1.Study on different levels of biochemical indexes in adult health examinees

Objective To investigate the metabolism of blood glucose and lipid levels in health examination of adults from Hain-an district,Wuhai city,Inner Mongolia and to discuss the preventive and health protection measures.Methods 1 395 adults receiv-ing health examination were enrolled.The glucose and lipid were tested and analyzed.Results The levels of glucose and triglyceride in males were significant higher than that in females,P <0.05.The differences of abnormal rates of each index between male and fe-male,among different age groups were statistically significant,P <0.05 as well.The overall level and abnormal rate of each index increased gradually with age(except HDL-C with irregular fluctuations),P <0.05.Conclusion There are certain abnormal rates in blood glucose and blood lipid levels in those examinees and the rates tend to increase with age.Effective health education needs to be strengthened and the preventive and treatment measures should be set up.
2.Alterations of CD4+CD8+T cells in peripheral blood of patients with tuberculosis and its clinical sig- nificance

Objective To characterize CD4+CD8+double-positive T ( DPT) cells in PBMCs from patients with tuberculosis(TB).Methods PBMCs were isolated from peripheral blood samples collected from patients with TB and healthy subjects.The subsets and percentages of CD4+T, CD8+T and DPT cells in PBMCs were determined by flow cytometry.Cell surface markers ( CD45RO, CCR7 and CD25 ) and intracellular cytokines ( IFN-γand TNF-α) were detected directly and after ESAT-6/PPD stimulation.Re-sults Patients with TB showed a significantly increased DPT cells as compared with the cured individuals and healthy subjects (P<0.005).The levels of DPT cells were gradually decreased down to normal upon the treatment of pharmacotherapy.DPT cells expressed higher levet of CD25 than CD4+T and CD8+T cells ( P<0.005 ) . DPT cells could express more IFN-γand TNF-αupon the stimulation of ESAT-6/PPD (P<0.005).The analysis of memory phenotype indicated that DPT cells were memory T cells.Conclusion DPT cells in peripheral blood of the patients with tuberculosis played a critical role in protective immunity against tuberculosis.The alterations of DPT cells in PBMCs during the period of pharmacotherapy might be a potential indicator for the prognosis of pulmonary tuberculosis.
3.Influence of acupuncture and moxibustion on proliferation and differentiation of neural stem cells in olfactory bulb of senescence-accelerated mouse P8.

This study was carried out to defect the effects of acupuncture and moxibustion therapy on the proliferation and differentiation of neural stem cells in the olfactory bulb of senescence-accelerated mouse P8. Immunofluorescence double staining methods(BrdU/Nestin, BrdU/MAP-2 and BrdU/beta-tubulin III) were used. The results show that both acupuncture and moxibustion could improve proliferation of neural stem cells, but only moxibustion could induce differentiation of neural stem cells into neuron.

4.Sharing database platform for acute pancreatitis

Acute pancreatitis ( AP ) is one of the common disorders of gastrointestinal system in clinical practice characterized by sudden onset, long course of disease, complex complications, severe conditions, heavy social and economic burden and the like. Recent years has seen a decreasing mortality due to the update and promotion of a series of therapeutic concepts. However, as the imbalance of comprehensive healthcare levels among different regions in China, substandard diagnosis and treatment of AP still exist, lowering the overall success rate of AP treatment. Moreover, the waste and miss of AP patients' data during the treatment period affect our original innovation in the field of AP research. Thus, after the accumulation of long-term exploration and practice, we integrated diverse data resources and established a single-disease database of AP by cooperating with computer and artificial intelligence ( AI) professionals, finished the deployment of hardware and network arrangement, generated a logical system of "business database→scientific database→sharing database", and built AP data sharing platform (www. ap-database. com), providing the fundamental functions including browse, search, upload and download ( with permissions) . We hope to share the abundant and free medical date with clinicians, computer or AI practitioners and attract communications and cooperation from different fields, which may deepen new values from current data mining and advance the research on AP.

Objective:
To explore the association between DNA methylation and body mass index (BMI) using Mendelian randomization analysis.
Methods:
A total of 469 participants were selected from the Chinese National Twin Registry in 2013, who were living in Shandong, Jiangsu, Zhejiang, and Sichuan provinces, and at least 18 years of age. A questionnaire survey and physical examination were conducted to collect demographic, clinical, and behavioral information. Peripheral blood cells were collected to detect genotype and methylation status. Association analyses between DNA methylation and BMI and between CpGs and cis-SNP were conducted. With rs748212 as the instrumental variable, the association between cg15053022 and BMI was explored using the Mendelian randomization method.
Results:
A total of 469 participants were selected. The mean age of participants was (44.8±13.2) years and the BMI was (25.0±3.8) kg/m2. Nine BMI-related DNA methylation sites were found and DNA methylation site cg15053022 in the ATP4A gene was negatively associated with cis-SNP rs748212 (β=-0.020); the mean methylation level of AA, AC, and CC were 0.212±0.025, 0.242±0.024, and 0.264±0.028, respectively. rs748212 was associated with BMI (β=0.04,
6. Efficacy of antiplatelet therapy with ticagrelor monotherapy after short-term double antibody therapy in patients with acute coronary syndrome after PCI

AIM: To observe the clinical efficacy of antiplatelet therapy with ticagrelor monotherapy after short-term double antibody therapy in patients with acute coronary syndrome (ACS) treated with percutaneous coronary intervention (PCI). METHODS: A total of 172 patients with ACS who had undergone PCI from October 2018 to April 2022 in our hospital and Subei People's Hospital of Jiangsu Province were selected and divided into three groups according to the patients' medication status: 50 patients in group A (tegretol alone group); 62 patients in group B (aspirin enteric coated tablets + tegretol group); and 60 patients in group C (aspirin enteric coated tablets + clopidogrel hydrogen sulfate group). The patients were observed for postoperative recurrent angina pectoris, in-stent restenosis, revascularization, heart failure, and bleeding. RESULTS: There were no significant differences in postoperative recurrent angina, in-stent restenosis, re-hematologic reconstruction, heart failure, and bleeding between the 3 groups. CONCLUSION: There is no increase in ischemic events, heart failure in patients treated with tegretol antiplatelet therapy alone after a short course of dual antibiotics after PCI and it reduces risk of bleeding.
7.Development, and reliability and validity testing of a knowledge needs questionnaire of respiratory rehabilitation training for in-service healthcare workers

ObjectiveTo develop a questionnaire on the knowledge needs of respiratory rehabilitation training for in-service healthcare workers, and test its reliability and validity. MethodsA theoretical framework was constructed based on the contents of the respiratory rehabilitation course modules of American Association of Cardiopulmonary Rehabilitation and American Association for Respiratory Care. The first draft of the questionnaire was prepared through literature searches and analyses, semi-structured interviews, and expert group discussions. The reliability and validity of the questionnaire was tested by interviewing 17 respiratory rehabilitation professionals and 16 respiratory rehabilitation experts by correspondence, and investigating 380 cases of in-service healthcare workers engaged in respiratory rehabilitation. ResultsThe knowledge needs questionnaire of respiratory rehabilitation training for in-service healthcare workers included progress in respiratory rehabilitation, physiological basis and diagnosis of chronic lung diseases, individualized assessment and management principles of chronic lung diseases, treatment of chronic lung diseases and guideline-recommended medications, oxygen therapy, exercise assessment and exercise prescription, tobacco addiction and cessation, nutritional assessment and weight management, psychological screening and intervention, self-management and health behavior promotion, and effectiveness assessment of respiratory rehabilitation, totaling eleven dimensions and 52 items. The total Cronbach's α was 0.921, the total folded half reliability was 0.904, the total retest reliability was 0.922, and the Cronbach's α of each dimension ranged from 0.909 to 0.953; the folded half reliabilities ranged from 0.882 to 0.924, and the retest reliabilities ranged from 0.908 to 0.950. The validity of the questionnaire's content was 0.902. In exploratory factor analysis, eleven metric factors were extracted, and their cumulative variance contribution rate was 75.324%. ConclusionThe questionnaire on the knowledge needs of respiratory rehabilitation training for in-service healthcare workers is reliable and valid, and can be used to assess the knowledge needs of healthcare workers participating in in-service training in respiratory rehabilitation.
8.Change trend of birth weight of twins in China, 1995-2012.

OBJECTIVETo understand the change trend of birth weight of twins in China from 1995 to 2012.
METHODSA total of 10 827 twins born between 1995 and 2012 registered in national twin registry system in 8 provinces in China were included in this study. A retrospective study was conducted to analyze the change trend of twin's birth weight.
RESULTSMean birth weight of twins decreased by 0.01 kg every five years after adjusting sex, ovum type and place of birth (P = 0.041). The birth weight decreased both in superior twin and in inferior twin over time. After 2007, the birth weight of superior twin decreased by 0.008 kg each year, while the birth weight of inferior twin decreased by 0.014 kg each year. Mean relative difference in birth weight and birth weight discordant rate decreased before 2002, then increased from the lowest point 8.16% and 16.20% to highest point 9.99% and 22.40% respectively.
CONCLUSIONThe birth weight of twins in China decreased between 1995 and 2012, while the birth weight discordant rate increased, suggesting that close attention should be paid to the discordance of twin body weight to reduce the risk of adverse health outcome.

9.Heritability of body mass index on Chinese adult twins from nine provinces/cities in China.

OBJECTIVETo explore the heritability of body mass index (BMI) in twins across different regions and genders in China.
METHODSA total of 11 122 adult twin pairs from the Chinese National Twin Registry were interviewed. A structural equation model was used to estimate the heritability of BMI.
RESULTSThis study included 6 226 monozygotic twin pairs and 4 896 dizygotic twin pairs, with the age range as 25-85 (39.0 ± 10.8) years. Under stratified analysis by region, results showed that the highest and lowest rates on heritability of BMI in men were seen in Tianjin and Zhejiang, as 67.8% (95% CI: 50.1%-85.8%) and 42.1% (95% CI: 27.2%-60.9%), while in women were seen in Sichuan and Heilongjiang as 56.2% (95% CI: 47.5%-70.0%) and 11.2% (95% CI: 0.0%-31.7%), respectively. Results from the stratified analysis showed that, by gender, the heritability of BMI in men was higher than that in women from the same region. The biggest differences of heritability of BMI between men and women were seen in Heilongjiang as 55.3% (95% CI: 35.5%-80.0%) and 11.2% (95% CI: 0-31.7%), while the smallest differences were seen in Sichuan as 61.5% (95% CI: 40.7%-86.4%) and 56.2% (95% CI: 47.5%-70.0%), respectively.
CONCLUSIONThe heritability of BMI across different regions and genders showed certain differences in the Chinese twins.

10.A co-twin control study on birth weight, overweight and obesity among children younger than 18 years old in China.

OBJECTIVETo analyze the associations between birth weight and overweight/obesity among children.
METHODSA total of 8 267 twin pairs younger than 18 years old from the Chinese National Twin Registry were included in the study. Associations between birth weight, childhood BMI and overweight/obesity were explored by this co-twin control study.
RESULTSAfter adjusting for sex and zygosity, when birth weight had an increase of 0.5 kg per fold, the OR values for overweight and obesity were 1.87(95%CI: 1.40-2.48) for 2-6 year olds, 1.69 (95%CI: 1.16-2.46) for 6-12 year olds and 1.28 (95%CI: 0.80-2.07) for 12-18 year olds.
RESULTSfrom the stratified analysis in the 2-6 year-olds, statistically significant differences were seen. When birth weight increased 0.5 kg per fold, the risk of overweight and obesity increased by 0.87 times among the dizygotic twins, more than that of the monozygotic twins (OR=1.86, 95%CI:1.24-2.81). The risk for male twins was 1.12 times higher than that of female twins (OR=1.65, 95%CI:1.11-2.44).
CONCLUSIONSBirth weight seemed associated with overweight and obesity for kids at early childhood or at age for schools. However, guidance on the implementation of public health interventions is still needed on these children.
