1.The expression of SQS1 gene and the content of glycyrrhizic acid of Glycyrrhiza uralensis Fisch. in different concentrations of Mn2+.

The transplants of one-year-old Glycyrrhiza uralensis Fisch. were subjected to five concentrations of MnSO4-H2O (0, 1.81, 18.1, 36.2 and 54.3 mg·L(-1)) culturing in vermiculite. qRT-PCR and HPLC were respectively used to measure the relative expression of SQS1 gene and the content of glycyrrhizic acid of G. uralensis in different concentrations of MnSO4·H2O. This is to explore discuss the effects of the expression of SQS1 gene and the accumulation of glycyrrhizic acid by Mn treatment. The results showed both the expression of SQS1 gene and the content of glycyrrhzic acid of G. uralensis tended to rise after the fall of the first with the increase of concentration of Mn treatment. And they were of very significant positive correlation (P<0.01, r=0.737). Relative expression of SQS1 gene reached the highest 7.90 under 18.1 mg·L(-1) MnSO4·H2O treatment. It was very significantly different between 18.1 mg·L(-1) concentration of MnSO4·H2O treatment and CK (0 mg·L(-1)), 1.81, 36.2 and 54.3 mg·L(-1) (P<0.01), and 1.75, 1.37, 1.37, 2.33 times respectively. The content of glycyrrhizic acid reached the highest under 1.81 and 18.1 mg·L(-1) MnSO4·H2O treatment, and there were not significant difference (P>0.05). It was very significantly different between them and other concentrations of MnSO4·H2O treatment (P<0.01). This study suggests the appropriate concentration of Mn treatment could certain promote the expression of SQS1 gene and the accumulation of glycyrrhizic acid of G. uralensis.

Objective To investigate the changing features of microtubule associated protein 2 (MAP2) expression, which was a neural dendrite marker, of epileptic rat and the intervention results after using Diazepam. 3.Clinical and MRI features of!multiple system atrophy

Objective To explore the clinical and MRI features multiple system atrophy (MSA) and the significance of diagnosing its clinical subtypes.Methods Clinical data and MRI features of 28 patients with MSA were retrospectively analyzed and compared.Results Cerebellar signs (75.0%) were dominant clinic features in olivopontocerebellar atrophy (OPCA). Brain MRI showed atrophy of pontine (91.7%) and cerebellar vermis (91.7%), fourth ventricle dilatation (83.8%) and T2WI high intensity in pontocerebellar region (63.6%). Extrapyramidal signs (80.0%) were found in striatonigral degeneration (SND) with apparent basal ganglia changes such as putamen atrophy (60.0%) and lineal T2WI high intensity of the lateral margin of the putamen (putamen slit) (80.0%). Shy-Drager syndrom (SDS) presented mainly with autonomic nerve system (81.8%), and autonomic nerve system failures appeared earlier and more severe than the other two subtypes of MSA. Conclusions MRI examination may be useful in diagnosis of MSA and its subtypes. Pontine atrophy, T2WI high intensity, especially pontine cross sign may support diagnosis of OPCA. However, putamen atrophy and putamen slit are the specific imaging signs in SND.

BACKGROUND: Plenty of materials have been used as stent in total auricular reconstruction with varying advantages and disadvantages. The in vitro prepared tissue engineered complex cartilage stent has been not applied in clinic.OBJECTIVE: To observe the ultrastructure changes of surrounding tissues following total auricular reconstruction with nickel-titanium (NiTi) shape memory alloy stent in vivo.DESIGN, TIME AND SETTING: The randomized, controlled, animal experiment was performed in the Central Laboratory of EENT Hospital of Fudan University, Anhui Provincial Hospital and Department of Nuclear Medicine, Huashan Hospital of Fudan University from April 2004 to October 2005.MATERIALS: A total of 12 New Zealand rabbits were randomized divided into the skin-expanded and unexpanded groups, with 6 animals in each group.METHODS: Each animal underwent subcutaneous implantation with a NiTi alloy stent like human auricle. In the skin-expanded group, the stent was implanted after skin expansion, while no treatment in the unexpanded group.MAIN OUTCOME MEASURES: The ultrastructure changes of surrounding tissues were detected by scanning electron microscope at months 6 and 12 after implantation.RESULTS: Structure of nickel-titanium alloy implant stent make fibrovascular ingrowth more easily. The surface of nickel-titanium alloy stent was smooth without corrosion. There was no evidence of infection in the host tissue. Microstructure of Ni-Ti stent wires showed no morphologic and functional changes after subcutaneously implanted in vivo, and the difference between 2 groups had no significance.CONCLUSION: NiTi shape memory alloy stent can smoothly pass the process of fibrovascularization with less complication.According to the examination by SEM, the nickel-titanium stent showed good biocompatibility.
6.Natural orifice transluminal endoscopic surgery in non-survival porcine models: access and technique

Objective To evaluate the feasibility, advantages and pitfalls of trans-gastric, trans-colonic and trans-umbilical peritoneoscopy, and to compare pure natural orifice transluminal endoscopic surgery (NOTES) with hybrid NOTES. Methods Two female pigs of 30kg were used in the study, one for pure NOTES, the other for hybrid NOTES. Each pig received trans-gastric, trans-colonic and trans-umbilical peritoneoscopy, respectively, followed by postmortem examination for organic lesions and closure of incisions.The two approaches were compared in terms of intra-operative situation and manipulation. Results The vital signs of the pigs were stable during the procedures, with no peri-incision organic lesions. There were blind areas in trans-gastric as well as in trans-colonic peritoneoscopy. It was easier to close the colonic incision than the gastric one. The situation in trans-umbilical peritoneoscopy was similar to laparoscopic surgery. Hybrid NOTES seemed superior to pure NOTES with regard to safe access, stable pneumo-peritoneum and adequate visualization. Conclusion Three routes for NOTES appear to be reasonable and feasible. Hybrid NOTES is more effective and safer than pure NOTES with current instruments.
7.Clinical application of auricular cartilage in intact bridge tympanomastoidectomy

OBJECTIVE To investigate and evaluate the effects of intact-bridge tympanoplasty using auricular cartilage in treating chronic otitis media. METHODS A retrospective review of 86 cases undergoing an intact-bridge tympanomastoidectomywas performed from July,2013 to May,2016.The auricular cartilage was placed on the attic and tympanic antrum entrance to reconstruct the attic. RESULTS The outcome of dry ear and well-epithelialized mastoid cavities was achieved after 4-6 weeks. After 6-month follow-up no recurrence occurred. The pure-tone average threshold improved by (25.4±6.3) dB HL after operation,and the mean air bone gap was elevated. CONCLUSION On the basis of combination of closed and open techniques,the IBM technique is a optimal cure strategy for chronic otitis media,which can not only remove the pathological changes thoroughly to prevent disease recurrence but also maximally improve the hearing of the diseased ear.The closure of attic with auricular cartilage can ensure the space of the ossicular chain.
8.Study of using postaural bipedicle musculoperiosteal flap in mastoid obliteration

OBJECTIVE To explore the dry ears and improving the auditory level of postaural bipedicle musculo-periosteal flap to obliterate the mastoid cavity. METHODS 78 cases(ears) of chronic otitis media were treated with radical mastoidectomy,open-ended tympanoplasty and conchaplasty,and received mastoid plombabge by postaural bipedicle musculo-periosteal flap.RESULTS In 78 cases(ears),the time of dry ears were reduced to 10 or 15 days and the auditory level in 58 ears was impoved(17.2?4.9) dB after treatment.CONCLUSION The surgery with postaural bipedicle musculo-periosteal flap to plombabge the mastoid cavity can achieve the epithelization in the operation cavity more quickly,achieve a short duration to dry ear shortly and improve the auditory level.

Moreover,mRNA and protein expressions of IL-1β and COX-2 in different experiment groups were determined by RT-PCR and Western blotting analysis. RESULTS:NF-κB p65 immunoreactivity began to increase in the nuclear fraction at 2 h (P<0.05),kept rising at 12 h (P<0.05) and returned to control level at 24 h after epilepsy seizures. Furthermore,3-AB sharply decreased the accumulation of NF-κB p65 in nucleus (P<0.05). In addition,3-AB significantly decreased the mRNA and protein expressions of IL-1β and COX-2 which obviously increased in hippocampus at 6 h after epilepsy seizures (P<0.05). CONCLUSION:Seizures triggers NF-κB nucleus translocation and promotes the expressions of IL-1β and COX-2 in hippocampus. In addition,poly (adenosine diphosphate-ribose) polymerase inhibition by 3-AB suppresses NF-κB associated inflammatory pathway in epileptic rat hippocampus.
