1.Influence of multidrug resistance polymorphism on CsA-associated hepatotoxicity

Objective To investigate the influence of MDR polymorphism on CsA-associated hepatotoxicity. Methods PCR/RFLP was used to analyze the genotypes of MDR exon26 in 187 recipients. CsA-associated hepatotoxicity was evaluated among groups being classified according to the genotypes. Results MDR exon26 takes 3 genotypes:CC,CT and TT, in the following ratio: 29.4%(55/187), 40.1%(75/187), 30. 5%(57/187). Among the same range of whole blood CsA concentration, incidence of CsA-associated hepatotoxicity is markedly higher in the CT/TT group than that in in patients with CsA-associated hepatotoxicity between the CC group and the CT/TT group is signifiimportant role in the development of CsA-associated hepatotoxicity.
2.Effect of hemorheology on ultrasonic doppler blood flow spectrum diagram.

The present research aims to point out the long-existing defect of analyzing the spectrum diagram only from the perspective of haemodynamics instead of haemorheology. In the light of the theories of haemodynamics and haemorheology, the causes of spectrum diagram formation of carotid artery blood at the rapid and slow flow can be clarified completely and accurately. Four conclusions have been drawn in the end. As long as the velocity gradient is large enough, obvious red blood cells concentrate to the shaft even in the big or bigger blood vessels; the spectrum diagram is the powerful proof of the two phase flow model of blood; the spectrum diagram can be completely and accurately analyzed only by combining haemodynamics with haemorheology; and only when the red blood cells concentrate to the shaft, the big or bigger blood vessels can be regarded as haemogeneous fluid.

3.The etiology and pharmacologic treatment of obesity

Obesity is caused by the imbalance between caloric intake and energy expenditure. It is also influenced by genetic factors. Many genes,such as ob gene, ? 3 receptor gene, ucp 1 gene, may involved in the prevalence of obesity. Pharmacologic treatments include anorexigenic agents, lipase inhibitor, ? 3 receptor agonists and many newer antiobesity drugs, which provide prospective ways in treating obesity.
4.Relationship between tumor angiogenesis and invasion, metastasis and prognosis of hepatocellular carcinoma

Objective To investigate the relationship between the tumor angiogenesis and the clinicopathological variables of hepatocellular carcinoma (HCC) and determine whether tumor angiogenesis can be used as an indicator for predicting invasion, metastasis and prognosis of HCC. Methods Using the immunohistochemical S-P method, CD34 expression was detected in liver tissue from 20 normal individuals, 20 patients with liver cirrhosis and 85 patients with HCC and juxtacancerous tissues and the microvascular density (MVD) determined in those from 85 patients with HCC. Results CD34 was mainly located in the cell cytoplasm of endothelial cells. The CD34 staining was confined to vessels in the portal triad in the above-mentioned various tissues. In HCC tissues, however, sinusoid-like vessels reacted intensively with anti-CD34 and the MVD was 156.5±62.4 (per 0.74 mm2). MVD was correlated to tumor size, number, differentiation, emboli in the portal vein and capsule (P<0.05 or 0.01) but not to HBsAg and AFP (P>0.05). It was also correlated with prognosis of HCC. The mean tumor-free survival of MVD ≥156 was 14 months while that of MVD <156 was 53 months (P<0.01). Conclusion The expression of CD34 in sinusoid-like vessels in HCC tissues represents the neovascularization. Angiogenesis in HCC is associated with the progression of HCC. MVD can be used as an indicator to judge the invasion, metastasis and prognosis of HCC.
5.Indications and taboo of fecal microbiota transplantation in children

Fecal microbiota transplantation(FMT),as a special organ transplantation,can be used to reconstruct the intestinal flora.It has been concerned by clinical practice,but it has not become a routine treatment method.With the gradual deepening of the study of FMT,the refractory difficile fusiform bacillus infection,inflammatory bowel disease,irritable bowel syndrome,chronic fatigue comprehensive syndrome and some metabolic disorders related to intestinal diseases were obtained using.In recent years,there are some reports about the application of FMT in children's diseases.Combining with the relevant literature at home and abroad,this paper reviews the indications and the taboo of the application of FMT in children,the aim is to provide a reference for FMT to be better applied in pediatric clinical.

8.A case report of enzyme replacement therapy for glycogen storage disease typeⅡ

Objective To study the clinical characteristics and the effect of enzyme replacement therapy for late-onset glycogen storage disease typeⅡ(GSDⅡ). Methods The clinical, laboratory data and the result of genetic testing were retrospectively analyzed in a GSDⅡchild, the effect of enzyme replacement therapy was followed up and the relevant literature was reviewed. Results The patient had motor regression after 1 year old, the serum creatine kinase level is from 675 to 1286 U/L. The EMG test showed myopathic change, acid alpha-glucosidase activity is 12.0 nmol/(g·min), next generation sequencing of genetic muscle diseases panel found the GAA compound heterozygous mutations, both were tiny variations, and muscle biopsies showed the typical pathological features of GSD. The patient was given human recombinant of alpha glucoside enzyme 20 mg per kilogram of body weight, once every other week for 1 year. The weakness of the patient’s muscle strength had no obvious aggravation. Conclusions Early and adequate enzyme replacement therapy is the only possible treatment for GSDⅡ.

Methods: Summarizing the experiences, lessons and questions in scientific research, edu-cation and clinical applying of hemorheologic in twenty and more years, clarifying the harm of high viscosity to health. Re-sults: Stating the foundation, necessity and feasibility of the overall and deep development have been possessed. Conclusions: Hemorheologic can be developed comprehensively and in depth provided that studying good experiences, cultivating talent, playing the role of learning organizations and conquering the existing questions.
10.Comparison of life quality between surgical castration and drug castration in advanced prostate cancer

Objective To explore the effects of surgical castration and intermittent androgen blockade (IAB) in patients with advanced prostate cancer.Methods 115 cases of advanced prostate cancer were selected including 48 cases with drug castration receiving IAB and 67 cases with surgical castration.Before the treatment, 6 months and 1 years after the treatment, the quality of life of the two groups was investigated by the quality of life questionnaire (EORTC QLQ-C30 +QLQ-PR25).Results Compared with before treatment, the general health status of the patients in the drug castration group and the surgical castration group were significantly improved (P<0.01), the whole body and the local pain were relieved (P<0.01) and the micturition symptoms were controlled ( P<0.01 ).The overall quality of life of drug castration group and surgical castration group were significantly decreased (P<0.05), and the degree of sexual function was particularly affected (P<0.01), but the body function of patients in drug castration group restored to a certain extent in drug castration group after 1 year’s treatment(P<0.01), while the quality of life in patients of surgical castration did not reverse.Conclusion Both of surgical castration and androgen deprivation therapy can reduce the quality of life of patients with advanced prostate cancer, but the adverse effects of drug treatment on the quality of life can be recovered in treatment intermission.
