1.Detection of the immunological competence of HBsAg pulsed Dendritic Vaccination

Objective:To explore the immunological competence of HBsAg-DCs vaccination prepared via HBsAg inducing DCs. Methods:The bone marrow cells of Lewis rat were cultured by RPMI1640 added with 4 ng/ml of granulocyte macrophage colony stimulating factor(GM-CSF)and 8 ng/ml of interlcukin-4(IL-4). 7 days late,the RPMI1640 were added with HBsAg(100 ?g/ml)for another 48h to obtain HBsAg-DCs. The morphologic changes of HBsAg-DCs were observed by light microscope and the phaenotype change were detected by flow cytometry (FCM);The immuno-competence of the HBsAg-DCs were judge by one-way mixed lymphocyte reaction (MLR). Results:There were many DCs growing adherence in cluster and the suspended cells were obviously increasing and much bigger after administrating HBsAg. 2.Inhibitory effects of TIMP-3 on endothelial cell proliferation

Objective: To investigate the inhibitory effect of TIMP-3 on endothelial cell(EC) proliferation.Methods: The inhibition of EC proliferation was analyzed by light microscopy,MTT,-TdR up-take,and flow cytometry et al.Results: A concentration-dependent and time-dependent proliferation inhibition and survival were demonstrated in the HUVEC incubated for 1~7days in the presence of TIMP-3.The proliferation inhibition was shown as the lessened cell growth,decreased counting of mitotic cells.TIMP-3 could also aggravate the increase of apoptotic cells and cell ratio in G2/M phase.Conclusion: TIMP-3 has significant inhibitory influence on EC proliferation,migration and can promote apoptosis of cells.

Hepatocellular carcinoma(HCC) is one of the most common malignant tumors in the world.According to the statistic data of Ministry of Public Health of China in 1995, the annual mortality of HCC was 20.4 per 0.1 million, and had the rise tendency. Surgery is commonly the main modality of HCC combined therapy. The high intensity focused ultrasound (HIFU), as a noninvasive therapy, with the mechanism of transient high temperatures, cavitation and mechanical effects, has been recognized. This review aimes to elucidate the immune changes in HCC after HIFU exposures and the mechanism.
5.The curative effect and influence of leibeirazole quadruple therapy on the treatment and expressions of IL-18 and IL-35 in patients with recurrent Hp positive gastric ulcer

Objective To investigate the effect and changes of IL -18 and IL -35 in patients with recurrent Hp positive gastric ulcer by treating with leibeirazole quadruple therapy.Methods 180 patients with recurrent Hp positive gastric ulcer were divided into two groups.The control group(90 cases)was applied amoxicillin,clarithromy-cin and colloidal bismuth pectin.The observation group was added leibeirazole.The effect and changes of IL -18 and IL -35 were observed before and after treatment in the two groups.Results The effective rate (86.67% vs. 73.33%,χ2 =5.000,P =0.025),inflammatory reaction rate(26.67% vs.51.11%,χ2 =11.314,P =0.008)and Hp positive rate(16.67 % vs.31.11%,χ2 =5.163,P =0.023)were higher in the observation group than in the control group.Expression of IL -18 was decreased after treatment[(70.80 ±11.21)ng/L vs.(88.83 ±7.13)ng/L, t =8.27,P =0.0210]in the observation group,but no change in the control group[(85.62 ±10.84)ng/L vs. (87.64 ±9.28)ng/L,t =0.78,P =0.4512).Expression of IL -35 didn't change after treatment in the two group (P >0.05).Conclusion Leibeirazole quadruple can increase the clinical effect,decrease the expression of IL -18 in patients with recurrent Hp positive gastric ulcer,it is worthy to be applied in clinical treatment.
6.Combination of transcatheter arterial chemoembiolization and antitumor drugs in the treatment of Hepatocellular carcinoma

Hepatocellular carcinoma(HCC)is a common world wide disease.Transcatheter arterial chemoembolization(TACE)has been the first choice for the unresectable midadvanced ones.However, TACE has its own limitation such as high recurrence rate and many complications, what's more, recurrence induced by neovascularization which is stimulated by activation of vascular endothelial growth factors promoted by ischemia and hypoxia in local tissues after the embolization always reduce the effect of treatment.And combination of TACE and anti-angiogenesis therapy is capable to make up the shortage of each other and reduce the recurrence rate, which has been a hot issue for non-surgical treatment of HCC.
7.Current progress in bystander effect of suicide gene therapy

Nowadays, tumor is one of the most challenging human health diseases, which oblige human tirelessly to studied for the tumor.Gene therapy is a promising treatment in oncotherapies. Suicide gene therapy is the most widely researched among gene therapies. At the same time, bystander effect take important role in the mechanism of suicide gene therapy. Therefore, more researchers devote themselves to studyinghow enhance the bystander effect in order to improve the effect of suicide gene therapy. This article reviewed in short how to augment bystander effect of suicide gene therapy against cancer.
8.Progress in the materials, design and related technique clinical application of temporomandibular joint prosthesis

OBJECTIVE:To summarize the materials, design, fit-shape and fixation techniques and clinical application of temporomandibular joint (TMJ) prosthesis.DATA SOURCES: An online search was undertaken to identify English articles about TMJ prosthesis published in Pubmed database from January 1998 to December 2006 using the key words of "temporomandibular joint, joint prosthesis".STUDY SELECTION: The collected literatures about TMJ prosthesis were sorted, and those with good pertinence were selected. For the literatures of the same field, those published recently or in authorized journals were selected.DATA EXTRACTION: Totally 69 literatures were collected, 28 were enrolled, and the other 41 were excluded, including 23 were irrelative with the aim of this study, and 18 were repetitive studies.DATA SYNTHESIS: Joint prosthesis is one of the fields that develop the fastest in orthopedic surgery in recent 30 years.TMJ replacement aims to enhance the function of TMJ, alleviate pain, and prevent serious complication. With the rapid developments of material science, tissue engineering, joint biomechanics and other related subjects, TMJ prosthesis has been significantly improved in the materials, design, fit-shape and fixation techniques. People have developed from simply imitating the outline form and mechanical motor of TMJ to pay more attention to its pathological function. It is very significant to trace the new progress in hip prosthesis and knee joint, and apply the good outcomes in the design and manufacture of TMJ prosthesis.CONCLUSION: Prosthetic material plays leading and promoting roles in the development of joint prosthesis, good design,fit-shape and fixation are the necessary conditions for prosthesis to act its role, and it is also necessary to investigate joint biomechanics. TMJ prosthesis should be further investigated, and it has good prospect.
9.Expressions of TLR4 and Dectin-1 in cornea tissue of rats with Aspergillus fungal keratitis and their effects in pathogenesis of keratitis

Objective To explore the expressions of Dectin-1 and Toll-like receptor 4 (TLR4)and the secretory charateristics of inflammatory factors of the Aspergillus fumigatus infected cornea of the rats,and to clarify theirs role in the pathogenesis of rat keratitis.Methods 40 SD rats were separated into control group (n = 10)and aspergillus keratitis group (n=30).The rats in aspergillus keratitis group were infected by Aspergillus fumigatus in two eyes for 12,24 and 48 h,and the cornea tissue was took and ELISA kit was used to detect the level of inflammatory cytokines IL-1,TNF-a,IL- 6,IL-10,and NF-κB;immunohistochemistry,Western blotting and RT-PCR methods were used to determine the Dectin-1 and TLR4 protein and mRNA expressions.Results The rat models were successfully established.The eyes of the rats in control group were limpid tested by corneal slit lamp, however,there existed a clear boundary film on the surface of eyes of the rats in aspergillus keratitis group infected for 12 h,white infiltrates and bloodshot in the eyes in 24 h group and the white infiltrates were further strengthened and almost covered the entire eyes in 48 h group. The order of inflammatory cytokines IL-1,TNF-α,IL-6,IL-10, NF-κB was:48 h group> 24 h group> 12 h group> control group (P < 0.05),so did the expression levels of Dectin-1 and TLR4 protein and mRNA in various groups tested by Western blotting and RT-PCR methods.The expression levels of Dectin-1 and TLR4 were increased with the prolongation of time investigated by immunohistochemical staining.Conclusion Dectin-1 and TLR4 are secreted in excess after the eyes are infected with fumigatus,which promotes the expressions of related inflammatory cytokines and results in the occurrence of keratitis.
10.TIL phenotype alteration in oral cancer tissue follwing induction chemotherapy via subcutaneous implantable drug pump

objective: To study the effects of induction chemotherapy via subcutaneous implantable drug pump on local immune status. Methods: 47 cases with node negative oral cancer were treated with MTX, CDDP, PYM via subcutaneous implantable drug pump(in 27 cases), or via vein for case control trial(in 20 cases). Local immune status of tumor tissue was detected by immunohistochemical technique before and after treatment. Results: The effectiveness of treatment with the drugs via subcutaneous implantable drug pump (DSIP) was observed in 21 out of 27 cases (77.8%) and that with the drugs via vein(DV) was in 13 out of 20(65.0%) respectively( P 0.05), while the number of positive cells of CD 4 and CD 4/CD 8 in the cases treated with DV was decreased( P
