2.Analysis on the surveillance results of brucellosis in Aba Prefecture of Sichuan Province during 2000-2007

Objective To clarify the human and animals brucellosis epidemic trends of ABa Prefectufe of Sichuan Province during 2000-2007,to strengthen the prevention and control and determine the strategy for its prevention and cure of brucellosis.Methods According to the National Surveillance of Brucella Document,serology[tube agglutination test(SAT)and plate agglutination test(PAT)]method was used to test the key people,SAT and rose Bengal plate test(RBPF)method to test the yaks and sheep in monitoring sites.Results There were 2 new human brucellosis during 2000-2007.Two thousand and eighty-eight human blood serum were detected.36 people were serohlogie positive,the detection rate was 1.72%(36/2088);the highest rate was in 2003.up to 2.65%(9/339).One hundred and eighty-two thousand,nine hundred and sixty-four cattle and sheep were quarantined.7368 were serologic positive,the detection rate was 4.03%(7368/182 964),4.16%(4844/116 356)for vak and 3.79%(2524/66 608)for sheep;the highest rate being 5.74%(195/3396)in 2001.Conclusions These general trends of human brucellosis is stable,while animals brucellosis has increased in ABa prefecture during 2000-2007.Effective prevention and cure should be adopted to prevent its outbreak.

Microtubule inhibitor has been a hot area of anticancer drugs research.Microtubule inhibitor exert an anti-tumor effect by promoting or inhibiting the microtubule aggregation to break the dynamic balance of microtubule,hindering the spindle forma-tion of tumor cells,and then blocking the process of cell divi-sion.Mitotic catastrophe is a cell death phenomenon that is caused by abnormal cell division and damage of spindle structure in cell mitosis phase.In recent years more and more attention has been paid to mitotic catastrophe cell death because it has
been confirmed clinically that microtubule inhibitors can induce mitotic catastrophe death of tumor cells.This paper reviews the latest research progress of microtubule inhibitors,and discusses the molecular mechanisms of mitotic catastrophe cell death tumor cells induced by microtubule inhibitors.
4.Multiple interventional models combined via anterior tibial veins in treatment of acute deep venous thrombosis

Objective To explore the efficacy of multiple interventional models combined through anterior tibial veins approach in treatment of acute deep venous thrombosis (DVT).Methods The clinical data and the imaging materials of 10 patients with acute DVT were analyzed retrospectively,and the interventional treatment strategies were discussed.Results The percutaneous mechanical thrombectomy (PMT) was performed after a temporary inferior vena cava filter implanted in all patients,and thrombus clearance rate of grade Ⅲ was obtained in 7 patients while 3 patients reached grade Ⅱ criteria.There were 7 patients underwent angioplasty,then the inferior vena cava filter was retrieved during the same procedure in 5 patients while 4 patients in the second stage and 1 patient rejected to retrieve the filter.All swelling limbs alleviated significantly after those procedures.There was no critical complications occured.The total hospital stay ranged from 4 to 7 days,with a mean time of (6.00±0.94) days.All patients were followed up with a mean time of (3.60± 1.84) months (ranged from 1 to 6 months).No thrombus recurrences of the previously affected vessels were reported.Conclusion The multiple interventional models combined via anterior tibial veins for acute DVT is simple,safe and effective,combined with mechanical thrombectomy device can reduce thrombus burden quickly,and combined with angioplasty can consolidate the therapeutic effect,all those methods combined can shorten the overall hospitalization time.
5.Application value of 3.0 T MRI in the preoperative staging and assessment of rectal cancer

Objective Toexploretheapplicationvalueof3.0TMRIinthepreoperativestagingandassessmentof rectal cancer. Methods Data of 103 patients with rectal cancer diagnosed by biopsy was collected. The T staging, situationsofparacoliclymphaticmetastasisandfasciainvolvement,anddistanceoftumoredgetoanalvergewereestimated bypreoperativeMRI.ThestageandassessmentofpreoperativeMRIandpostoperativepathologywerecompared. Results MRIshowedthattherewere5patientswithstageT1-2,72patientswithstageT3,and26patientswithstageT4.And pathologicaldiagnosisshowedthattherewere5patientswithstageT1,16patientswithstageT2,60patientswithstageT3, and22patientswithstageT4.ThesensitivityandspecificityofstageT1-2,T3,andT4were23.8%and100%,96.7%and 67.4%,90.9%and92.6%,respectively.ResultsofMRIwerebroadlyconsonantwiththeresultsofpathology(Kappa=0.748, P<0.01).MRIshowedthattherewere74patientswithparacoliclymphaticmetastasis.Pathologicaldiagnosisaftersurgery showedthattherewere66patientswithparacoliclymphaticmetastasis.ThesensitivityandspecificityforMRIdiagnosis were92.4%and64.9%,respectively(Kappa=0.601,P<0.01).Amongthe18patientsreceivedanalresection,thedifferent valueofdistanceoftumoredgetoanalvergewaslessthanorequalto6mmbetweenMRIandpostoperativespecimen,with nosignificantdifference(P>0.05).Totally65patientswithoutfasciainvolvementdiagnosedbyMRIwereconsonantwith theresultsofpostoperativepathology.Amongthe38patientswithfasciainvolvementdiagnosedbyMRI,12patientswere false-positive.Thesensitivityandspecificitywere100.0%and84.4%,respectively(Kappa=0.732, P=0.000).Conclusion 3.0TMRIcanprovideaccuratepreoperativestagingandevaluationforpatientswithrectalcancerbeforetheoperation,and helpinclinicaldiagnosisandtreatment.
6.Expression of vascular endothelial growth factor in patients with aplastic anemia and its significance.

This study was aimed to investigate the expression of vascular endothelial growth factor (VEGF) in patients with aplastic anemia. The gene expressions of VEGF in mononuclear cells of bone marrow from 7 cases of aplastic anemia and 12 normal controls were detected by semi-quantitative reverse transcriptase polymerase chain reaction (RT-PCR). The expressions of VEGF in bone marrow from 20 cases of aplastic anemia and 20 normal controls were also determined by immunohistochemistry assay. The results showed that expression of VEGF mRNA was found in 2 out of 7 (28.57%) bone marrow of patients and in 10 out of 12 (83.33%) bone marrow of normal controls. The VEGF mRNA in patients with aplastic anemia was significantly lower than that in normal controls (P < 0.05). No patients with aplastic anemia showed immunohistochemical staining of VEGF in bone marrow, while 5 out of 20 (25%) normal controls exhibited VEGF positive cells. Bone marrow of aplastic anemia patients contained less VEGF than that of normal persons (P < 0.05). In conclusion, when compared with normal controls, VEGF expression decreased significantly in patients with aplastic anemia at gene transcription level and protein translation level, it may be related to the defect of angiogenesis and thus hematopoiesis in bone marrow of patients with aplastic anemia.

7.Studies on the aberrant methylation of 14-3-3 sigma gene in human breast cancer

Objective To observe the methylation and expression of 14-3-3 sigma gene in human breast cancer.Methods 40 breast cancer tissues and 18 mammary gland tissues with benign lesions were analyzed by methylation specific PCR(MSP),RT-PCR,and Western-blot(WB)so as to detect the methylation status and expression of 14-3-3 sigma mRNA or protein.Results Methylation of 14-3-3 sigma gene was detectable in 85%(34/40)of patients with breast cancers.RT-PCR showed negative in 12.5% of breast cancers(5/40),WB also indicated that 14-3-3 sigma was not detected in 32 of 40 breast carcinomas (80%).Furthermore,both RT-PCR and WB were negative in 30 of 34 positive cases by MSP.While methylation of 14-3-3 sigma was not detectable and its expression was demonstrated by RT-PCR and WB among 18 cases of benign breast diseases.These evidences proposed that methylation of 14-3-3 sigma gene had great relevance with its silence.Conclusion Methylation and loss of expression in 14-3-3 sigma gene were high frequent events in breast cancers.And methylation of 14-3-3 sigma gene might be related to its loss.
8.MiR-182-5p Knockdown Targeting PTEN Inhibits Cell Proliferation and Invasion of Breast Cancer Cells

PURPOSE: Breast cancer (BC) is one of the most common malignant tumors, affecting a significant number of women worldwide. MicroRNAs (miRNAs) have been reported to play important roles in tumorigenesis. The aim of this study was to determine the roles of miR-182-5p in BC progression. MATERIALS AND METHODS: The expressions of miR-182-5p and phosphatase and tensin homolog deleted on chromosome 10 (PTEN) were measured in BC tissues and cells by quantitative real-time polymerase chain reaction or Western blot. Cell proliferation and invasion were detected by cell counting kit-8 assay and trans-well assay, respectively. The interaction between miR-182-5p and PTEN was probed by bioinformatics analysis, luciferase activity, and RNA immunoprecipitation. A murine xenograft model was established to investigate the role of miR-182-5p in BC progression in vivo. RESULTS: An abundance of miR-182-5p was noted in BC tissues and cells. High expression of miR-182-5p was associated with poor survival. Abrogation of miR-182-5p inhibited cell proliferation and invasion in BC cells. Interestingly, PTEN was indicated as a target of miR-182-5p, and its restoration reversed miR-182-5p-mediated promotion of proliferation and invasion of BC cells. Moreover, depletion of miR-182-5p suppressed tumor growth via up-regulating PTEN expression in the murine xenograft model. CONCLUSION: MiR-182-5p exhaustion blocked cell proliferation and invasion by regulating PTEN expression, providing a novel therapeutic avenue for treatment of BC.

Objective To explore the expression of cardiotrophin-1(CT-1) in myocardium and peripheral blood plasma of neonatal rat with asphyxia and the regulative effect of neuregulin-1(NRG-1).Methods Ninety seven-day-old neonatal rats were randomly divided into 3 groups:asphyxia group (n=40),normal control group (n=10)and NRG-1 pretreatment group (n=40).The model of neonatal rat with asphyxia was prepared by the way of ligation of carotid combined with low supply of oxygen.NRG-1(1 mg/kg) was given to NRG-1 pretreatment group by intraperitoneal injection 30 min before asphyxia.The separated plasma of peripheral blood and myocardium antetheca of aortic ventricle of heart were taken at the time point of 6,12,24 and 48 h.The expression of CT-1 in peripheral blood plasma was detected by enzyme linked immunoadsorbent assay,and that of myocardium was determined by Western blot.Results The expressions of CT-1 protein in peripheral blood plasma of asphyxia group were significantly higher than those of normal control group at each time point,and reached the peak at 24 h(Pa

OBJECTIVETo establish a new method on stratification analysis when the stratification limits of confounding factors was not clear or contradictory.
METHODData on a study of diabetes mellitus in Guangdong province collected in the year of 1997 and 1998 was analyzed using cluster-stratification analysis.
RESULTSThe efficiency of stratification analysis was improved and the confounding bias was effectively controlled with information bias avoided when the clusters-stratification analysis was applied.
CONCLUSIONThe problem was logically solved using cluster analysis as an assistant stratification means.
