis loci were more than 0.45 cm, the AUC was 0.726 (sensitivity was 90.20%, specificity was 48.90%).CONCLUSION In bilateral or right PTC patients with metastasis of level Ⅵ-1 lymph nodes, especially when the number of level Ⅵ-1 metastatic lymph nodes was greater than 2cm and the metastasis loci were more than 0.45 cm, we should dissect the LN-prRLN.
5.Morphological observation of Tritrichomonas muris isolated from Xinjiang Cricetulus migratorius and analysis of its 16 S rRNA gene

Objective To conduct morphological observation and gene identification of the strain of flagellate iso -lated from Cricetulus migratorius in the Xinjiang Research Center for Experimental Animals .Methods The ileocecal con-tents of C.migratorius were microscopically examined on direct smear with Wright-Giemsa staining , and the total RNA iso-lated from Xinjiang C.migratorius was extracted and 16S rRNA was amplified by PCR , and then sequenced .Furthermore the homology was compared and the phylogenetic tree was developed using MEGA 5.22 software.Results Morphological observation indicated that the isolated flagellate was Tritrichomonas muris.The 16S rRNA gene sequence of the Xinjiang C. migratorius isolate shared highly homology with that of other Tritrichomonas.Phylogenetic tree analysis indicated that the 16S rRNA gene of Xinjiang C.migratorius isolate was classified into a subgroup with T.muris 16S rRNA (U85966.1), but was relatively distant relative from other related tritrichomonas.Conclusions The flagellate isolated from Xinjiang C. migratorius is identified to be T.muris by both morphological observation and 16S rRNA gene analysis.
6.8.5/11.5F transurethral seminal vesiculoscopy in the diagnosis and treatment of refractory hematospermia.

OBJECTIVETo investigate the clinical application value of 8.5/11.5 F transurethral seminal vesiculoscopy in the diagnosis and treatment of refractory hematospermia.
METHODSWe retrospectively analyzed 78 cases of refractory hematospermia diagnosed and treated by 8.5/11.5 F transurethral seminal vesiculoscopy from June 2012 to June 2014. The patients underwent serum PSA examination, transrectal ultrasonography, seminal vesicle ultrasonography, and pelvis CT or MRI before surgery, and all received transurethral seminal vesiculoscopy under the 8.5/11.5 F rigid ureteroscope.
RESULTSOperations were all successfully accomplished, which revealed abnormal opening of the ejaculatory duct in 5 cases, mucosal inflammatory hyperemia in the prostatic utricle and seminal vesicle in 78, dark red mucilage substance in the seminal vesicle in 34, seminal vesicle stones in 19, small polyp in the seminal vesicle in 2, and ejaculatory duct or seminal vesicle cyst in 4. All the patients received symptomatic treatment during the surgery. After surgery, hematouria was found in 13 cases, which disappeared within 2 weeks, pelvic hematoma in 1 case, which was cured by conservative treatment within 3 months, and epididymitis in 2 cases, which was controlled by anti-infection treatment. Hematospermia recurred in 3 cases during the 1-year postoperative follow-up.
CONCLUSION8.5/11.5 F transurethral seminal vesiculoscopy, with its advantages of easy operation, wide field of vision, large channel for operation, and few complications, deserves general clinical application in the diagnosis and treatment of refractory hematospermia.

7.IFN-gamma and TGF-beta1, levels in the expressed prostatic secretions of patients with chronic abacterial prostatitis.

OBJECTIVETo investigate the levels of pro-inflammatory cytokine IFN-gamma and anti-inflammatory cytokine TGF-beta1, in the expressed prostatic secretion (EPS) of men with chronic abacterial prostatitis and their clinical significance.
METHODSThe levels of IFN-gamma and TGF-beta1, in the EPS of 20 patients with inflammatory chronic pelvic pain syndrome (type III A), 20 patients with non-inflammatory chronic pelvic pain syndrome (type Ill B) and 10 healthy men were measured using enzyme-linked immunosorbent assay (ELISA). The results were analysed comparatively with NIH-chronic prostatitis symptom index (NIH-CPSI).
RESULTSIFN--gamma and TGF-beta1 levels were higher in III ([14.92 +/- 7. 85)], [8477.50 +/- 4612.45] ng/L) and III B ([13.74 +/- 5.96], [7946.50 +/- 5044.06] ng/L) prostatitis patients than those in the controls ([7.47 +/- 1.49], [2462.50 +/- 985.31] ng/L), P < 0.05 and P < 0.001 respectively. There was no statistically significant difference in cytokine levels between III A and Il B prostatitis patients. No correlation was found between NIH-CPSI and cytokine levels, r = 0.02, P = 0.86, r = 0.31, P = 0.76.
CONCLUSIONIFN-gamma and TGF-beta1, play a very important role in the etiology of chronic abacterial prostatitis and can be the objective parameters in the diagnosis of chronic abacterial prostatitis.

8.Changes of the mitochondrial DNA copy number and the antioxidant system in the PBMC of hepatocellular carcinoma.

OBJECTIVETo investigate the relationship between the changes of the copy numbers of mtDNA in peripheral blood mono-nucle- ar cell(PBMC) and the disordered of antioxidant capacity of hepatocellular carcinoma (HCC) patients.
METHODSThe Ficoll Hypaque method was used to isolate the PBMC from blood specimens. The ND1 gene of the mitochondrial was amplified by real-time PCR; meantime β-actin was served as a quantitative standard marker; the difference of mtDNA copy number in PBMC was compared between HCC and healthy control group. The level of reactive oxygen species (ROS) in PBMC was determined by flow cytometry. The change of total antioxidant capacity (T- AOC) of plasma was detected by the biochemistry examination.
RESULTSThe copy numbers of ND1 gene in PBMC of HCC was 73% that of the healthy control group,which suggested a decrease of the copy numbers of mtDNA in HCC. The levels of ROS of PBMC in HCC was (417. 82 ± 110.62) and (301.82 ± 75.54) in control group, which showed that the levels of ROS of PBMC in HCC were significant higher than that in control group (P < 0.01).Plasma T-AOC in HCC was (1.30 ± 0.85), and (3.20 ± 1.62) in control. The T-AOC of plasma of HCC was significantly lower than in control group (P < 0.01).
CONCLUSIONThere was a certain relationship between the decrease of the copy numbers of mtDNA and the disordered antioxidant capacity in hepatocellular carcinoma, which may be associated with the development of hepatocellular carcinoma.

9.Emergency operation for the treatment of intertrochanteric fractures in elderly patients.

OBJECTIVETo evaluate the emergency therapeutic effects of intertrochanteric fractures in elderly patients.
METHODSA retrospective study was designed. From March 2006 to March 2012, 254 patients with intertrochanteric fractures treated in Mianyang Central Hospital were reviewed. The patients with old fractures, pathological fractures, multiple trauma and severe medical comorbidities of admission were excluded. So 127 patients with intertrochanteric fractures over 70 years old were included in the study, including 38 males and 89 females, and the patients were divided into 2 groups: 56 patients underwent emergency operation within 24 h after injury (emergency operation group), 71 patients underwent an operation more than 24 h after injury (not emergency operation group). The following data were collected retrospectively for all patients: (1) in-hospitalcomplications, including electrolyte disturbance, hidden blood loss, delirium, deep vein thrombosis, upper gastrointestinal hemorrhage, pulmonary infection, urinary tract infection, cardiovascular and cerebrovascular disease, bedsore. (2) prognosis, ineluding in-hospital mortality, mortality in the 1st year, length of hospital stay, fracture healing times, hip score. Comparisons between the two groups were made using SPSS13.0. The quantitative data and categorical data were analyzed by t test and chi2 test, respectively.
RESULTSThe main medical complications in emergency operation group were hidden blood loss, delirium, pulmonary infection, electrolyte disturbances, urinary tract infections, and in not emergency operation group followed respectively by hidden blood loss, lung infection, electrolyte disturbances, delirium, and pulmonary infection. In-hospital mortality was zero, and 1 in the 1st year in emergency operation group; while in not emergency operation group were 4 and 7 respectively. An average length of hospital stay with (9 +/- 3) d in emergency operation group was lower than (17 +/- 8) d in not emergency operation group. Fracture healing time was (12 +/- 2) weeks in both groups. The significant variables with P < 0.05 between the two groups were electrolyte disorders, delirium, deep venous thrombosis, pulmonary infection, urinary tract infection, bedsore, in-hospital mortality, mortality in the 1st year, and length of hospital stay. The differences of hidden blood loss, upper gastrointestinal hemorrhage, cardiovascular and cerebrovascular disease, fracture healing times, hip score between the two groups had no significant differences (P > 0.05).
CONCLUSIONThe emergency surgery (within 24 h after injury) reduces length of hospital stay, and may also reduce complications and mortality.

10.The clinical value of the ultrafine needle CO_2 splenoportography

Objective To evaluate the clinical efficacy of percutaneous ultra-fine needle CO_2 splenoportography (CO_2-SP).Methods CO_2-SP and 3D-CE-MRA were performed in 36 patients.The imaging quality of the methods was compared by a scoring criterion setup based on the visualization of the trunk,intrahepatic branches of the portal vein and collateral vessels.Results Transient mild abdominal discomfort was presented in 19 patients(52.8% )receiving CO_2-SP.One patient developed snbcapsular splenic hematoma and was discharged with clinical stability several days later after conservative treatment. The imaging quality of the intrahepatic branches of the portal vein with CO_2-SP was much more superior to 3D-CE-MRA (the score was 232 and 198 respectively,t=4.52,P0.05 ).Conclusion Ultrafine needle CO_2-SP is a minimally invasive and safe procedure,able to provide dynamic and clearer imaging of the intrahepatic branches of the portal vein.
