4.Inhibitory effects of docosahexaenoic acid on the growth of incubated human pancreatic cancer cell lines

Objective To investigate the effects of docosahexaenoic acid (DHA) on the growth of human pancreatic cancer cell lines. Methods Human pancreatic cancer cell lines Patu8988 and SW1990 were treated with DHA. The cell proliferation was evaluated by MTT assay. Cell cycle, apoptosis, and cyclooxygenase-2 ex-pression were evaluated by flow cytometry. Results After incubation of pancreatic cancer cell with DHA for 24 to 72 hours, cell proliferation significantly decreased (P<0. 01) and apoptosis increased, both of which were time- dependent and dose-dependent After incubation with 50μg/ml DHA for 24 hours, cyclooxygenase-2 expression of pancreatic cancer cell lines significantly decreased (P < 0. 05). Conclusions DHA can inhibit the growth of hu- man pancreatic cancer cells via decreasing cell proliferation and inducing apoptosis. These effects may be associat- ed with the decrease of cyclooxygenase-2 expression.
5.Rural migrant workers' coping style,traditional personality and poverty attribution style

ObjectiveTo explore the relative model for psychological resources and poverty attributions of migrant workers in adversities in the aim of understanding poverty problems in China Mainland and find out the factors for tolerance in systematic unjustness.MethodsChina Mainland Poverty Attribution Questionnaire (CMPAQ) ,Simple Coping Style Questionnaire(SCSQ),Resilience Scale (CD-RISC),Locus of Control subscale (I_E) and Thrift subscale (T_E) of CPAI-2 were used upon 398 rural migrant workers,in which 301 were valid.Results ①Deseription statistic: compared with national model,migrant workers score higher in positive coping style subsonic(1.92±0.45),and lower in negative coping style subscale (1.46±0.66),lower in CD-RISC (60.98±14.26) ,higher in I_E subscale (5.53±2.17) and lower in T_E subscale 7.48 + 1.77) ; Scores on the subscales of CMPAQ were separately overburden-fate (3.25±0.90),institution-unjustness (3.45±1.03),personal-competence(3.72±0.78),personal-quality (3.03±1.04),and geographical-environment (3.07±0.97).②Regress analysis:positive coping style can clearly positively predict personal-competence and personalquality attribution,while negative coping style can clearly positively predict all other factors.Resilience(CD-RISC)can clearly positively predict personal-competence attribution.The standard regression coefficient (β) of inner locus of control (I_E) to overburden-fate was negatively significant,while that of thrift (T_E) to all the poverty attribution factors was not significant.All variables other than negative coping style couldn' t clearly predict institution-unjustness attribution.ConclusionThe study discovered some positive psychological resources in rural migrant workers.These resources help them attribute poverty to personal competence but stand in tolerance for institutional unjustness.

7.Study on the effect of perindopril and benazepril on nude mice bearing esophageal squamous cell carcinoma

ObjectiveTo investigate whether there was inhibitory effect of angiotensin-Ⅰ converting enzyme inhibitors (ACEI) on esophageal carcinoma cell line.Methods The highest expression of vascular endothelial growth factor (VEGF) at mRNA level was screened in esophageal squamous cell carcinoma cell lines KYSE30,TE-1,EC109 and EC9706 by reverse transcriptionpolymerase chain reaction (RT-PCR) and which was transplanted and established nude mice xenografts model.After tumor formed,the nude mice were evenly divided into control group,perindopril group and benazepril group (n=6) of which 0.9% saline,4 mg/kg perindopril and 6 mg/kg benazepril was given respectively to nude mice in each group.The tumor size and the tumor inhibitory rate were measured.The expression of CD31 in xenograft tumor was tested by immunohistochemistry and the microvessel density (MVD) was assessed.ResultsThe highest expression of VEGF at mRNA level was in EC9706 cell line.At second and third week,there was no significant difference in xenograft tumor size between each group. At fourth and fifth week,there was no significant difference in xenograft tumor size between perindopril group and control group,and the tumor inhibitory rate was 24.6 % and 21.1 %.There was significant difference between benazepril group and control group (t=-2.450 and -3.120,P=0.035 and 0.008),and the tumor inhibitory rate was 33.1% and 45.4%.After treated with perindopril and benazepril,the expression of CD31 in xenograft tumor of nude mice decreased.The MVD of benazepril group was significantly lower than that of control group (10.98±1.18 vs 13.98 ± 1.76; t =-3.732,P =0.002),and there was no significant difference between perindopril group (12.41±1.15) and control group (t=-2.053,P=0.07).ConclusionsBenazepril (6 mg/kg) could significantly inhibit the growth of EC9706 cell line formed xenograft tumor of nude mice.One of the possible tumor inhibitory mechanisms was inhibiting new vessel forming in tumor.Perindopril (4 mg/kg) may have the similar effect.
10.Predictive value of soluble urokinase plasminogen activator receptor in late-onset sepsis in newborn

Objective To investigate the dynamic changes of soluble urokinase-type plasminogen activator receptor (suPAR) and its predictive value in late-onset sepsis in the newborn.Method To collect the data of neonates aged 7 days and older,who were diagonsed to have infections.They were admitted to neonatal intensive care unit of our Hospital from January 2014 to January 2015.The group of sepsis and nonseptic group were assigned according to the diagnostic criteria of sepsis,and a control group was selected without infection.Blood cultures were collected in patients on the first day when infection was identified and the serum suPAR and CRP were measured on the first day,fourth day and tenth day respectively.The controls were tested with suPAR and CRP when infection was excluded.The levels of blood suPAR and CRP in the three groups were compared and the receiver-operating characteristic curve was performed according to serum suPAR level of neonates with sepsis on the first day.Result A total of 65 infants with infections (40 were septic and 25 were non-septic) were enrolled in this study and 20 patients were selected as control group.There were significant differences in serum suPAR and CRP levels between the patients with and without infection (P < 0.001).The level of suPAR in the survivors of the sepsis group was significantly decreased as time went by,and the difference was statistically significant on the 10th day compared with the 1 st day [9.3 (8.2,13.1) ng/ml vs.18.9 (14.8,24.7) ng/ml,P < 0.05].The level of CRP increased first initially and then decreased with time,while the highest level was on the 4th day and the difference was statistically significant compared with the 10th day [19.0 ( 6.8,56.4) mg/L vs.6.4 (2.5,12.0) mg/L,P < 0.05].The levels of serum suPAR and CRP in non-sepsis group were not significantly different (P > 0.05).There were no deaths in the sepsis group and the non-septic group,but the levels of suPAR between survivals and deaths in the infection groups were statistically significant [15.4(10.6,21.6) ng/ml vs.22.6 (15.4,31.9) ng/ml,Z =-2.063,P =0.039].The area under the receiver-operating characteristic curve of serum suPAR was 0.955 (95% CI 0.906 ~ 1.000,P <0.001),and the sensitivity was 90% and the specificity was 100% when the suPAR level was 10.9 ng/ml.Conclusion Early elevated serum suPAR levels were prominently related to the severity of neonatal late-onset sepsis.The level of first day suPAR has a high sensitivity and specificity in the prognosis of sepsis and can be helpful to predict the prognosis.
