2.Basal-like breast cancer

Basal-like breast cancer (BLBC) is a basal cell phenotype and associated with different levels of expression in basal cell keratin and (or) myoepithelial markers in breast cancer,which has a unique genetic phenotype and morphological characteristics.BLBC is prevalent in young woman,and it is easy to relapse and metastasis. Additionally,most BLBC has expression loss of estrogen receptor and progesterone receptor and human epidermal growth factor receptor 2,which limits the targeted therapeutic options for these predominantly triple-negative breast cancers.
3.Intratympanic Gentamycin for Intractable Meniere Disease

Objective To explore the effect of intratympanic gentamycin on intractable Meniere's disease.Methods 10 patients diagnosed of intractable Meniere disease were analyzed respectively.Results 9 patients got A and 2 patients got B in controlling vertigo.1 patient gots B and 9 patients got C in hearing result.Conclusion Intratympanic gentamycin for intractable Meniere disease is an effective method.It should be studied further.
4.The ultrastrucural and immunohistochemical study of BBB in streptozocin induced hyperglycemic rats

Objective To investigate the hyperglycemic effect on ultrastructural morphology and intercellular adhension molecule 1(ICAM 1) expression of BBB. Methods Hyperglycemia model was established by intravenous injection of streptozocin in SD rats. A dynamic observation on the changes of the ultrastructure and the ICAM 1 expression on endothelium of BBB in SD rats with hyperglycemia for 3 days to 8 months was carried out by transmission electron microscopy and immunohistochemical method. Results Swelling was slight in endothelium of BBB of rats with hyperglycemia for 3 days, became obvious and occurred in endfoot of astrocyte for 1 to 2 months, and some microthrombi appeared in cavity of capillary with various malformation and apoptosis was found in endothelium of BBB and neuron surrounding abnormal capillary for 3 to 8 months. With immunohistochemical study, ICAM 1 was expressed sparsely on endothelium of BBB in 3 days after injection of streptozocin, became markedly strong in 3 months, and progressed further in 6 to 8 months. There was no changes on BBB in both SD rats and negative controls. Conclusion Long term hyperglycemia could cause obvious ultrastructural changes of BBB, associated with ICAM 1 expression on endothelium of BBB induced by hyperglycemia
5.James Sheehy Myringoplasty

Objective To apply James Sheehy myringoplasty for treatment of large perforation without residual rim and to observe the efficacy.Methods Between January 2001 and June 2003, 60 patients with large perforation without residual rim had underwent James Sheehy myringoplasty, in which 36 were males and 24 were females, aged from 16 to 64 years and with an average of 45.23 years. Preoperative air-bone gap was 5 to 45 dB, averaged 28.5 dB. Fellow-up was done more than 6 months on 54 patients.The key technic: (1) a vascular skin flap was made on the posterior canal wall and the anterior canal wall skin was excised. (2) canaloplasty was performed until the whole tympanic sulcus can be seen under one microscope view. (3) if necessary, the ossicular chain was explored. (4) overlay technic was used to repair perforation with temporal fascia. (5) the free anterior skin and the posterior canal vascular flap were replaced.Results 52 patients were healed in one stage. In 2 patient, two needle tip-like perforations appeared in the center of the grafting tympanic membrane 3 months after surgery, which were closed after gelatin sponge patching. The shape of the tympanic membrane looked as normal. Symptoms of temporomandabular joint appeared in 6 patients and disappeared 3 days later. Lateral displacement of the graft happened in 2 patients.Conclusion James Sheehy myringoplasty is a reliable technique for treatment of large perforation without residual rim, with high success rate, less postoperative response and good shape of new tympanic membrane.
6.HLA-DPB1, DQB1 genes and their correlation with autoantibodies in type 1 diabetes mellitus

Objective To investigate HLA-DPB1, DQB1 gene alleles and their correlation with autoantibodies in type 1 diabetes mellitus. Methods Sequence-based typing was used to determine the alleles of HLA-DPB1 and -DQB1 in 68 patients with type 1 diabetes and 50 healthy controls. Autoantibodies 〔glutamic acid decarboxylase antibody (GADA), islet cell antibody (ICA), insulin autoantibody (IAA)〕were detected by ELISA. Objective To observe the changes and significance of the activity of nuclear factor kappa B(NF-?B)in peripheral blood mononuclear cell(PBMC)of the critical patients with hyperglycemia,who were treated by intensive insulin therapy.Methods One hundred and twelve critical patients with hyperglycemia were randomly assigned to receive intensive insulin therapy(maintenance of blood glucose at a level between 4.4~6.1mmol/L,57 patients)or to receive conventional treatment(maintenance of blood glucose at a level between 8.0~11.1mmol/L,55 patients).2ml venous blood was collected to extract nucleoprotein in PBMC,and the NF-?B activity of PBMC was detected by electrophoretic mobility shift assay(EMSA)on 24h,4d and 10d.NF-?B oligonucleotide probe was labeled by digoxin.The Gel-Imaging system(UVP company)was used to record optical density values(OD)which represented the NF-?B activity.Results NF-?B activities in PBMC of the patients who received intensive insulin therapy(44.69?10.46,34.47?14.32 and 25.95?11.50 at the 24th hour,4th day and 10th day,respectively)were significantly lower than that of the patients who received conventional treatment(50.37?11.54,43.76?10.27 and 35.87?9.97 at 24th hour,4th day and 10th day,respectively,P

Objective:To study the curative efficacy of decitabine combine with CAG regimen in the treatment of acute myeloid leukemia and its effects on the serum Interferon-γ (IFN-γ),alpha hydroxybutyrate dehydrogenase (HBDH) and lactate dehydrogenase (LDH) levels.Methods:70 cases of patients with acute myeloid leukemia who were treated from February 2013 to October 2016 in our hospital were selected as research objects.According to the random number table,the patients were divided into the observation group (n=35) and the control group (n=35).Both groups of patients were treated conventional treatment.The control group was treated with CAG scheme,intravenous injection of aclarubicin,20 mg each time,granulocyte colony stimulating factor,300 μg each time,subcutaneous intravenous cytarabine,10~15 mg/m2 each time,while the observation group was treated with intravenous drip of decitabine on the basis of control group,15 mg/m2 each time.Then the therapeutic effect,serum interferon-γ (IFN-γ),alpha hydroxybutyrate dehydrogenase (HBDH),lactate dehydrogenase (LDH) levels before and after treatment,incidence of adverse reactions were compared between two groups.Results:After treatment,the total effective of observation group was significantly higher than that of the control group [91.42%(32/35)vs68.57%(24/35)] (P <0.05);the serum IFN-γ,HBDH,LDH levels were significantly higher than those of the control group [(3.21± 1.01)pg/ml vs.(5.13 ± 1.90)pg/mL,(103.62± 26.39)U/L vs.(118.80± 28.60)U/L,(101.36± 27.32)U/L vs.(123.08 ± 30.59)U/L] (P <0.05);the incidence rate of adverse reactions was significantly lower than that of the control group [20.00%(7/35) vs.42.85%(15/35)(P <0.05)].Conclusion:Decitabine combined with CAG regimen was more effective for acute myeloid leukemia than CAG regimen alone,which might be related to reduce the serum levels of IFN-γ, HBDH and LDH.
9.Lipid-lowering effects of gallic acid on glutamate-induced obese mice

AIM To study lipid-lowering effects of gallic acid on glutamate-induced obesity mice.METHODS The obese model was established through subcutaneous injection of 3mg/(g · d)sodium glutamate into neonatal mice.After the model was established,the mice were divided into normal control group,model group,positive control group [simvastatin 30 mg/(kg · d)],high-,and low-dose group of gallic acid [400,200 mg/(kg · d)],and were intragastrically administered for ten weeks.Mice in each group after the last administration were fasted for 12 h except water.Blood was sampled from mouse eyes.The organs and adipose were obtained to determine the organ index and fat index.The levels of HDL-C,TG,LDL-C and TC in serum and liver were determined by using the corresponding reagent kit,and the serum leptin level was determined by ELISA kit and simultaneous determination of SOD,GSH-Px and MDA levels in liver.RESULTS Compared with the normal control group,the body weight and fat weight significantly increased in the model group;the levels of TC,TG and LDL-C in serum and liver significantly increased;the serum leptin level significantly reduced;the activity levels of SOD and GSH-Px in the liver significantly reduced;and the level of MDA significantly increased.Compared with the model control group,the body weight and fat weight significantly reduced in the gallic acid group mice and the levels of TC and TG significantly reduced in the serum and liver;SOD and GSH-Px levels significantly increased,MDA level significantly decreased in the liver.CONCLUSION Gallic acid can significantly reduce the blood lipid level of glutamate-induced obese mice.
10.Influence of pathologic factors on phenotypic transformation of renal tubular epithelial cells in vitro

Objective To study the effects of high albumin, high glucose and low bovine serum on the phenotypic transformation of renal tubular epithelial cells. Methods The normal human kidney proximal tubular eel] line (HKC) was cultured for 30 days in the presence of high albumin (1.5 g/L), high glucose(25 mmol/L) and low bovine serum(2% ) . Morphological changes were observed by electronic microscopy. Immunohistochemistry stain was used to examine the expression of cytokeratin, vimentin, a-SMA, collagen Ⅰ and TGF-pl protein. Western blot was applied to further detect the process of collagen I protein expression, and in situ hybridization was used to examined the expression of collagen Ⅰ gene. Results Renal tubular epithelial cells cultured in high albumin, high glucose and low bovine serum showed obvious morphologic changes, including elongated shape, decrease of microvilli and mitochondria, and increase of rough endoplasmic reticulum under electronic microscopy. Immunohistochemistry stain revealed the reduction of cytokeratin, and enhancement of vimentin, ?-SMA, TGF-?1 and collagen Ⅰ. Western blot demonstrated that the expression of collagen Ⅰincreased in a time-dependent manner, and in situ hybridization showed that collagen type Ⅰ mRNA increased as well. Conclusion High albumin, high glucose and low bovine serum induce phenotypic transformation of renal tubular epithelial cells into mesanchymal cells.
