1.Fast and Almost 100% Efficiency Site-directed Mutagenesis by The Megaprimer PCR Method

A novel PCR-based mutagenesis method was reported, in which there is no need to purify megaprimers or design a special flanking primer. This method used one mutagenic primer and two sequencing primers (T_m≤58℃) as flanking primers. After first round PCR, 12.5 μl first PCR production was directly added into 50 μl second PCR system as template and megaprimer, and 10 rounds of asymmetrical PCR at high temperature of annealing (68 ℃ ) was to add in initiation of second PCR. This additional step greatly has increased the efficiency of mutagenesis via 600 bp or 800 bp long megaprimer. The results demonstrated that this method can achieve high fidelity, 97%~ 98% efficiency, high yield.
2.Study progress of dental pulp stem cells in tissue engineering.

In recent years, modern tissue engineering is becoming emerging and developing rapidly, and the acquisition, cultivation and differentiation of seed cells is the premise and foundation of the construction of tissue engineering, so more and more scholars pay attention to stem cells as seed cells for tissue engineering construction. Dental pulp stem cells (DPSCs) is a kind of adult stem cells derived from dental pulp, and as a new kind of seed cells of tissue engineering, the study of DPSCs presents important significance in tissue and organ regeneration. In this review, we introduced the progress of studies on dental pulp stem cells and discussed their clinical application prospects.

3.Research progress of tumor cells and tumor microenvironmentderived exosomal miRNAs in carcinogenesis, tumor diagnosis and treatment

Exosomal miRNAs, derived from tumor cells or their microenvironment, could promote the proliferation, migration and invasion of tumor cells;and enhance tumor metastasis via regulating information exchange between tumor cells and immune cells or metastatic target organs;and induce tumor resistance to cisplatin and gemcitabine;meanwhile, detecting the exosomal miRNAs in the serum and saliva of cancer patients suggested the potential of application in cancer diagnosis and prognosis assessment.
4.Olfactory ensheathing cell transplantation in treatment of spinal cord injury:an update

The treatment of spinal cord injury is always a stubborn problem for neurosurgeons because nerve cell cannot regenerate by itself and the glia scar can prevent the axonal regeneration. Therefore, to decrease the neuron death and formation of scar and to increase axonal regeneration are the keys for clinical treatment. Many studies showed that the olfactory ensheathing cells(OEC) can promote axonal regeneration and prover axonal growth,bringing hope for treament of spine injury, but the result is not satisfactory in clinic. In order to improve the clinical effect of OES transplantation many studies combine it with genetic engineering and tissue engineering and some progress is made.
5.Effects of soy oligosaccharides and peptides on blood lipid metabolism of rats

Objective To investigate the effects of soy oligosaccharides and peptides on lipid levels in rat plasma.Methods Sixty healthy SD rats were randomly divided into five groups fed respectively with normal diet(ND group),high-fatty diet(HFD group),HF diet+2% soy oligosaccharides(SO group),HF diet + 3% soy peptides(SP group),HF diet+2% soy oligosaccharides+3% peptides(SOP group) for 8 weeks,and the concentrations of blood lipid and apolipoprotein were measured at 28 d and 56 d.Results As compared with HFD group,the serum levels of total cholesterol(TC),triglycerol(TG),very low density lipoprotein cholesterol(VLDL-C) and low density lipoprotein cholesterol(LDL-C) and atherosclerosis index(AI) in SO,SP and SOP groups decreased significantly,while high density lipoprotein cholesterol(HDL-C) increased significantly;the level of plasma apolipoprotein A-Ⅰ(apoA-Ⅰ) increased significantly,while apolipoprotein B(apoB) and apoB/apoA-Ⅰ ratio decreased significantly,especially in SOP group.Conclusion Soy oligosaccharides and peptides can prevent hyperlipidemia by adjusting blood lipid metabolism in vivo effectively,especially when they are combined.
6.INVESTIGATION ON PROTECTIVE EFFECTS OF DNA VACCINE PCDSJ32 FORSCHISTOSOMA JAPONICA BY DIFFERENT INJECTION TIMES AND CHALLENGE TIME AFTER THE LAST INOCULATION IN MICE

Aim Protective immunity of DNA vaccine pCDSj32 for Schistosoma japonica by different injection times and challenge time after the last inoculation in mice was investigated. Methods 100μg/100μl DNA vaccine pCDSj32 was injected into the quadriceps muscles of the BALB/c mice. The mice were divided into once immunized groups and three times immunized groups. Challenge infection was applied 4wk or 8wk after the last inoculation with 40 cercariae each mouse. The adult reduction rates and the EPG in the livers of mice were detected 6wk after challenge. Results Substantially protective immunity was obtained when compared the immunized groups with the control. The adult reduction rates were 35.61~44.44 % ,and the egg reduction rates in the livers were 39. 64~69.06%. Conclusion The protective immunity could be influenced by different times of DNA vaccine inoculation and the challenge time after the last immunization. The best protective results could be obtained with inoculation once combined with the challenge 8wk after the immunization of DNA vaccine.
7.Discussion on teaching of medical aesthetics in military medical university

Aesthetic qualities of the medical staff directly affect the quality of disease prevention and control.Military medical university is responsible for the task of training highly qualified military medical personnel.Medical aesthetics education in military medical university is necessary not only for medical development and military modernization,but also for the overall improvement of the quality of medical students.The meaning of medical aesthetics teaching,teaching content and teaching methods were discussed in this article in combination with medical aesthetics teaching practice carried out in military medical university,aiming at strengthening medical aesthetics education of cadets and training them to be social and military needed high-quality medical personnel.
8.FUNGAL INFECTION FOLLOWING ABDOMINAL SURGERY

Objective:To inquire into the prophylaxis and treatment of fungal infection following abdominal surgery.Methods: Retrospectively analyzed the diagnoses,the inducing factors, the treatments and the progreoses of 136 cases fungal infections complicated by abdominal sugery from 1990 to 1999 June. Results : Analyzed and explained that the fungal kinds and invasions relate to the abuse of artibiotic, oldness and weakness, the use of immunosuppression and the low immunity function. Couclusion : We should raise the cognition of the fungal infection following abdominal surgery and achieve early diagnosis and prompt treatment in order to avoid the serious consequences of fungal septicemia and multiple organ dysfunction syndrome.
10.Effect of Early Rehabilitation Nursing on Secondary Disturbance in Hemiplegics after Stroke

ObjectiveTo study the effect of early rehabilitation nursing on secondary disturbance in hemiplegics after stroke.Methods140 patients were randomly divided into two groups , rehabilitation nursing group (70 cases) and control group (70 cases). Patients in the rehabilitation nursing group were given clinical treatment and regularly convalescent nursing, while those in control group were given clinical treatment and unguided self-training. Evaluation was performed in pre-treatment and 12 weeks post-treatment respectively.ResultsSecondary disturbance rate in the rehabilitation nursing group was significant lower than that in the control group (P<0.01). The motor scores and ADL scores measured by Barthel Index in both groups raised after 12 weeks treatment, but the scores in the early rehabilitation nursing group were obviously superior to those in the control group (P<0.01).ConclusionEarly rehabilitation nursing for hemiplegics afte sroke may prevent or lessen the secondary disturbance, as well as obviously improve motor function of the limbs and ADL.
