1.Research development and clinical significance of genetic polymorphism of CYP450

Cytochrome P450 is the very most important enzyme of drug metabolism.It involves in biotransformation of various endogenous and exogenous compounds,especially for clinical drugs.There are many polymorphisms of gene and phenotype,resulting in significant individual variability of different kinds of compounds.This article reviews the genetic polymorphisms of CYP,the characters and clinical meanings of diverse subgroups that relevant to drug metabolism in recent years.The major object of this article is reasonably explaining and predicting the interaction of clinical drugs and the drug adverse reaction in order to providing scientific evidences for the individual administration in clinic.
2.Effects of mindfulness - based stress reduction on compliance in primary open angle glaucoma patients with topical medication

AIM: To explore the effects of mindfulness- based stress reduction ( MBSR ) on anxiety, compliance and visual function in primary open angle glaucoma( POAG) patients. ·METHODS:Totally 64 patients, who were treated with topical medication and were diagnosed with POAG in the department of ophthalmology at Shaanxi Provincial People's Hospital, were randomly divided into the intervention group ( n=32 ) and the control group ( n=32 ) . Both groups received conventional mental health education while the intervention group was complemented with MBSR intervention. The patient's anxiety status, compliance rates and MD before and after the intervention were evaluated. ·RESULTS:After the MBSR intervention, the scores of HAMA of the intervention group were significantly lower than that of the control group (P<0. 01). The compliance of patients in intervention group was higher than that of the control group (P<0. 01). ·CONCLUSION:The MBSR intervention can effectively relieve the anxiety of patients and improve the compliance in primary open angle glaucoma patients. It can be promoted and put into clinical application.
3.E-cadherin promoter methylation and demethylation in epithelial ovarian carcinoma cells

Objective To investigate the cytidylyl phosphate guanosine(CpG) islands methylation status of E-cadherin (E-cad) promoter region in human ovarian carcinoma cell lines (ES-2,3 AO, SKOV3 ), and the effect of 5-azacytidine-2 '-deoxycytidines (5-Aza-CdR ) on the cell proliferative ability, invasion and the expression of E-cad protein. Methods Methylation specific PCR(MSP) was used to detect CpG islands methylation status of E-cad promoter region in ES-2,3AO and SKOV3 cell lines. After treated with different concentrations of 5-Aza-CdR, morphological changes of cell lines were observed under microscope. The proliferative ability was evaluated by methyl thiazolyl tetrazolium(MTT) assay. E-cad protein expression was detected by western-blot and cellular invasion was investigated by 24-well matrigel invasion chambers. Results Hypermethylatian status of CpG islands of E-cad promoter region was observed in ES-2 and SKOV3 cell lines, but not in 3AO cell lines. After treated with 5-Aza-CdR (0.1,1.0,10.0 μmol/L), ES-2 and SKOV3 cell lines displayed morphological evidence of differentiation. 5-Aza-CdR was found to decrease proliferation as evidenced by cell growth curve , to increase the level of E-cad protein expression (P < 0.01 ), and effectively inhibit the ability of cell invasion(P <0.01 ). Conclusions CpG hypermethylation is an important mechanism of E-cad gene inactivation in ES-2 and SKOV3 cell lines. 5-Aza-CdR be found to inhibit proliferation and invasion, and increase the expression of E-cad probably by the inhibition of hypermethylation.
4.Effect of basic fibroblast growth factor on the formation of blast-colony-forming cell derived from embryonic stem cells

BACKGROUND:Some studies show that basic fibroblast growth factor(bFGF)strongly expresses dudng the process of embryonic stem cells differentiation into hematopoietic stem cell.yolk sac blooding.and fetal liver hematopoiesis.OBJECTIVE:To study the regulation of bFGF on the blast-colony-forming cell(BL-CFC)by adding bFGF in the medium of embryoid body generation.METHODS:The third to fifth generations of the pdmaw mouse embryonic fibroblasts were recovered,and then incubated with the DMEM medium containing mitomvcin C for 2.5 hours in order to lose the proliferative capacity.Then cells were suspended into single cell by trypsinization and inoculated in the gelatin-coated bottle at the density of 10×104/cm2.After culturing for 24 hours,mouse embryonic stem cells(mESC)of D3 were recovered and placed on the feeder layer cells.According to the composition of medium in embryoid body generation.mESCs were divided into two groups:group A:standard medium+VEGF+SCF;group B:standard medium+VEGF+SCF+bFGF.Each group was cultured for 3 days and 6 days respectively,and the cloning number of BL-CFC was quantified,as well as Flk-1+ expression was observed by immunofluorescence staining.Positive number and average absorbance were analyzed using IMAGE-PRO PLUS imaging analysis system.RESULTS AND CONCLUSION:Adding bFGF in the course of embryoid body growth could significantly increase the number of BL-CFC(P<0.01).and the positive results of Flk-1 and the average absorbance were also increased significantly(P<0.01).bFGF effectively promoted embryoid body amplification and proliferation of BL-CFC.
5.Signal molecules related to self-renewal of hemopoietic stem cells: Effects and pathways

BACKGROUND: There are small amount of hematopoietic stem cells, which are easy to divide in vitro and show the difficulty in applying to transplantation.OBJECTIVE: This paper has focused on the role and means of the Wnt, Notch, Bmi_1, Shh, HOXB4 signaling molecule in the maintenance of hematopoietic stem cell self-renewal and regulation. METHODS: With the key words of "HSC, Wnt, Notch, Bmi_1, Shh, HOXB4" for the search, we searched PubMed database (2002-01/2008-12) in English. Literatures closely related to the hematopoietic stem cell self-renewal related signaling molecules were included. Repetitive research and Meta analysis were excluded.RESULTS AND CONCLUSION: The computer initially retrieved 216 documents, of which 30 documents for research. Hematopoietic stem cells are self-renewing, have strong differentiation and growth and regeneration capacities, can produce various types of blood cells ancestor cells, are widely used to treat blood diseases, but the hematopoietic stem cell differentiation in vitro demonstrated that the difficulties used in transplantation. How to make hematopoietic stem cells in vitro amplification and processing, while maintaining hematopoietic stem cell self-renewal characteristics is of a key issue. In recent years, different signaling pathways to enhance the ability of hematopoietic stem cell self-renewal signaling molecule have been a research hotspot. The article focused on the role and means of the Wnt, Notch, Bmi_1, Shh, HOXB4 in the maintenance of hematopoietic stem cell self-renewal and regulation and found that both the above-mentioned five signaling molecules can enhance hematopoietic stem cell self-renewal function. There are also a number of factors playing an important role in the maintenance of hematopoietic stem cell self-renewal process, such as endogenous factors, a series of transcription factors Oct-4, Ehox, Nanog, SCL, Runx1 and so on, to explore how their regulatory networks formed by the interaction control self-renewal of hematopoietic stem cells will become a key point in the research of self-renewal of hematopoietic stem cells.

7.Relationships among Social-emotional Competence, Adaptive Behavior and Language Skills of Hearing-impaired Children

Objective To explore the influence of social-emotional competence and adaptive behavior on language skills of hearing-impaired children. Methods 68 hearing-impaired children aged 1~3 years were investigated with questionnaire. Results Externalizing behavior and deregulation negatively correlated with language skills (P<0.05), while communication positively correlated (P<0.05). Multiple regression showed that externalizing behavior was the independent factor related with language skills of hearing-impaired children. Conclusion Social-emotional competence relates with the language skills of hearing-impaired children.
8.Effects and mechanisms of low concentration dopamine on hydrogen peroxide-induced apoptosis in cultured neonatal rat cardiomyocytes.

OBJECTIVETo study the effects of low concentration dopamine(DA) on hydrogen peroxide-induced apoptosis in cultured rat cardiomyocytes as well as the possible molecular mechanisms.
METHODSCultured neonatal rat cardiomyocytes were randomly divided into the following groups: control group (control), hydrogen peroxide group (H2O2), pretreated with low concentration dopamine ( DA + H2O2), dopamine receptor l(DR1) antagonist group (DR1 + DA + H2O2), dopamine receptor 2(DR2) antagonist group (DR2 + DA + H2O2). The cell apoptosis was then assessed by MTT and flow cytometry. The cellular ultrastructure changes were observed by transmission electron micro- scope. The activity of lactate dehydrogenase(LDH )and superoxide dismutase (SOD) in cell medium was analyzed by colorimetry. The protein expressions of Cytochrone c, Caspase 3 and Caspase 9 were obtained by Western blot.
RESULTSCompared with hydrogen peroxide group, low concentration dopamine(10 µmol/L) decreased the apoptosis rate and the expression of protein of apoptosis related protein, enhanced SOD activity, decreased LDH activity. DR1 antagonist SCH-23390 treatment inhibited dopamine induced cardiac protective effect. DR2 antagonist haloperido treatment had no changes compared with dopamine group.
CONCLUSIONAbove findings indicate that low concentration dopanine inhibits apoptosis induced by hydrogen peroxide in neonatal rat cardiomyocytes, which is partly associated with the activation of DR1.

9.Effects of androgen on autoimmune ovary failure of mouse

Objective To identify the effects of androgen on mouse model of autoimmune ovary failure.MethodsFifty mice were randomly divided into five groups as control group,early therapeutic group and its model group,late therapeutic group and its model group.Autoimmune POF was induced in forty mice by Pzp3.Then,the animals were randomly divided into four groups: two were treated with androgen from the 15th and 20th day after immune.The control group was treated with distilled water.The therapeutic efficacy of each treatment was evaluated by the changes in sexual cycles of mice,the serum level of AzpAb,infiltration of cells positively expressing CD45 in the ovary as well as the percentage of growing follicles of the ovary.Results The sexual cycles,serum level of AzpAb,infiltration of CD45 cells in the ovary of each model group were more significant than those in control group;early therapeutic group was less than model group;and serum level of AzpAb in early therapeutic group was lower than that of late therapeutic group.The percentage of growing follicles in the ovary of each model group was lower than that of control group;early therapeutic group was higher than model group and late therapeutic group.Conclusion The use of androgen in mice with autoimmune POF may notably ameliorate the pathogenetic conditions of the disorder,and the treatment during prophase was apparently more effective than that during the advanced stage.
10.Analysis of the medical treatment willingness and place among migrant workers in some areas of Shanghai and relevant influencing factors

Objective · To understand the medical treatment willingness and place among migrant workers in Shanghai and relevant influencing factors. Methods · The questionnaire survey was used to investigate migrant workers in some areas of Shanghai and the data were analyzed with constituent ratio,χ2 test, and Logistic regression. Results · Regional differences in medical treatment willingness and place existed between migrant workers in surveyed areas and local workers. Differences between actual and intended medical treatment places existed among migrant workers. Main factors influencing the medical care seeking behavior of migrant workers were medical insurance, economic factors, and demographic factors. Conclusion · The medical security system and the accessibility of basic medical services for migrant workers should be further improved.
