1.Detection of the expression of hypoxia-inducible factor 1 alpha in gastric mucosal disease by using tissue chip technique

Objective To study the significance of expression of hypoxia-inducible factor 1 alpha(HIF-1?) in gastric mucosal diseases. Methods Using tissue chip technique to creat tissue chip in 120 cases of vary gastric mucosal disease,and using S-P immunohistochemical methods to detect the expressions of HIF-1? in these tissue chips. Results The positive rates of HIF-1? was 28.3% in 120 tissue chips of gastric mucosal diseases. The positive rates of HIF-1? was 0, 10.0%, 40.0% and 63.3% in chronic superficial gastritis, chronic atrophic gastritis with intestinal metaplasia, chronic atrophic gastritis with dysplasia and intestinal type gastric cancer, respectively. The positive rates of HIF-1? of intestinal type gastric cancer and chronic atrophic gastritis with dysplasia were significantly higher than that in chronic superficial gastritis and chronic atrophic gastritis with intestinal metaplasia(P

3.Expression of p53 and E-cadherin in gastric adenocarcinoma and their relationship with prognosis

Purpose To investigate the protein expression of p53 and E-cadherin in gastric adenocarcinoma and the effects of them and the clinicopathological parameters on prognosis of patients.Methods Expression of p53,E-cadherin in 150 cases of gastric adenocarcinoma was detected by tissue microarray and immunohistochemistry.The effects of them and the clinicopathological parameters on the prognosis of patients were studied by Kaplan-Meier method to calculate and compare the cumulative survival rate and postoperative mean survival time.Multivariate analysis was also performed by COX regression.Results The positive rate of p53 and E-cadherin expression in 150 cases of gastric adenocarcinoma was 31.3% and 91.3%,respectively.74 cases were followed up, and the 1-,3-,and 5-year survival rates were 83.8%,70.3% and 63.5%,respectively.The invasion, lymph node status and pTNM stages were prognostic factors;multivariate analysis revealed that p53,age and lymph node status were independent prognostic invasion, lymph node status and pTNM stages are important prognostic factors in gastric adenocarcinoma. Nevertheless, E-cadherin and histological classification are not prognostic factors.
4.The microsurgical treatment of subdural extramedullary tumors in ventral spinal

Objective To investigate the microsurgical treatment of subdural extramedullary tumors in ventral spinal.Methods Retrospectively analyzed 16 cases of subdural extramedullary tumors in ventral spinal which resected by microsurgical technique from January 2000 to December 2013.By summarized the tumor character,location,extent of resection and rehef of symptoms.Results All of 16 cases were successfully resected,in which 14 cases completely resected,2 cases mostly resected.A higher response rate of local symptoms and neurological symptoms obtained postoperatively,2 patients got worse sensory disorders,movement disorder compared with preoperative,cerebrospinal fluid leakage occured in 1 patient.Conclusion Resection of the subdural extramedullary tumors in ventral spinal with microsurgical technique has the advantage of more clearly anatomical level,minimal tissue injury of spinal cord and surrounding tissue,higher tumor removal rate.

Objective To investigate the different expressions of adiponectin receptor 2(AdipoR2) mRNA in liver between diet-induced obese rats and controls.Methods The level of AdipoR2 mRNA expression was detected by reverse transcriptase polymerase chain reaction(RT-PCR).Insulin sensitivity was evaluated by insulin sensitivity index(ISI).Results Diet-induced obese rats showed higher expression of AdipoR2 mRNA compare to controls.The ISI of diet-induced obese rats was lower than that of controls.Conclusion High energy diet induceds AdipoR2 expression in liver.
9.Interaction relationship between secondary metabolites in Glycyrrhiza uralensis at condition of short-term exogenous glycyrrhizic acid simulation.

In order to study the interaction relationship between secondary metabolites in Glycyrrhiza uralensis, and find out which secondary metabolite is significantly related to the content of glycyrrhizic acid, artificial applying ammonium glycyrrhetate solution was used to establish a high glycyrrhizic acid environment. The change of the 4 secondary metabolites was analyzed within 72 h after glycyrrhizic acid stimulation, while correlation statistical soft was applied to analyze the correlation of glycyrrhizic acid and other compositions. It turned out that it is feasible to establish high glycyrrhizic acid environment by glycyrrhizic acid root soaking in the concentration of 1.0 mmol x L(-1). There was significant positive correlation between glycyrrhizic acid and liquorice glycosides in short-term glycyrrhizic acid stimulation environment. It is concluded that glycyrrhizic acid accumulation internal of G. uralensis could be effected by artificial exogenous glycyrrhizic acid stimulation in certain case, and its accumulation was significantly related to the content of liquorice glycosides.

10.Preparation, identification and application of polyclonal antibody against major outer membrane protein of Chlamydia trachomatis E

Objective To express major outer membrane protein (MOMP) of Chlamydia trachomatis E and to prepare rabbit polyclonal antibody. Methods The recombinant plasmid MOMP/ pGEX6p-l prepared by our lab was introduced into E. coli. The protein was expressed and purified by gel recycling, then was injected into New Zealand rabbits to produce polyclonal antibodies. Enzyme linked immunosorbent assay (ELISA) was used to detect the titer of antibody. The antibody specificity was identified by Western blot and immunofluorescence. Results The fusion recombinant protein glutathione S-transferase (GST)-MOMP was successfully expressed in E. coli. The titer of antibody recombinant protein detected by Western blot and to the endogenous MOMP of Chlamydia trachomatis in vitro detected by immunofluorescence. Conclusions The recombinant MOMP is successfully expressed and the MOMP antibody with high titer and high specificity is obtained. which will be helpful for Chlamydia trachomatis detection and related clinical research.
