1.Application of Gukang Gold Far-Infrared Magnetic Therapeutic Paste to Treatment of Lateral Epicondylitis

Objective To apply Gukang gold far-infrared magnetic therapeutic paste to the treatment of lateral epicondylitis.Methods 64 cases of lateral epicondylitis patients were randomly divided into treatment group and control group,each with 32 ones.The subjects from the treatment group were treated with Gukang gold far-infrared magnetic therapeutic paste,and the remained with ibuprofen sustained-release capsules.The results after one and two courses were analyzed statistically.Results After two-course treatment,the patient from the treatment group had an higher improvement rate than the others from the control group,with a P value less than 0.05.The treatment course of the treatment group was shorter than that of the control group,with a P value less than 0.05.Conclusion When compared with ibuprofen sustainedrelease capsule,Gukang gold far-infrared magnetic therapeutic paste is more effective for the treatment of lateral epicondylitis.
2.Inhibitive Effects of Somatostatin on LPS-stimulated Rat Kupffer Cells

OBJECTIVE To investigate the effects of somatostatin(SST)on expression of NO,TNF? and 5-lipoxygenase(5-LO)in primary cultured rat Kupffer cells induced by LPS.METHODS Kupffer cells isolated from Sprague-Dawley rats were cultured in the presence of LPS added alone or with SST for 12,24 and 48 h,and the production of TNF? was assessed in culture supernatants by ELISA and that of nitric oxide(NO)by nitrate reductase method.5-LO mRNA expression was assessed by semiquantitative RT-PCR.RESULTS Vehicle-stimulated Kupffer cells produced a basal amount of NO,TNF? and expressed 5-LO mRNA.Kupffer cells stimulated by LPS secreted significantly increased amounts of NO and TNF?(P0.05).CONCLUSIONS SST modulates NO,TNF? production and 5-LO mRNA expression by LPS-stlimulated Kupffer cells.
3.Bioluminescence imaging evaluation of the inhibitory effect of lidamycin on lung metastasis of human fibrosarcoma in athymic mice.

This study is to investigate the inhibitory effect of lidamycin (LDM) and its combination with methotrexate (MTX) on lung metastasis of fibrosarcoma by bioluminescence imaging in athymic mice. A stable luciferase transfected HT-1080 cell line was constructed and the capability to establish experimental lung metastasis in athymic mice was confirmed. The optical imaging system was applied to evaluate the formation of lung metastasis in vivo. In addition, metastatic nodules were counted for the evaluation of inhibition rates. As shown, the fluorescent intensity of luciferase-transfected HT-1080 cells was colinear with the cell population and the minimal detected cell population was 100 cells/well. Optical imaging showed that the fluorescent intensity of treated group was apparently lower than that of the control. The inhibition rates of lung metastasis by LDM alone at 0.025 mg x kg(-1) and 0.05 mg x kg(-1) were 53.9% and 75.9%, respectively, while that of MTX alone at 0.5 mg x kg(-1) was 70.2%. The combination of LDM at 0.025 mg x kg(-1) and MTX at 0.5 mg x kg(-1) showed an inhibition rate of 88.7%. The coefficient of drug interaction (CDI) was 0.82. The results herein demonstrated that LDM alone had strong anti-metastasis effect on human fibrosarcoma HT-1080 and the inhibition efficacy is strengthened when combined with MTX.
4.The synergistic effect of lidamycin and rituximab on human B cell lymphoma.

This study aimed to investigate the synergistic effect of lidamycin (LDM) and rituximab on human B cell lymphoma Ramos cells. Cell proliferation was measured using MTS assay, cell apoptosis was analyzed by Annexin V-FITC/PI assay, the expression of apoptosis related proteins was analyzed by Western blotting, and the in vivo lymphoma inhibition was verified using BALB/c mice inoculated via tail vein using Ramos cells which stably expressed pEGFP-N1 plasmid. The results showed that, after the pretreatment with rituximab for 48 h, rituximab and LDM showed significantly synergistic effects on cell proliferation. Cells in combined treatment group had a higher apoptosis rate than that in LDM treatment group. Compared with the LDM treatment group, the expression of apoptosis-related proteins such as Cleaved caspase-3, Cleaved caspase-7, Cleaved caspase-9 and Cleaved PARP in combined treatment groups increased, and expression of cIAP-2 and Bcl-2 decreased. The result of in vivo experiment showed that, in the combined treatment group, the survival time of BALB/c mice was significantly longer than the mice in control group and LDM treatment group, and the degree of tumor accumulation and metastasis to lymph nodes and spleen was lower.
5.Pharmacokinetics of Balofloxacin Tablets in Chinese Healthy Volunteers

OBJECTIVE:To study the pharmacokinetics of balofloxacin tablets in Chinese healthy volunteers.METHODS:A single dose of 100 or 200 mg balofloxacin tablets were given to 12 healthy volunteers in a randomized crossover design.Concentrations of balofloxacin in plasma and urine were determined by HPLC with data processed by DAS(drug and statistics)software.RESULTS:After administration of balofloxacin 100 mg and 200 mg,the Cmax were(0.970?0.245)?g?mL-1 and(1.849?0.466)?g?mL-1;the tmax were(1.25?1.10)h and(1.24?0.81)h;the t1/2 were(7.14?1.01)h and(7.11?0.72)h;the AUC0~36 were(7.309?1.368)?g?h?mL-1 and(15.214?1.727)?g?h?mL-1;AUC0~∞ were(7.531?1.386)?g?h?mL-1 and(15.695?1.762)?g?h?mL-1;the accumulative eliminating rates within 36 h in urine were(64.47?11.56)% and(63.24?11.93)%,respectively.CONCLUSION:The pharmacokinetics of balofloxacin in healthy volunteer after oral administration was characterized by high peak concentration and long half life.The method is sensitive,accurate,reliable and specific,and can satisfy the requirements for pharmaceutical study.
6.Effect of kebimin decoction on level of Th1、Th2-typed cytokines in patients with allergic rhinitis

Objective:To observe the effects of sera Th1/Th2-typed cytokines in patients allergic rhinitis (AR) who were treated with kebimin decoction(KD),and to study the therapeutic mechanism of KD in AR.Methods:60 patients of AR were randomly divided into two groups (treating group,TG and control group,CG). Patients in TG were treated with KD,while those in CG were treated with Xinfang Rhinitis Capsule before and after treatment,the levels of serum Th1-typed cytokines IFN-?、IL-2、IL-12 and Th2-typed cytokines IL-4、IL-5、IL-10 were determined by enzyme-linked immune sorbent assay (ELISA); and compared with 30 healthy controls.Results:The levels of serum Th2-typed cytokines IL-4、IL-5、IL-10 were higher and the levels of Th1-typed cytokines IFN-?、IL-2、IL-12 were lower in AR than that in healthy control ( P 05).Conclusion:Kebimin decoction is highly effective in treating allergic rhinitis by regulating the levels of Th1、Th2-typed cytokines,correcting the imbalance Th1/Th2-typed cytokines network.
7.Analysis on drug resistance of 196 strains of Mycobacterium tuberculosis in patients with HIV/AIDS complicating tubercolosis

Objective To study the drug resistance situation of Mycobacterium tuberculosis strains isolated from the patients with HIV/AIDS complicating tuberculosis (TB).Methods One hundred and ninety-six inpatients with AIDS complicating TB in our hospital from January 2012 to December 2015 were collected.The drug resistance situation of Mycobacterium tuberculosis strains cultured from sputum/tissue fluid/fiber liquid and pathological tissue submitted samples in the patients with AIDS complicating active TB was analyzed.The BACTEC MGIT960 systems was adopted to perform the bacterial identification.The drug sensitivity test was conducted by using 960 culture testing system.Results One hundred and ninety-six strains of Mycobacterium tuberculosis were cultured,the total drug resistance rate of Mycobacterium tuberculosis was 26.02%.These cases were divided into the >100/μL and ≤100/μL groups according to different CD4+ T cells count.The drug resistance situation of Mycobacterium tuberculosis to 4 kinds of first line anti-TB drugs were compared between these two groups.The comparison results found that the resistance rate of Mycobacterium tuberculosis strains to the first line anti-TB drugs had no statistical difference between the two groups (P>0.05).The resistance rates of first line anti-TB drugs from high to low were isoniazid,rifampicin,streptomycin and ethambutol.The drug resistance rate of Mycobacterium tuberculosis strains to 5 types of drug resistant TB had no statistical difference(P>0.05).Conclusion The drug resistance rate in the initial patients with HIV complicating TB is consistent with the average initial drugresistance level of TB patients in our country.The TB drug resistance rate in the patients with HIV complicating TB has no correlation with CD4+ T cells count.

Objective:To observe the clinical efficacy of Tuina(Chinese therapeutic massage)plus electroacupuncture in treating migraine due to liver-Yang hyperactivity and the effects on the serum levels of calcitonin gene-related peptide(CGRP)and prostaglandin(PG)E2. Methods:A total of 122 patients with migraine due to liver-Yang hyperactivity were recruited and randomized into a control group and an observation group,each consisting of 61 cases.The control group was given Tong Nao Huo Luo acupuncture(acupuncture treatment for unblocking brain collaterals),and the observation group was Tuina treatment focusing on cervical Ashi points in addition to the intervention received by the control group.Both groups were treated once daily for 21 consecutive days.When the intervention finished,the two groups were observed for changes in the headache score,symptom and sign scores of traditional Chinese medicine(TCM),the severity of impact on life,and serum CGRP and PGE2 levels.The clinical efficacy was compared after 21 d of treatment. Results:The observation group had a higher total effective rate than the control group,90.2%versus 73.8%(P<0.05);after treatment,the headache and TCM symptom and sign scores decreased in both groups(P<0.05)and were lower in the observation group than in the control group(P<0.05);the migraine's impact on life was less severe in the observation group than in the control group(P<0.05);the levels of serum CGRP and PGE2 dropped in the two groups(P<0.05)and were lower in the observation group(P<0.05). Conclusion:Tong Nao Huo Luo acupuncture can produce more significant efficacy in treating migraine due to liver-Yang hyperactivity when combined with cervical Tuina at Ashi points,better alleviating the headache,improving TCM symptoms and body signs,and reducing the impact of headache on life.The mechanism may be associated with inhibiting the expression of serum pain factors CGRP and PGE2.
10.Effect of electroporation mediated transfecting recombinant plasmid pIRES-hVEGF165-EGFP on angiogene-sis of distraction area during early mandibular distraction osteogenesis

Objective To explore the effect of electroporation mediated gene therapy on angiogene-sis of the distraction area during early mandibular distraction osteogenesis (DO). Methods Thirty-two New-Zeland rabbits were randomly divided into 4 groups: group A: recombinant plasmid pIRES-VEGF165-EGFP and electroporation; group B: recombinant plasmid pIRES-VEGF165-EGFP; group C: normal saline (NS) and electroporation and group D: control group. The rabbits were sacrificed at 1d, 3d, 7d and 14d after injection, respectively. The distraction area tissue was removed for histological examination and electron microscopy, and immunohistochemical stain for CD34 was performed to detect the microvessel density. Results Generation of vascular endothelial cells (VEC) in the group A and group B were active, and majority of VEC in groups C and D took on early change of cataplasia and apoptosis. The immunohistochemical stain for CD34 showed that it expressed weakly at the first day after transfection, and at 3,7,14 days after transfection, CD34 of VECs in the distraction area expressed positively. CD34 expression was the strongest in group A (P<0. 05), and there was significant difference among three groups and different time, respectively.Compared to each other, CD34 of VECs expressed positively with a tendency to rise in the groups A and B. But it fluctuated at the level of the expression at the first day in the groups C and D. Conclusion Electroporation-mediated transfecting recombinant plasmid could promote angiogenesis during early stage of mandibular DO. It could promote local vascular proliferation and penetration, increase the blood flow of broken ends in fractured bone. It indicates that electroporation-mediated transfecting recombinant plasmid play an important role in regulating and promoting growth and reparative process of the bone.
