1. Effect of TY501 on carbon tetrachloride-induced liver fibrosis in mice

Objective To explore the protective effects of glycyrrhetic acid derivatives (TY501) on carbon tetrachloride-induced liver fibrosis in mice. Methods The mice liver fibrosis was established by classical CCl4, and TY501 was given in three kinds of dosage (45, 15, and 5 mg/kg), with colchicine serving as control. After 30 d treatment, the liver function, fibrosis and the levels of Hyp and TGF-β1 were detected. Moreover, the histopathological changes were detected by HE and Masson staining. Results Compared with model group, the levels of ALT, AST, ALP, ALB, HA, LN, PCIII, CIV, Hyp, and TGF-β1 were all decreased, with high-dose group being the most significant (P < 0.05, 0.01). Histopathological examination showed that liver fibrosis of those mice treated with TY501 was improved to some extent. Conclusion TY501 can significantly protect carbon tetrachloride-induced hepatic fibrosis.

Objective To determine the contents of four fiavonoids(trifolirhizin,nor-kurarinone,xanthonumal and kuraridin)in 25 samples of Sophora flavescens Air.in the market.Methods The determination was carried out on Phe- nomenex Luna C_(18)column(250 mm?4.6 mm,5?m),using Acetonitrile-H_2O as mobile phase,at a flow rate of 1.0 mL/min and detected at the wavelength of 310 nm for trifolirhizin,294 nm for nor-kurarinone and,365 nm for xan- thonumal and kuraridin,and column temperature at 35℃.Results The contents of 4 flavonoids in 25 samples of Sophora flavescens Air.in market were in the ranges of 0.05 %~0.32 % for trifolirhizin,0.05 %~0.51% for nor- kurarinone,0.55?10~(-3)%~4.87?10~(-3)% for xanthonumal and 0.06 %~0.23 % for kuraridin.Conclusion The contents of 4 flavonoids in the samples from the market vary notably,and this should be taken into the quality control of Sophora flavescens Ait.
3.Comparative studies on codon usage bias of Ganoderma lucidum based on analysis of genomic and transcriptomic data.

Codon usage bias is an important characteristic of genetic information transfer in organisms. Analysis of codon usage bias of different species is important for understanding the rules on genetic information transfer. The previous method for analysis of codon usage bias is mainly based on genomic data. However, this method is greatly limited, because the genome sequences of higher organisms are still not available up to now. In this study, we found that we could obtain the same optimal codons of Ganoderma lucidum (Curtis: Fr.) P. Karst based on its whole genomic data or large-scale transcriptomic data from its liquid-cultured hyphae, primordium and fruiting body, separately. This result indicated the feasibility to understand the codon usage bias based on the large-scale transcriptomic data. By calculating the proportion of rare codons of Escherichia coli and Saccharomyces cerevisiae in 26 terpene synthases (TS) of G. lucidum, we found that the rare codons of S. cerevisiae have a higher proportion in TS genes, while the rare codons of E. coli have relatively lower, suggesting that the TS genes of G. lucidum are possibly more difficult to be expressed in S. cerevisiae than in E. coli. Chemical synthesis of TS genes according to the yeast optimal codons will be an effective way to solve the problem on the mismatch of gene codon bias between the foreign genes and the host strain.

4.Analysis of 572 Cases of Drug-induced Liver Injury Induced by Anti-infective Agents

OBJECTIVE:To investigate the condition and characteristics of drug-induced liver injury (DILI) of anti-infective agents and provide reference for the prevention and treatment of anti-infective agents related DILI. METHODS:Based on retrospective analysis,a total of 572 DILI reports of anti-infective agents were collected from PLA ADR monitoring center during 2009 to 2013, and then analyzed statistically in terms of patient’s age and gender,main diagonosis,categories of DILI-inducing drugs,type,route of administration,occurrence time,lab indicator,DILI types and clinical manifestations,the application of liver protective drugs,out-comes,etc. RESULTS:Among 572 DILI cases,there were 412 cases(72.03%)of male patients and 160 cases(27.97%)of female patients,and average age of the patients was(44.54±23.75)years old. ADRs were related to 57 kinds of anti-infective agents in 6 cat-egories. Rifampin was the most frequent in suspected drugs,followed by isoniazid,moxifloxacin,fluconazole,azithromycin,cefurox-ime, cefoperazone/sulbactam, levofloxacin, cefoxitin and voriconazole. Intravenous infusion was the main administration route (74.48%). The occurrence time of ADRs was mainly within two weeks (86.19%). Hepatocellular damage (93.33%) was the main type in 360 cases of ADR for evaluation of liver injury types. The majority of cases(82.17%)were cured or improved after drug with-drawal and symptomatic treatment. CONCLUSIONS:Cephalosporin,fluoroquinolones,antituberculosis and antifungal drugs might be the common agents which caused liver injury. Hepatocellular damage is the most frequent type. Most of patients have a favourable prognosis. Clinical medical staffs should strengthen the awareness of DILI caused by anti-infective agents and ehance the prevetion of it.
5.Effects of thoracoscopic lobectomy on early-stage non-small cell lung cancer

Objective To discuss effects of thoracoscopic lobectomy on early-stage non-small cell lung cancer . Methods 60 patients with non-small cell lung cancer patients were randomly divided into two groups of 30 cases in each group using thoracoscopic lobectomy (VATS) treatment,as the VATS group,another group using traditional open surgery(TOS) therapy,TOS as a group.(1) Operative time,blood loss,postoperative drainage time,drainage, postoperative pain,postoperative bed days,after several days of hospitalization were compared between the two groups . (2) 1,3 and 5 days after surgery,serum C-reactive protein(CRP) and interleukin-6(IL-6)levels were compared. Results (1)were operative time,postoperative drainage time,pain time,bed time and hospitalization time in VATS group were (69.50 ±15.33)min,(60.23 ±9.75)h,(46.75 ±7.36)h,(3.54 ±1.58)d,(9.50 ±2.50)d,com-pared with TOS group (86.77 ±19.56)min,(66.50 ±10.50)h,(58.36 ±9.58)h,(5.50 ±1.55)d,(13.55 ± 3.18)d short d observed group blood loss ,drainage volume(161.55 ±25.54)mL,(155.75 ±23.18)mL,compared with TOS group(186.58 ±25.35)mL,(169.65 ±26.87)mL less,the difference was statistically significant (t =39.14,28.19,54.11,6.03,7.24,98.36,76.34,all P<0.05).(2) VATS group after 1,3 and 5 days CRP serum and were(79.58 ±22.15) mg/L,(97.23 ±28.16) mg/L,(98.44 ±27.19) mg/L,IL-6,respectively (69.36 ± 17.32) ng/L, ( 96.33 ±25.67 ) ng/L, ( 103.16 ±28.89 ) ng/L, were higher than the TOS group ( 97.84 ± 25.37)mg/L,(119.17 ±31.77)mg/L,(130.81 ±33.29)mg/L,(76.83 ±21.97)ng/L,(121.15 ±32.64)ng/L, (122.08 ±30.74)ng/L,the difference was statistically significant (t=70.12,58.33,69.17,64.34,79.58,92.17,all P<0.05).Conclusion VATS lobectomy for non-small cell lung cancer has better effects and safety .
6.The preventive effect of Chinese medicine with Replenishing kidney and invigorating Qi on rabbits osteoarthritis

Objective To explore the mechanism of preventive effect of Chinese medicine with Replenishing kidney and invigorating Qi on osteoarthritis. Methods 72 male Long-eared white rabbits aged 4 months were randomly divided into six groups, A group (blank control group), B group (model group), C group (traditional Chinese medicine high-dose prevention group), D group (Chinese herbal medicine with medium-dose prevention group), E group (Chinese herbal medicine with low-dose prevention group), F group (glucose-amino acid hydrochloride capsules prevention group).All the animals apart from A group were established osteoarthritis model by immobilizing knee joint with plaster cast for 6 weeks. In the same time of immobilization, traditional Chinese medicine and glucose-amino acid hydrochloride capsules were given to C, D, E and F group for 4 weeks. Physiological saline was given to B group. After 6 weeks of modeling, synovial fluid was extracted and the changes of TNF-α in it were analyzed by ELISA. Results Articular cartilage degeneration of B group was most obviously compared with C, D, E and F group. There was a significant difference of TNF-α level in comparison of A and B group (P<0.05), B and C, D, E and F group (P<0.05), and among C, D and E group. Conclusion Chinese medicine with replenishing kidney and invigorating Qi can prevent osteoarthritis by reducing TNF-α level in synovial fluid, enhancing cartilage cell metabolism, and slowing down cartilage degradation.
7.Proteotyping: A New Approach Studying Influenza Virus Evolution at the Protein Level

Phylogenetic methods have been widely used to detect the evolution of influenza viruses.However, previous phylogenetic studies of influenza viruses do not make full use of the genetic information at the protein level and therefore cannot distinguish the subtle differences among viral genes. Proteotyping is a new approach to study influenza virus evolution. It aimed at mining the potential genetic information of the viral gene at the protein level by visualizing unique amino acid signatures (proteotypes). Neuraminidase gene fragments of some H5N1 avian influenza viruses were used as an example to illustrate how the proteotyping method worked. Bayesian analysis confirmed that the NA gene tree was mainly divided into three lineages. The NA proteotype analysis further suggested there might be multiple proteotypes within these three lineages and even within single genotypes. At the same time, some proteotypes might even involve more than one genotype. In particular, it also discovered some amino acids of viruses of some genotypes might co-reassort. All these results proved this approach could provide additional information in contrast to results from standard phylogenetic tree analysis.
8.The application of intermittent splenic artery occlusion for treating iatrogenic splenic injury: an animal experiment

Objective To evaluate the value of intermittent splenic artery occlusion in the treatment of iatrogenic splenic injury and the cause of ischemia reperfusion injury.Methods Experimental animals using dogs were divided into experimental group and control group according to whether the splenic artery was blocked.The animal model of iatrogenic splenic injury was established by general anesthesia.Experimental group was treated with intermittent splenic artery blockage combined with gelatin sponge oppression of the wound to stop bleeding.The first time of splenic artery occlusion was 15 min and continued for 10 min and for another 5 rmin if still bleeding.Then hemostatic effect after removing blocking and oppression was observed.Control group was only treated with gelatin sponge compression to stop bleeding.Blood samples and tissue samples were collected before blocking and after reperfusion for 2 hours in experimental group,and before compression and relieving the compression for 2 hours in control group.IL-1 (interleukin),SOD (superoxide disproportionation enzyme),MPO (myeloperoxidase),MDA (malondialdehyde) and caspase-3 (aspartame acid cysteine protease) were detected.Results The control group still had bleeding,while the experimental group had successfully stopped bleeding.Serum markers before and after occlusion in the experinental group were IL-1 (124.4 ± 106.8 vs.121.2 ± 105.1),SOD (4.7 ± 2.7 vs.5.2 ± 1.8) and MDA (8.8 ±6.5 vs10.8 ±7.5).There were no significant differences on serum markers in two groups after occlusion/oppression including IL-1 (121.2 ± 105.1 vs.162.8 ± 73.8),SOD (5.2 ± 1.8 vs.4.7 ± 2.8) and MDA (10.8 ±7.5 vs.9.6 ±6.6) (P>0.05).Histological indicators before and after occlusion in the experimental group included MPO (0.62 ±0.23 vs.0.68 ±0.21) and Caspase-3 (0.90 ±0.29 vs.0.86 ± 0.26),and there was no statistical difference on MPO (0.68 ±0.21 vs.0.86 ±0.23 after two sets of experiments) and Caspase-3 level (0.86 ± 0.26 vs.1.21 ± 0.18) (P > 0.05) in two groups after occlusion/ oppression.Conclusion The therapeutic effect of intermittent splenic artery occlusion combined with gelatin sponge compression hemostasis is safe and effective and without obvious ischemia reperfusion injury.
9.Biodistribution and radioimmunoimaging of 131 I-Herceptin in healthy KM mice and nude mouse models bearing human ovarian cancer xenografts

Objective To study the biodistribution of anti-HER-2/neu monoclonal antibody Herceptin labeled by 131I(131I-Herceptin) in healthy KM mice and nude mice bearing human ovarian cancer xenografts and radioimmunoimaging (RII) of the nude xenografts-bearing mice.Methods 131I-Herceptin was prepared using Iodogen method.The labeling efficiency, radiochemical purity, stability and immunocompetence were measured.The percentage activity of injection dose per gram of tissue (%ID/g) and the radioactivity ratio of tumor to non-tumor tissue (T/NT) were calculated for each time point.The optimal time for imaging was investigated by comparing the 131I-Herceptin SPECT for the nude mouse models bearing ovarian cancer xenografts at different time points.Results The labeling efficiency and radiochemical purity of 131I-Herceptin were 89.8% and 98.4%, respectively.The labeling was stable and had good immunocompetence.131 I-Herceptin was cleared rapidly mainly through liver, spleen and kidneys, consistent with first order two-compartment model.The uptake of 131I-Herceptin in the tumors bearing human SKOV-3 xenografts was much higher than that in nontumor tissue.The% ID/g was 18.08 in the tumor at 24 h post injection.The T/NT ratio increased with time and was 27.27 at 72 h post injection.The tumors in nude mice bearing SKOV-3 xenografts could be visualized on 131I-Herceptin SPECT imaging 2 h post injection; definitely identiffed 48 h post injection and the radioactivity ratio of tumor to contralateral tissue was 11.44 at 120 h post injection.However, the tumor in nude mice bearing HO-8910 xenografts did not show abnormal uptake of 131 I-Herceptin at each time point.Conclusions 131 I-Herceptin is a good radiopharmaceutical targeting SK-OV-3 xeuografts and it may be useful in imaging carcinoma of ovary and target therapy of its metastases with high HER-2/neu expression.
