1.TMBS-a new Nonpoisonous Substrate of HRP for Demanstrating Specific Antibody in Enzyme Immunoassay
Chinese Journal of Immunology 1985;0(02):-
This paper reported a new home-made nonpoisonous substrate of horseradish peroxidase(HRP),3,3′,5,5′-tetramethylbenzidine sulphate(TMBS),used in ELISA with the aqueous soluble egg antigen(SEA)as antigen;proved patients'sera as antibo- dy and the antihuman IgG was replaced by protein A conjugate. The optimun condition obtained by orthogonal design was 1.0 millimolarity of TMBS,1.2 millimolarity of H_2O_2,and pH value at 5.0.The best time to stop reaction with H_2SO_4,was 30-60 min after adding substrate TMBS.At the same time,TMB and OPD were used as compared substrates. It showed that the reaction colour of both TMBS and TMB remained much more stable than that of OPD when being exposured in light and oxygen. After the solutions of TMBS and TMB were stored under 4℃ in refrigerator for 60 days, the coefficient of variation among different experiments was less than 10%. The results in detecting specific antibody in sera from 28 patients with proved schistosomiasis also showed that the sensitivity of TMBS was remarkably higher than OPD.It was found that TMBS used in enzyme immunoassay for demonstrating specific antibody was suitable not only for optical density(OD)read by spectrophotometer but also for estimation judged by naked-eye.During TMBS and TMB used in indirect immunoperoxidase method(IIP),the specific antibody in the tissue can be easily dete- cted and located as well.

Acta Pharmaceutica Sinica 2010;45(7):895-900
The paper is aimed to provide a novel index, named as multidimensional spatial triangular area, for the evaluation of the release-absorption correlation of multiple component traditional Chinese medicines (TCMs). The applicability of the method was demonstrated by the example data. The method and standard practice for evaluation of the release-absorption correlation for western medicines with single compound could not be applied to TCMs with multiple components. The release percentage or absorption percentage of the multiple components for TCMs at the sampling time was a point in the multidimensional space. The area of the triangle formed byt the sequential three points rrepresented the changing characteristics of the components' release and absorption kinetics. The side lengths of the triangle could be calculated from the spatial distances between each two of the sequential three points. Then the triangle area could be obtained by the side lengths. The in vitro release-in vivo absorption correlation of the multiple components could be represented by the correlation between the integrating values of the release triangle areas and that of the absorption triangle areas. The results of the examples indicated that the multidimensional spatial triangular area method could treat the multiple components in a holistic way, in line with the holism the hi he TCMs. Therefore, the multidimensional spatial triangular area method provided new methodology for the release-absorption correlation of the TCMs with multiple components.

Objective:To investigate the effect of genistein,a soybean-derived isoflavone,on thestimulating effect on bone resorption of IL-1?.Methods:The osteoclasts(OCs)were isolated with themethods of Yu Shifeng. The rat calvaria were cultured as an organ. The cells in the experiment weregrown in four respectively:Control(without genistein or IL-1?),10~(-6) mol/L genistein,10 ?g/L IL-1?and 10~(-6) mol/L genistein+10 ?g/L IL-1?.The area of bone resorption, the concentration of Ca~(2+) in thesupernatant liquid of OCs cultures and mice calvaria were tested. The contents of acid phosphatase(ACP)were also examined by biochemistry method. The index of bone resorption was counted as the ra-tio of the experiment average and control ones, which indicated the increase in bone resorption when itwas above 1.0.Results:The area of bone resorption of 10~(-6) mol/L genistein+10 ?g/L IL-1? increasedcompared with that of 10~(-6) mol/L genistein, while the concentration of Ca~(2+) in the supernatant liquid ofOCs cultures decreased significantly. The index of bone resorption of 10~(-6) mol/L genistein+10 ?g/L IL-1? lied between 10~(-6) mol/L genistein and 10 ?g/L IL-1?.In the organ culture, there was no differencein the content of ACP among all the groups,The index of bone resorption of 10~(-6) mol/L genistein+10?g/L IL-1? was below that of 10?g/L IL-1?,but both were above 1.0.The index of bone resorptionwas below 1.0 in the group of 10~(-6) mol/L genistein. Conclusion:Genistein can suppress obviously thebone resorption simulated by single IL-1?.

Objective: To evaluate pharmacokinetic and pharmacodynamic of repaglinide tablets in Chinese subjects. Methods: Twenty healthy male volunteers were enrolled in the study. A single dose (4 mg) of repaglinide tablets was givenorally. Plasma concentrations of repaglinide were determined by HPLC method. Blood glucose and serum insulin leve1s weremeasured by biochemistry and radioimmunoassay methods respectively. Results: Plasma concentration-time curve conformedto one-compartment open model. The pharmacokinetic parameters were as follows: tmax (0. 75?0.43 ) h,cmax (54.44?24.97)ng/ml, t1/2 (0. 80?0. 31) h, MRT (1. 55?0. 41) h, C1/F (61. 43?20. 10) L/h and AUC (73. 34?29.95) h? ng/ml. Thelevel of serum insulin was raised and the level of blood glucose decreased after administration of repaglinide. The highest levelof serum insulin was (l26. 24?95.93) mU/L at 0.75h and blood glucose level reached its lowerest vaIue (2. 34I0.44) mmol/L 1 h after oral administration. Conclusion: Repaglinide is characterised by fast-acting and short effects on in-sulin secretion. It decreases serum glucose level by stimulating insulin secretion from the pancreatic ?-cells. It is a novel oralprandial glucose regulator for the treatment of type 2 diabetes mellitus.

China Pharmacy 2016;27(5):635-637
OBJECTIVE:To explore the characteristics and regularities of the adverse drug reactions(ADRs)induced by irino-tecan,in order to provide reference for clinical application. METHODS:In retrospective study,56 cases of irinotecan-induced ADR collected from our hospital during Jun. 2011-Jan.2015 were analyzed statistically in respects of gender, age, disease types, ECOG score,medication,organs/systems involved in ADR,clinical manifestations, outcome, etc. RESULTS:All 56 patients suf-fered from ADR,among which the percentage of hematopoietic system accounted for the highest(100%),and followed by diges-tive system;the grade Ⅰ-Ⅱ ADRs accounted for 87.20%,the grade Ⅲ-Ⅳ ADRs accounted for 12.80%. 64.50% of ADRs oc-curred within 1 week. In respect of conversion,85.71%(48/56)of the patients recovered with supportive care and continued che-motherapy,the remaining 8 cases had to reduce the dose of chemotherapy and then recovered. CONCLUSIONS:With regular dose, irinotecan is well tolerated. Since ADRs induced by irinotecan are slight, all the ADRs are well managed. It is important to enhance follow-up observations of patients to prevent some serious ADRs.
