1.Induction of human mesenchymal stem cells in vitro and related identification

OBJECTIVE To establish the isolation and identification system of human mesenchymal stem cells(hMSCs)in vitro, and then to establish a stable cultured system in which the hMSCs can be induced to osteoblasts, chondroblasts and lipoblasts in vitro. To study the possibility if hMSCs can be used as the seed cells in bone tissue engineering.This study would lay a foundation for the clinical application of tissue engineering. METHODS 5ml bone marrow of the patient was aspirated from iliac crest. The human MSCs were isolated using Percoll gradient centrifugation. The hMSCs were cultured in DMEM medium in vitro, FACS(fluorescence axtivated cell sorter)was used to identify the phenotype of hMSCs. The hMSCs were cultured in vitro, and induced to osteoblasts, chondrocytes and lipocytes. The changes of phenotype were tested by mmunohistochemisty and molecular biology technique. RESULTS The hMSCs were isolated by Percoll gradient centrifugation from patient’s bone marrow. The expression rate of MSC markers including CD105, CD166, CD29 were(78?6)%, (43?7)%, (69? 12)% respectively. The phenotype of osteoblasts induced from hMSCs was verified by the mineralized nodes formation when cultured in vitro and Col, and OCN expression showed by immunohistochemistryand RT-PCR. The phenotype of chondrocytes induced from hMSCs was verified by type, collagen, SOX9 expression with immunohistochemical staining technique and type, collagen, aggrecan mRNA expression with RT-PCR. The lipocytes’ phenotype was verified by positive result of oil red O staining and PPAR 2mRNA expression by RT-PCR. CONCLUSION The highly pure hMSCs can be harvested by means of density gradient centrifugation, and the hMSCs have multi-potentiality of cell differentiation via induced culture in vitro. The third passage of hMSCs can be induced to express osteoplast phenotype and meet the qualitative and quantitative demands of seed cells when bone tissue engineering was used in clinical practice.
2.Hotspots in text mining of biomedical field

The high frequency subject terms were extracted from the PubMed-covered papers published from January 2000 to March 2015 on text mining of biomedical field to generate the matrix of high frequency subject terms and their source papers.The co-occurrence of high frequency subject terms in a same paper was analyzed by clustering analysis.The hotspots in text mining of biomedical field were analyzed according to the clustering analysis of high frequency subject terms and their corresponding class labels, which showed that the hotspots in text mining of bio-medical field were the basic technologies of text mining, application of text mining in biomedical informatics and in extraction of drugs-related facts.

The external stimuli and other reasons may result in hyperplasia and abnormal angle of the upper corner of the thyroid cartilage. The upper corner of the thyroid cartilage has anatomical variations. To oppression stimulate carotid artery, cervical sympathetic dry section and so on, could causing pharyngeal foreign body sensation and sore throat. By surgical removal of the thyroid cartilage hyperplasia upper corner, postoperative symptoms would be resolved.

4.The study of resveratrol protective effects and mechanism on tissue-engineered cartilage

Objective To investigate the mechanism of protective effects of resveratrol on tissueengineered cartilage.Methods The chondrogenesis of alginate-encapsulated bone marrow mesenchymal stem cells (BMSCs) were evaluated by toluidine blue staining and immunostain.The morphology of BMSCs-derived chondrocytes cultured on chitosan-gelatin scaffolds (CGS) was evaluated by scanning electron microscope and laser confocal microscope.When these cells on CGS were pre-stimulated with interleukin-1β (IL-1β) or cotreated with IL-1β and resveratrol in the absence and presence of specific β1-integrin blocking antibody,collagen type Ⅱ,aggrecan,matrix metalloproteinase-13 (MMP-13) expression,and the translocation of nuclear factor kappaB (NF-κB) were analyzed by Western blotting.ANOVA was used for statistical analysis.Results Alginate bead culture plus conditional medium together could induce the cartilage-specific collagen type Ⅱ,aggrecan expression and extracellular matrix accumulation in differentiated chondrocytes.CGS supported differentiated cell attachment,proliferation,and migration.When those cells cultured on CGS were stimulated with IL-1β alone,collagen type Ⅱ and aggrecan expression was inhibited.However,MMP-13 expression increased.By Western blotting semi-quantitative analysis,the expression level of cartilage-specific collagen type Ⅱ of the control group was 0.484±0.006; the expression level of resveratrol intervention group was 0.474±0.014.The difference between these two groups was not statistically significant (P>0.05).The expression level of the IL-1β intervention group reduced to 0.155±0.009,which was statistically significant different from the above two groups(P<0.05).Resveratrol could antagonist the negative effect of IL-1β,and increase collagen type Ⅱ to 0.468±0.014,the difference between these two was statistically significant (P<0.05),and no significant difference when compared to the control group (P>0.05).Specific β1-integrin blocking antibody could abrogate these effects of resveratrol,decrease collagen Ⅱ expression to 0.169±0.011,the difference was significant (P<0.05),but there was no difference when compared to the IL-1β group (P>0.05).Aggrecan semi-quantitative expression has the same trend in the expression of type Ⅱ collagen while the expression of MMP-13,NF-κB had the reversal trend.These indicated that the resveratrol reversed the catabolic effects by reducing the nuclear translocation of NF-κB.Specific β1-integrin blocking antibody abrogated these effects of resveratrol.Conclusion Resveratrol,by regulating β1-integrin,acts as a NF-κB nuclear trans-location inhibitor to protect tissue-engineered cartilage.
5.The impact factors analysis in using extracorporeal shock wave lithotripsy for patients with ureteral calculi

Objective To summarize the impact factors of extracorporeal shock wave lithotripsy (ESWL) in ureteral calculi patients.Methods The data of 287 patients with ureteral calculi from January 2010 to December 2012 in the Aerospace General Hospital of Beijing who received the ESWL were retrospectively analyzed.The relationship between ESWL' s effect and patients' gender, age, the stone' s size, the stone's location, the length of the course and the stone bed' s polyps hyperplasia was analyzed.Results The Partial correlation course and chi-square test showed that, there was statistically significant difference between the ESWL treatment effect and patient' s age, patient' s length of the disease course and patient' s stone size (P<0.01).The value of OR between patient' s age and ESWL treatment effect was 1.905, showed that with the age of the patients increased every 10 years, the success rate of ESWL was reduced by 1.905 times.The value of OR between the patient' s length of the disease course and ESWL treatment effect was 2.809,indicated that with the duration of the course of prolonged every 1 month, the ESWL success rate decreased by 2.809 times.The value of OR between stone size and ESWL effect was 2.277, showed that the ratio between the stone diameter of 1.1-2.0 cm and 0.6-1.0 cm, the ESWL success rate reduced by 2.277 times.For the surgery patients who were failure to ESWL treatment,we found that there has statistical significance between the patients' length of disease course and stone bed' s polyps hyperplasia(P<0.01), but the relationship between the stone bed' s polyps hyperplasia and the gender, the age, the stone' s position, the stone ' s size had no statistical significance (P > 0.05) .Conclusion The patient ' s age, the length of the disease course, the stone ' s size and stone' s position are significantly affect to the effect of ESWL.There have closely relationship between the length of the disease course and the stone bed' s polyps hyperplasia, which is the reason why the longer length of the course, the worse the ESWL' s effect.This has very important instructed meaning for anticipating the treatment effect by the comprehensive evaluation of the patients before ESWL treatment.
6.Laparotomy and Cool-tip radiofrequency ablation for large liver tumors: short-term results

Objective To study the results of laparotomy and Cool-tip radiofrequency ablation to treat large liver tumors.Methods Laparotomy and Cool-tip radiofrequency ablation were carried out on 64 patients with large hepatic cancer.To destroy the tumor completely,for tumors of 3.0~4.0 cm in diameter,7 ablations were required; for 4.0~5.0 cm in diameter 15 ablations; for 5.0~6.0 cm in diameter 19 ablations; for 6.0~7.0 cm in diameter 40 ablations.Result The complete necrosis rate of laparotomy and radiofrequency ablation was 93.75% (60/64).The short-term results were good.Conclusions Laparotomy and Cool-tip multipoint overlapping radiofrequency ablation for large liver tumors (tumor diameter>3 cm) could result in a high complete necrosis rate and a low complication rate.It is a good radical treatment for unresectable and large liver cancer.
7.The development and application of stabilizers for ELISA test kit

Objective To develop ELISA kit stabilizer for each component ,with which the shelf life of kit could meet the com-mercial need .Methods A series of stabilizers for microplate ,enzyme-labeled antibody and standards were prepared by the means of experience and orthogonal approach ,through controlled trials ,screening out the best stabilizer formulations .Results A kind of coa-ted stabilizer (mass fraction :bovine serum albumin 0 .5% ,gelatin 0 .25% ,trehalose 5% ,PEG4000 0 .1% ) and a kind of enzyme la-beled antibody stabilizer (mass fraction :Bovine serum albumin 1% ,peptone 1% ,sucrose 10% ,trehalose 5% ,PEG4000 0 .25% ) were screened out .Conclusion The ELISA kits can be stable at 4 ℃ for 1 year after treated with the obtained stabilizers .
8.Analysis of Imaging Appearances of Chest in Patients with AIDS

Objective To analyse imaging appearances of chest in AIDS and to supply the evidence for imaging diagnosis this lesions. Methods 46 cases of AIDS who had chest X-ray films and CT were collected and their imaging appearances were reviewed. Results 20 cases showed bilateral lung diffuse lesions which were network shadow with mottle, punctate, patchy, nodular and opacification. 10 cases showed unilateral lung field lesions,of them,cavity in the left upper lung was presented in one patient. 5 cases showed thickness or turbulence of texture in bilateral lungs. 11 cases were negative on chest film.Conclusion The characteristics of X-ray film or CT of lung can reflect the chest pathological changes in patient with AIDS.
9.Mechanism of Coix Seed Extract in Improving Free Fatty Acid Metabolism in Rats with Fatty Liver

Objective:To observe the effect of coix seed extract on non-alcoholic fatty liver disease (NAFLD), and discuss the mechanism of coix seed extract in the treatment of NAFLD in the repect of free fatty acid. Methods:Totally 60 SD rats were randomly divided into 6 groups with 10 ones in each, the normal group, the model group, the positive group ( Xuezhikang capsules, 1 g · kg-1), three coix seed extract groups respectively at high (20 g·kg-1), medium (10 g·kg-1) and low (5 g·kg-1) dosage. The normal group was given basic diet, the other 5 groups were given high fat diet to establish NAFLD rat model, the modeling time was 8 weeks, and the drug treatment started from the 5th week till the 8th week. The liver weight and fat weight and index were observed after the drug intervention. The serum total cholesterol (TC), alanine aminotransferase (ALT), aspartate aminotransferase (AST), free fatty acid (FFA), fatty acid synthase (FAS), acetyl coenzyme A carboxylase (ACCa-se), C two acyl coenzyme A (MALONYL-CoA) , adenosine content of protein kinase ( AMPK) and adiponectin ( ADP) activated by glycosides were detected. Results: Com-pared with those in the normal group, the lipid metabolism related indices in the model group were significantly abnormal (P<0. 05), which suggested that the model was established successfully. After the drug intervention, the lipid metabolism related indicators in the positive group and the three drug groups were adjusted in varying degrees. Compared with those in the model group, TC, FFA and AMPK expression levels increased notably in the high dosage group and the positive group (P<0. 05), and the levels of ALT, AST, FAS, ACCa-se, MALONYL-CoA decreased at the same time (P<0. 05). Coix seed extract could decrease the body weight and liver wet weight in NAFLD rats and improve the related index significantly. Conclusion: The findings indicated that coix seed extract is highly effective in improving the pharmacological effect on NAFLD induced by high-fat diet, and the mechanism is achieved through ADP-AMPK-ACCase-malony-CoA-FFA axis.
10.Expression of serotonin transporters in postmortem brains of Parkinson's disease patients

Objective To study serotonin transporters(SERT) in postmortem human brains of Parkinson's disease(PD).Methods Immunoautoradiography was used to show SERT distribution in postmortem PD brains.ResultsIn comparison with healthy group,SERT decreased in dorsal raphe nucleus,substantia nigra and striatum in PD brains.Quantitative analysis showed that 25.9%、31.1%、27.2% and 24.7% of labeling intensity of SERT decreased in putamen,caudate nucleus,substantia nigra and ventrotegmental area as compared with corresponding control respectively.Among the four regions studied of dorsal raphe nucleus,SERT labeling intensity significantly decreased by 38.9%,37.3%,16.5% and 37.0% of corresponding control respectively in ventral part,dorsal part,caudal part and interfascicular part.Median raphe nucleus did not show the decreased SERT labeling.ConclusionDecreased SERT expression in three regions of postmortem PD brains indicates that a dysfunction of serotonergic raphe system may contribute to the etiology of Parkinson's disease.
