1.Effects of 60Co Irradiation with Different Doses on the Volatile Components in Huodan Pill

OBJECTIVE:To establish a method for the contents determination of volatile components in Huodan pill and study the effects of 60Co irradiation with different doses on the volatile components in Huodan pill. METHODS:GC-MS was used to de-termine the contents of patchouli alcohol and the ralative contents of β-patchouliene,α-guaiacene,seychellesene,α-patchouliene,α-bulnesene,5,11-diene guaiacyl,and unidentified objects in Huodan pill after 60Co irradiation with different doses. Column was HP-5MS by programmed temperature,volume temperature was 250℃,split ratio was 50:1,detector was triple quadrupole mass spectrometer detector,MS1 quadrupole temperature was 150℃,MS2 was 150℃,carrier gas was helium,flow rate of column was 1.2 ml/min,and the volume injection was 1 μl;ion source was EI,bombarding energy was 70 eV,temperature of ion source was 230℃,temperature of transmission line was 280℃ and scan range was 50-500 amu. RESULTS:The linear range of patchouli alcohol was 0.103 1-2.062 0μg(r=0.999 4);RSDs of precision,reproducibility and stability tests were lower than 1.0%;recovery was 98.05%-102.32%(RSD=1.8%,n=9). 60Co irradiation with different doses had little effects on the 8 volatile components in Huodan pill. CONCLUSIONS:60Co irradiation can be used for sterilization on Huodan pill,but irradiation dose should be controlled. The method is simple,good reproducibility,and can be used for the contents determination of volatile components in Huodan pill.
2.Determination of Residual Solvents in α-Ketophenylalanine Calcium by Gas Chromatography

To develop a method for the determination of residual solvents inα-ketophenylalanine calcium by capillary gas chromatography. Methods:The residual solvents were separated on a DB-624(30 m × 0. 32 mm, 0. 25 μm) capillary chromato-graphic column with temperature programming. The column temperature was maintained at 40℃ for 1 min,and then raised to 180℃at a rate of 10℃·min-1 and maintained for 2 min. N2 was used as the carrier gas, and FID was used as the detector with temperature of 250 ℃. The injector temperature was 200 ℃ and the split ratio was 10∶1, and direct injection was adopted. Methanol, ethanol, ethyl acetate and tetrahydrofuran in α-ketoleucine calcium were detected using an external standard method. Results:The four solvents were separated completely. There was a good linear relationship between the peak area and the concentration of each solvent ( r=0. 997 2-0. 999 5). The average recovery of the four solvents was 95. 47%-100. 26%(RSD≤4. 7%, n=9). Conclusion:The method is rap-id, simple, accurate and sensitive, and can be used in the determination of residual solvents in α-ketophenylalanine calcium.
3.Simultaneous Determination of 3 Isoflavones in Shenqi Granule by HPLC

OBJECTIVE:To establish a method for simultaneous determination of the confents of calycosin-7-glucoside,calyco-sin and formononetin in Shenqi granule. METHODS:HPLC was performed on the column of Welch Ultimate XB-C18 with mobile phase of acetonitrile-0.2% Formic acid solution (gradient elution) at flow rate of 1.0 ml/min,detection wavelength was 250 nm, column temperature was 35 ℃,and injection volume was 10 μl. RESULTS:The linear range of sample quantity was 0.008-0.32 μg for calycosin-7-glucoside(r=0.999 9),0.005-0.20 μg for calycosin(r=0.999 7)and 0.007-0.28 μg for formononetin(r=0.999 8);RSDs of precsion,stability and reproducibility tests were no more than 1.59%;recoveries were 98.17%-99.17%(RSD=0.67%, n=6),98.03%-99.66%(RSD=0.57%,n=6) and 98.16%-99.12%(RSD=0.43%,n=6). CONCLUSIONS:The method is sim-ple,accurate and reliable,and can be used for the content determination of Shenqi granule ofiso flavones.
4.Comparison of anxiety and depression state among patients with different type of vestibular peripheral vertigo.

OBJECTIVE:
To investigate and analyze the status of anxiety and depression among patients with four types of peripheral vertigo.
METHOD:
The clinical data of patients with one of the four types of peripheral vertigo, namely benign paroxysmal positional vertigo (BPPV), vestibular migraine (VM), Menière's disease (MD), and vestibular neuritis (VN), were collected. Thorough otological and neuro-otological examinations were performed on these patients, and their status of anxiety and depression were assessed using self-rating anxiety scale (SAS) and self-rating depression scale (SDS).
RESULT:
A total of 129 patients with one of the four types of peripheral vertigo(49 cases of BPPV, 37 cases of VM, 28 cases of MD and 15 cases of VN) were included in the study. The scores of SAS and SDS were higher in the patients with VM or MD than those with BPPV or VN (P < 0.05), and the incidence of anxiety (VM = 45.9% MD = 50.0%) and depression (VM = 27.0% MD = 28.6%) were higher in the patients with VM or MD than those with BPPV or VN (P < 0.05). Paired comparisons showed the differences between the incidences of BPPV and MV groups, BPPV and MD groups, and MD and VN groups were statistically significant (P < 0.05).
CONCLUSION
Among patients with different types of peripheral vertigo, anxiety/depression is more common in patients with VM or MD. This may be due to the different mechanisms involved in different types of vertigo, as well as differences in the prevention and self-control of the patients against the vertigo.

5.Clinical Observation of Mirtazapine Combined with Citalopram in the Treatment of Sleep Disorder of De-pressive Patients

OBJECTIVE:To investigate the clinical efficacy and safety of mirtazapine combined with citalopram in the treat-ment of sleep disorder in depressive patients. METHODS:One hundred and sixty-five depressive patients with sleep disorder were selected and divided into control group (82 cases) and treatment group (83 cases) according to random number table. Control group took Escitalopram oxalate tablet 10 mg,once every night,increasing to 20 mg according to disease condition;treatment group was additionally given Mirtazapine tablet 15 mg,once every night,increasing to 30 mg one week later. Both groups re-ceived treatment for consecutive 6 months. HAMD-17 and MADRS were observed in 2 groups before and after treatment,and the sleep quality of 2 groups were evaluated by PSQI before and after treatment;the sleep structure was measured by using polysom-nography before and after treatment;clinical efficacies and the occurrence of ADR were compared between 2 groups. RESULTS:Before treatment,there was no statistical significance in HAMD-17,MADRS and PSQI score,sleep structure between 2 groups (P>0.05);after treatment,above scores and indexes of 2 groups were all improved significantly,and the treatment group was sig-nificantly better than the control group,with statistical significance(P<0.05). Total response rate of treatment group was 97.47%, which was significantly higher than 78.95%of control group,with statistical significance(P<0.05). There was no statistical signif-icance in the incidence of ADR between 2 groups(P>0.05). CONCLUSIONS:Citalopram combined with mirtazapine shows sig-nificant therapeutic efficacy for sleep disorder of depressive patients,and can significantly improve sleep structure,adjust sleep cy-cle and improve sleep quality with good safety.
6. Mechanism of transcutaneous auricular vagus nerve stimulation on polycystic ovary syndrome

Non-invasive transcutaneous auricular vagus nerve stimulation (ta-VNS) is established on the basis of auricular acupuncture and the principle of vagus nerve stimulation (VNS). The results of the comparative study of 4 indications of VNS show that ta-VNS achieves almost the equivalent effect as VNS. Moreover, the ta-VNS is advantageous at safety, economy and portability. Regarding the clinical prevention and treatment of polycystic ovary syndrome (PCOS), it is still controversial for the effect of acupuncture and moxibustion therapy including auricular acupuncture. Moreover, it is the difficulty for the principle of acupoint selection and the treatment mechanism of this therapy to be in line with modern medicine. As neuroendocrine disease, PCOS and the ovary, its target organ are all associated with the vagus nerve. It is a new idea to introduce the ta-VNS in treatment of PCOS by regulating the reproductive and endocrinal disturbance of PCOS in terms of vagus nerve and improving the acupuncture and moxibustion therapy, such as the auricular acupuncture. In the paper, the potential mechanism of the ta-VNS in treatment of PCOS is presented: 1)the ta-VNS treats PCOS by improving insulin resistance; 2)the ta-VNS relieves PCOS-induced psychological disorders through the treatment of depression; 3)the ta-VNS is applicable in treatment of PCOS complications, such as hypertension and diabetes.
7.Indirect Enzyme-Linked Immunosorbent Assay Based on the Nucleocapsid Protein of SARS-Like Coronaviruses

The nucleocapsid protein (N) is a major structural protein of coronaviruses. The N protein of bat SARS-like coronavirus (SL-CoV) has a high similarity with that of SARS-CoV. In this study, the SL-CoV N protein was expressed in Escherichia coli, purified and used as antigen. An Indirect Enzyme-Linked Immunosorbent Assay (indirect ELISA) was developed for detection of SARS- or SL-CoV infections in bat populations. The detection of 573 bat sera with this indirect ELISA demonstrated that SL-CoVs consistently circulate in Rhinilophus species, further supporting the proposal that bats are natural reservoirs of SL-CoVs. This method uses 1-2 μl of serum sample and can be used for preliminary screening of infections by SARS- or SL-CoV with a small amount of serum sample.
8.Value of SPECT/CT imaging for follow-up of bone metastases

Objective To retrospectively evaluate the value of SPECT/CT imaging for follow-up of bone metastases.Methods A total of 178 patients with bone metastases (387 lesions) underwent 2 or more events of whole-body bone scintigraphy (WBS) and SPECT/CT imaging.Sequential images were analyzed by 2 experienced,nuclear medicine physicians.Each lesion was interpreted as progressive,remissive or stable in WBS or SPECT/CT independently.Reasons for the discordance between WBS and SPECT/CT results were analyzed.The results of clinical follow-up,including clinical symptoms,tumor markers,serum ALP,radiograph,CT and MRI,were likewise classified as progressive,remissive or stable.The x2 test was used to compare the differences between the two imaging methods.Results The follow-up results of the two imaging methods were consistent in 313 (80.88%,313/387) lesions,including 208 in progression and 105 in remission or stable condition.Among the 74(19.12%,74/387) lesions showing discordance,48 showed remission or stable conditions on WBS but progression on SPECT/CT (64.86%,48/74) ; while 26 showed progression on WBS but remission or stable condition on SPECT/CT (35.14%,26/74).There was a statistically significant difference of the follow-up results between WBS and SPECT/CT (x2 =6.54,P <0.05).Conclusion SPECT/CT is more valuable than WBS for follow-up of bone metastases.
9.MR diffusion tensor imaging of optic radiation in chronic Leber's hereditary optic neuropathy

Objective To observe quantitatively the optic radiation alterations in chronic Leber's hereditary optic neuropathy (LHON) using MR diffusion tensor imaging at 3.0 T.Methods Twenty-five patients with chronic LHON and twenty-eight sex-and age-matched disease-free control subjects wereenrolled from March 2011 to December 2014.The whole brain DTI examination and retinal opticalcoherence tomography were performed in all the subjects.The parameters values of optic radiation includingfractional anisotropy(FA),mean diffusivity(MD),principal eigenvalue(λ//),orthogonal eigenvalue(λ⊥) and average retinal nerve fiber layer (RNFL) thickness and temporal retinal nerve fiber layer(RNFL) thickness were measured.For DTI and OCT parameters,differences between patients and controls and differences right and left of the patient group were evaluated using independent-samples t test and paired t test respectively.Results In LHON patients,the DEC maps showed that the bilateral optic radiation fibers were obviously sparse and slender.The values of FA,MD,λ// and λ⊥ in the right optic radiation were 0.523±0.050,(0.903±0.061) × 10-9 mm2/s,(4.999 ±-0.097) × 10-9 mm2/s and (4.126-± 0.065) × 10-9 mm2/s respectively,and the corresponding values of left optic radiation were 0.537±0.041,(1.057±0.573)× 10-9 mm2/s,(5.052±0.119)×10-9 mm2/s and (4.138±0.072)×1009 mm2/s.Compared with optic radiation in the control group,the bilateral FA was significant reduced,the bilateral λ⊥ value and right MD value were significant increased (t=-6.524,-6.045,6.932,6.454,3.471,P<0.05).However,there were no significant difference of the bilateral λ〃 and left MD (t=-0.457,1.221,1.833,P>0.05).There were no significant differences in FA,MD,λ//and λ⊥ between the left and right eyes in LHON patients(P>0.05).The values of average RNFL thickness and temporal RNFL thickness in right eyes were (59.36± 10.94) and (41.72±9.43) μm respectively,and the corresponding values of left eyes were (60.12±9.81),(44.72±9.43) μm.The average and temporal RNFL thickness were significantly thinned in LHON patients compared with the control group(t=-16.357,-10.398,-17.169,-9.672,P<0.05).There was no significant difference in average RNFL thickness between the left and right eyes (t=0.959,P>0.05),temporal RNFL thickness between the left and right eyes was statistically significant (t=2.645,P<0.05).Conclusion The bilateral optic radiation had significant degeneration and atrophy in chronic LHON.
10.Effects of Nrf2-ARE signal path on levrtiracetam anti-epileptic andlevrtiracetam on learning and memorizing ability

Aim To explore the effects of Nrf2-ARE signal path on levrtiracetam anti-epileptic and levrtiracetam on learning and memorizing ability.Methods Thirty-six SD rats were divided into normal saline group, levrtiracetam group, model group and treatment group.Each group recruited nine rats.Tests of Morries water maze were given to the rats to evaluate their learning and memorizing ability.The protein expression of nuclear factor (erythroid-derived2)-like2 (Nrf2), heme oxygenase 1(HO-1) and NAD(P)H quinone oxidoreductase(NQO1) were examined by Western blot.Results Compared with model group, levrtiracetam could shorten the plateau period in epileptic rats (P<0.05), and increase the expression of Nrf2 protein, HO-1 protein and NQO1 protein in hippocampus(P<0.05).Conclusions Levrtiracetam could improve the learning and memorizing ability in epileptic rats.Levrtiracetam may increase the expression of HO-1 protein and NQO1 protein through the Nrf2-ARE pathway and play a part in antiepileptic effects.
