1.595nm tunable pulsed dye laser in treatment of children heamangioma of skin

Objective To evaluate of the clinical efficacy and side-effect of 595nm tunable pulsed dye laser in treatment of children heamangioma of skin. Methods A retrospective study was conducted according to 76 children patients with heamangioma of skin (including 53 cases of nevus flammenus, 20 strawberry hemangioma and 3 cavernous hemangioma) treated with 595nm tunable pulsed dye laser. We evaluated the efficacy and side-effect of the treatment. Results The effectiveness rates were 83.2% and 86.63%, respectively, in nevus flammenus and strawberry hemangioma, but there were disappointed efficacy obtained in patients with cavernous hemangioma treated with 595nm tunable pulsed dye laser. There was 7.9% of the side-effect rate in the children with heamangioma of skin treated by 595 nm tunable pulsed dye laser. Conclusion There is satisfactory clinical efficacy and lower side-effect for children hemangioma treated by 595nm tunable pulsed dye laser.
2.Construction and Application of the Nursing Quality Control Platform Based on Informatization in Tertiary Hospitals of Qing-hai Province

Purpose/Significance To establish an informatization platform for nursing quality control in tertiary hospitals of Qinghai province,and to analyze its influence on nursing quality.Method/Process According to the needs of nursing quality management in Qing-hai province,based on the nursing quality data platform,a nursing quality index monitoring module is established.A tertiary hospital is selected as a pilot hospital,and the nursing quality data of this hospital from January to June 2021 is selected as the control group by u-sing the historical comparative study method,and the nursing quality data collected by the informatization platform from January to June 2022 is selected as the observation group.The changes of nursing quality before and after the application of informatization platform are compared.Result/Conclusion After the application of the informatization platform for nursing quality control,the rate of physical restraint and unplanned extubation of endotracheal intubation of hospitalized patients decreases significantly,and the score of patients'satisfaction with nursing work increases significantly.The informatization platform has strong operability and clinical application value.
3.Construction of HNP-3 Mature Peptide Bait Plasmid of Yeast Two-hybrid System and Detecting Its Self-activating and Toxic Effect

Objective To construct the bait plasmid of HNP-3 mature peptide in yeast two-hybrid system and examine whether the recombinant bait plasmid has self-activating and toxicity effect.Methods Using RT-PCR technique,the cDNA fragments of HNP-3 mature peptide gene were amplified from the extracted RNA in cultured HL-60 cells.The fragment was firstly cloned into pBluescript-SK-II vector,confirmed by sequencing,then sub-cloned into the bait plasmid pGBKT7 and identified with PCR and sequence analysis techniques.The recombinant plasmid was introduced into the yeast cell AH109,and its self-activating and toxicity effect was tested by auxotrophic selective culture.Results DNA sequencing indicated that the inserted fragment in pBluescript-SK-II vector was HNP-3 mature peptide gene sequence,and the sub-cloned recombinant pGBKT7-HNP-3 was no mismatch.The recombinant bait plasmid didn't have self-activating effect and did not show toxicity to yeast AH109 cell.Conclusion The bait plasmid of HNP-3 mature peptide was constructed successfully.This was helpful for investigating the proteins interacting with HNP-3 mature peptide by yeast two-hybrid technique.
4.Optimization of extraction technology for total saponins from Rhizoma Panacis Majoris using orthogonal test

Objective To optimize extraction technology for total saponins from Rhizoma Panacis Majoris ( zhuzishen) and provide a theoretical basis for development and utilization of saponins from Rhizoma Panacis Majoris. Methods Ultraviolet spectrophotometry was employed to detect content of total saponins from Rhizoma Panacis Majoris with detection wave length set at 540 nm. And effects of ethanol concentration, solvent quantity, extraction time and extraction times on extraction process were investiga-ted by using orthogonal test design. Results Linear range of total saponins from Rhizoma Panacis Majo-ris was 0. 052~0. 182 mg, and the average recovery rate was 98. 64%. The best extraction technology for total saponins from Rhizoma Panacis Majoris was to extract 3 times with 10 times treatment of 70%ethanol for 1 h per time. The average yield rate of total saponins from Rhizoma Panacis Majoris was 12. 32%. Conclusion This established ultraviolet spectrophotometry was accurate and reliable to detect contents of total saponins from Rhizoma Panacis Majoris. At the same time, this optimized extraction technology of total saponins from Rhizoma Panacis Majoris was simple, stable and feasible.
5.Multi-index comprehensive evaluation and comparison of different soften-ing methods for cortex cinnamomi

Objective To compare the effects of three different softening methods ( soaking, sprinkle-moistening and decompression-quenching methods) on cortex cinnamomi, contents of water extraction, volatile oil content and cinnamaldehyde and cinnamic acid were analyzed to determine the best softening method of cinnamon. Methods Water extraction and volatile oil contents of each sample were deter-mined according to China Pharmacopoeia criteria. And the high performance liquid chromatography ( HPLC) method was used for determining the content of cinnamic acid and cinnamicaldehyde. Multi-in-dex comprehensive evaluation method was used to analyze the results. Results The order of comprehen-sive evaluation scores was decompression-quenched > soaked > the original medicine> sprinkle-moin-stening. Conclusion Cortex cinnamomi processed by the softening method of decompression-quenching seems to have the characteristic of the best luster and richest fragrance; furthermore, this method seems to take the least time and damage to the fewest effective components of the medicine, proving itself to be the best softening process.
6.Characteristic of intestinal absorption of euphorbia factor L1 by rats single pass intestinal perfusion moder in situ

Objective To study the characteristic of absorption of euphorbia factor L1 in intestine of rats, and to observe the effects of P-glycoprotein(P-gp)and multidrug resistance-associated protein(MRP2)on intestinal absorption of euphorbia factor L1 .Methods The contents of euphorbia factor L1 of intestinal perfusion fluid of duodenum, jejunum, ileum and colon in the rats in situ single-pass intestinal perfusion model were determined by using HPLC.The drug absorption rate constant( Ka ) and the apparent absorp-tion coefficient( Papp ) in four intestinal regions were calculated.Results Ka and Papp of euphorbia factor L1 at colon were the highest of the whole rat intestine.Significant increase of Ka and Papp showed at rat colon when co-perfused with verapamil hydrochloride, by contrast, decrease of Ka and Papp found when co-perfused with indomethacin.Conclusion We inferred that verapamil hydrochloride be the substrate of P-gp and that indomethacin be not the substrate of MRP2 .
