AIM: To investigate how brain-dead state affects the heart structure and function and the effect of PKC-? in BA-Ma mini pigs.METHODS: Ten Ba-Ma mini pigs were randomized into 2 groups: brain-dead group(n=5),and control group(n=5).The brain-dead model was made by increasing intracranial pressure,while the control group was maintained anesthesia for 24 h.The concentrations of cTnT,TNF-?,IL-1? and IL-6 in serum were determined at 6,12 and 24 h after brain death.At 24 h,heart tissues were observed by HE staining and electron microscope.The expression of PKC-? was detected by immunohistochemistry and RT-PCR.RESULTS:(1) Histological changes of myocardium: flaky bleeding under endocardium and dissolution of myocardium were found in optical microscope.In electron microscope dropsical mitochondria and confluent muscle fiber were found.(2) Changes of serum cTnT: serum cTnT for brain-dead group began to increase gradually since 6 h,and were significantly higher at each time point than those in control group(P
2.Killing effects in vitro of a new photosensitizer black titanium dioxide-mediated photodynamics on prostate cancer cells

Objective:To investigate the killing effects in vitro of a new photosensitizer black titanium dioxide b-TP-700-mediated photodynamics on prostate cancer cell line PC-3 cells.Methods:The b-TP-700 was successfully prepared by using solid phase in situ thermal reduction method. PC-3 cells were treated with 31.25, 62.5, 125, 250, 500, 1 000 μg/ml b-TP-700 for 12, 24, 48 h. The cell activity was detected by using CCK-8 method, and the morphology of PC-3 cells was observed by using confocal fluorescence microscope, the toxicity of b-TP-700 to PC-3 cells was also judged. PC-3 cells were treated with different concentrations of b-TP-700 (31.25, 62.5, 125, 250, 500 μg/ml) and irradiated with laser (1 W/cm, 808 nm) for 1, 3, 5, 7 min. The cell activity was detected by using CCK-8 method. The production of reactive oxygen species in PC-3 cells cultured in 250 μg/ml b-TP-700 after laser irradiation for 5 min was detected by using confocal fluorescence microscopy. The non-laser irradiation cells were selected as the control group.Results:There were no statistically significant differences in cell activity of PC-3 cells treated with different concentrations of b-TP-700 for the same time ( F value was 1.26, 0.39 and 0.37, respectively; all P > 0.05). PC-3 cells were treated with 1 000 μg/ml b-TP-700 for 24 h, no obvious morphological changes and no obvious dead cells were observed under fluorescence microscope. When PC-3 cells were treated with the same concentration of b-TP-700, the activity of PC-3 cells was decreased with the extension of laser excitation time. Under the same laser excitation time, the activity of PC-3 cells was decreased with the increase of b-TP-700 concentration (all P < 0.05). Under laser irradiation, obvious green fluorescence could be detected in the PC-3 cells treated with 250 μg/ml b-TP-700, while almost no green fluorescence could be observed in the control group. Conclusions:A new photosensitizer b-TP-700 has the good reactive oxygen generation ability. b-TP-700-mediated photodynamics show obvious killing effects on prostate cancer PC-3 cells in vitro, which is expected to be a new method for the treatment of prostate cancer.
3.Infection and inflammatory response in elderly patients with oral and maxillofacial surgery

Objective To investigate the characteristics and influencing factors of infection in elderly patients with oral and maxillofacial surgery,and to explore the effect of infection on inflammatory response-related proteins,in order to guide the clinical treatment.Methods A total of 653 cases of patients aged more than 60 who were treated with oral and maxillofacial surgery during January 2004 to December 2015 in Shenzhen People's hospital was involved and divided into infected group (n =48) and uninfected group (n =605) according to whether postoperative nosocomial infection was occurred.The characteristics and influencing factors of infection were observed.A total of 171 cases of healthy volunteers whose age and sex were matched with patients were collected as control group.The expressions of inflammatory responserelated proteins in 3 groups were compared.Results The main site of postoperative nosocomial infection was lower respiratory tract,with 15 cases of patients,accounting for 31.3%.A total of 48 strains of pathogens was isolated,the main pathogenic bacteria were Streptococcus pyogenes,with 8 strains,accounting for 16.7%.Streptococcus pyogenes had high drug resistance to erythromycin and clindamycin,and had high sensitivity to penicillin G and meropenem.Aged more than 70,operation time more than 3 h,the primary diseases as malignant tumor were the independent risk factors for postoperative nosocomial infection (P <0.05).Blood interleukin (IL)-1,IL-2,IL-6,IL-10,tumor necrosis factor α (TNF-α),transforming growth factor β (TGF-β) and interferon-γ (IFN-γ) in infected group were more than uninfected group and control group.The differences were statistically significant (P < 0.05).Conclusions The rate of infection in elderly patients with oral and maxillofacial surgery is still high in our hospital.The main site of infection is lower respiratory tract and the main pathogens were Streptococcus pyogenes.Aged more than 70,operation time more than 3 h,the primary diseases as malignant tumor were the independent risk factors for postoperative nosocomial infection.Infection can lead to peripheral inflammatory factor levels increase significantly.
