Aim To explore the function of immune modulation of anti-HBV iRNA in patients with chronic hepatitis B(CHB) Methods Peripheral blood lymphocyte iRNA was prepared from anti-HBs positive human body with HBV complete clearance after HBV infection(h-iRNA). The effect of h-iRNA on HBV specific lymphocyte proliferative response of peripheral lymphocyte from patients with CHB was observed by using MTT method and was compared with that by HBV specific iRNA from animal immunized only by HBsAg(a-iRNA).Results Both h-iRNA and a-iRNA increased the level of peripheral lymphocyte proliferative response to HBsAg in patients with CHB to some degree. In group of HBcAg, only h-iRNA showed its enhancement of HBcAg specific lymphocyte proliferative response. Conclusions h-iRNA can increase HBV specific lymphocyte proliferative response in patients with CHB and the function of increasing HBcAg specific lymphocyte proliterative response contributes to HBV clearance .

Objective To construct and identify the incompetent-replication adenovirus carrying the fusiongene composed of the encoding gene of prepropeptide of mouse nerve growth factor(PN)and human beta-endorphin(?-EP)geue.Methods The gene segments of PN obtained from total RNA of the submandibular glandof a 2-week old Kumning mouse were amplified by RT-PCR and joined with the segment of ?-EP to form the fusiongene which was sequenced. The fusion gene contained in the incompetent-replication adenovirus was formed in theBJ-Ad Easy-1 susceptible cells and identified by PCR so as to choose the positive clone without wild vectors.Thecorrect clone was amplified and purified. The titers of adenovirus were determined using the specific 50% tissueculture infection dosage(TCID 50)method.Three days after the adenovirns was transferred into the cultured A431cells,RT-PCR was performed to showed the transcribed mRNA of this fusion gene and the intracellular ?-EPexpression was quanlitatively detected by inummo-histological method. Finally the concentration of human ?-EP inthe culture medium was determined by quantitative radio-immunoassay on 1st,3rd and 7th day afterinfeetion. Results The sequence of the fusion gene was correct. The titer of recombinant adenovirus Ad-NEP was1.5?10~10 pfu/ml.Three days after infection a 475 bp segment was amplified by RT-PCR and abundant orangegranules were shown in the infected cell. The ?-EP concentration in the culture medium was significantly higher inAd-NEP group than in the control group on 1st,3rd and 7th day(P

Objective:To study the expression of coxsackievirus group B3 gene fragment encoding VP1 in procaryon and to explore its application. Methods:Stablie expression of VP1 gene of CVB3 in E.coli was obtained. The expressed protein was purified by NAT chromatography and its immunoactivity was identified by indirect ELISA.Results:The expressed product of VP1,similar to native protein antigen of CVB3,could strong bind with the mouse's antibody serum against CVB3(polyclonal antibody).Irrelevant monoclonal antibody as contrast presents negative activity. Using the expressed VP1 product,we have had a special IgM ELISA for the patient's serum of the clinical acute viral myocarditis. The result was same with the cellular protein antigens of the tissue-cultivated CVB3.Conclusion:The protein antigen CVB3-VP1 which is obtained by the method of gene engineering has character of high product, and its immunoactivity after being purified was basically unchanged. At present, this kind of antigen is difficult to be obtained from the viral cullture medium and a potent hazard for being infected by this virus may take place in such manipulateion. By the method of gene engineering we can obtain antigenic VP1 of CVB3 and use as immuneogen for the detection of serum antibody,by which to provide reliable test reference for the early-stage diagnosis and clinical therapy of the acute myocarditis.

OBJECTIVETo observe the effect of nourishing Yin, strengthening Qi and activating blood decoction on Fas/FasL in salivary glands of NOD mice with Sjogren's syndrome and their mRNA expression.
METHODThirty-two NOD mice were randomly divided into the model group, the traditional Chinese medicine group (TCM group, orally given 0.4 mL nourishing Yin, strengthening Qi and activating blood decoction as per 100 g x kg(-1) everyday), the hydroxychloroquine group (given 0.4 mL hydroxychloroquine as per 60 mg x kg(-1) everyday), the traditional Chinese medicine and western medicine group (TCM WM group, given nourishing Yin, Strengthening Qi and activating blood decoction 50 g x kg(-1) and hydroxychloroquine 60 mg x kg(-1), 0.4 mL everyday), with eight mice in each group. Eight Balb/C mice were selected as the normal control group (normal group). All of mice were killed after eight weeks, and their submaxillary glands were dissected. The expression levels of Fas/FasL were examined by immunohistochemical method, and the FasL mRNA was detected by RT-PCR.
RESULTThe expression levels of Fas/FasL in salivary glands of the model group were higher than that of other groups (P < 0.05). The expression level of FasL of the normal group was much lower than that in the hydroxychloroquine group (P < 0.05). The relative expression level of Fas mRNA in salivary glands of the model group was higher than that in other groups, but the control group was notably lower than other groups (P < 0.05). The expression level of FasL mRNA in salivary glands of the model group was higher than that in TCM and TCM WM groups (P < 0.05). But the expression level in TCM WM group was notably lower than the hydroxychloroquine group (P < 0.05).
CONCLUSIONThe nourishing Yin, strengthening Qi and activating blood decoction could down-regulate the expression level of Fas/FasL in salivary glands of NOD mice with Sjogren's syndrome and their mRNA expression, and had a better efficacy after being combined with hydroxychloroquine. The nourishing Yin, strengthening Qi and activating blood decoction might treat the Sjogren's Syndrome by reducing apoptosis which is regulated by Fas/FasL

Blood-eye barrier (BEB) is one of the most important structures of organism to maintain homeostasis of the eye. However, it is the major constraint for the medication of intraocular diseases. Traditional Chinese medicines have distinctive advantages for the treatment of intraocular diseases, which can be used to regulate the physiological function of human body with low toxicity. In this article, we have briefly summarized the feature of BEB, with the domestic and foreign literatures combined, and mainly reviewed current progress in the field of study on the permeability of traditional Chinese medicines and effective components in BEB and promoting methods.
