1.Expression and purification of the fusion hepatitis C virus hypervariable region 1 protein and it's preliminary application

Objective To clone and express hypervariable region 1 (HVR1) fragments of different genotypes of HCV strains from different cities of China.Then detect the reactivity of the expressed HVR1 fusion proteins with sera of chronic he-patitis C patients to analyzing it's signification in clinical utilization. Methods HVR1 genes of four HCV strains (genotype 1b and 2a) were amplified from pGEMT-E2 plasmids and cloned into pQE40 vectors, respectively to construct four recombinant prokaryotic expression plasmids which expressed HVR1 as fusion proteins with DHFR in Escherichia coli strain TG1. Then we used the purified DHFR-HVR1 proteins to detect the anti-HVR1 antibodies in 70 serum samples of chronic hepatitis C patients. Results Four DHFR- HVR1 fusion proteins were successfully expressed in E.coli (320~ 800 ?g fusion proteins per 100 ml culture). Each fusion protein (SH1b, BJ1b, SD1b and SD2a) reacted with 72.8%(51/70), 60%(42/70), 48.6%(34/70), and 58.6%(41/30) of the anti-HCV positive patients' sera, respectively by ELISA. 57% (4/7) of the non responders' sera taken before therapy reacted with all four HVR1 fusion proteins, while only 15.3% (2/13) of the sera from responders reacted with all of them. 2.Correlation between perceived partner responsiveness and fear of disease progression in young patients with cervical cancer after surgery

Objective:To explore the status quo of young patients′ perception of partner responsiveness and fear of disease progression after surgery of cervical cancer, and to analyze the correlation between the two to provide a basis for targeted intervention.Methods:From October 2018 to June 2020, a convenient sampling method was used to select 121 young patients after radical resection of cervical cancer who were treated in East Branch of Qingdao Municipal Hospital. It was investigated by the Perceived Partner Responsiveness Scale and the Fear of Progression Questionnaire-Short Form in cancer patients.Results:The total score of perceived partner responsiveness of young patients with cervical cancer after surgery was (41.30 ± 6.14) points, and the total score of fear of disease progression was (31.10 ± 6.58) points; perceived partner responsiveness was negatively correlated with fear of disease progression ( r=-0.72, P<0.05). Multiple linear regression analysis showed that work status and perceived partner responsiveness were the influencing factors of patients' fear of disease progression ( t=11.54, 18.40, all P<0.05). Conclusions:The perception of partner responsiveness in young patients with cervical cancer after surgery is closely related to the fear of disease progression. Perception of partner responsiveness can affect patients′ fear of disease progression; it can improve the patients′ perception of partner responsiveness to improve their fear of disease progression.
3.Expression of hsa-miR-422a in hypertrophic scars: a bioinformatics analysis

Objective:To evaluate the expression level of hsa-miR-422a in hypertrophic scars and to identify the target genes of hsa-miR-422a along with their biological functions using bioinformatics approaches.Methods:From June 2020 to December 2020, tissue samples of 3 hypertrophic scar and 3 normal skin were collected from patients (3 males, 3 females, aged 20-42 years) in Department of Plastic and Reconstructive Surgery, Shanghai Ninth People′s Hospital, Shanghai Jiaotong University School of Medicine. Primary fibroblasts were isolated and cultured. Real-time quantitative PCR was performed to quantify the expression of hsa-miR-422a. To construct a ceRNA network, starbase and Target Scandata bases were utilized to predict genes as well as long noncoding RNAs (lncRNAs) that may sponge hsa-miR-422a. GO and KEGG pathway enrichment analyses were conducted on the target genes of hsa-miR-422a; protein-protein interaction (PPI) networks were constructed to identify the hub genes whose functions were predicted by functional enrichment analyses. The expression of hub genes was validated through real-time quantitative PCR in hypertrophic scars.Results:The expression of hsa-miR-422a was significantly lower in the hypertrophic scar tissue samples and fibroblasts compared to that in the normal skin ( P<0.05). 133 target genes as well as 1033 lncRNAs were predicted by starBase and TargetScandata bases and used to construct an hsa-miR-422a-centered ceRNA network. PPI networks of the target genes revealed 10 hub genes, including MAPK1, GRB2, and IGF1R, which were discovered to be related to protein serine/threonine/tyrosine kinase activity, ubiquitin protein ligase binding, fibroblast growth factor receptor signaling pathway, muscle cell proliferation, and many others; besides, they may be involved in FoxO, mTOR, Toll-like receptor, Ras, MAPK, PI3K-Akt signaling pathways and signaling pathways regulating pluripotency of stem cells. Three hub genes (MAPK1, GRB2, and IGF1R) were significantly upregulated in hypertrophic scars ( P<0.05). Conclusions:hsa-miR-422a is significantly downregulated in the hypertrophic scars and may target hub genes such as MAPK1 in ceRNA networks, ultimately modulating hypertrophic scar formation.
