2. Effect of lactuside B on the expression of CHOP mRNA in rats' cerebral cortex and hippocampus after cerebral ischemia-reperfusion injury
Chinese Pharmaceutical Journal 2013;48(1):39-43
OBJECTIVE: To investigate the effect of lactuside B on CHOP mRNA expression in rat's cerebral cortex and hippocampus after cerebral ischemia injury. METHODS: Male SD rats of 280-320 g were randomly given brain ischemia-reperfusion treatment for 24 and 72 h respectively and then divided into sham operation group, model group, positive control group, and lactuside B 12.5 mg · kg-1 group, 25 mg · kg-1 group and 50 mg · kg-1 group. There were a total of 12 groups with 4 rats in each group. The cerebral ischemia-reperfusion model was established by cerebral artery occlusion for 2 h. The animals with neurological missing symptom scores of 1-3 were selected for the experiment. All the animals were intraperitoneally injected corresponding experimental drugs after reperfusion for 24 and 72 h and then sacrificed. RT-PCR was used to detected the expression of CHOP mRNA in the cerebral and hippocampus at different time. RESULTS: After cerebral ischemia-reperfusion injury for 24 and 72 h, the neurological deficits scores of the model group animals were significantly higher compared with the sham group (P < 0.01); the neurological deficit scores of the animals were decreased for positive control group and all lactuside B groups compared with model group (P < 0.01). All of the three dosage(12.5, 25 and 50 mg · kg-1) of lactuside B could reduce CHOP mRNA expression in cerebral cortex and hippocampus compared with the model group (P < 0.05, P < 0.01), and the majority of lactuside B groups were superior to the positive control drug nimodip-ine(P < 0.01). There was a good dose-effect relationship for lactuside B in general, but only the middle dose group showed good time-effect relationship. CONCLUSION: The results suggest that the anti-cerebral ischemia effect of lactuside B may be related with the down-regulation of CHOP mRNA expression in cortex and hippocampus. Lactuside B is worthy of development. Copyright 2013 by the Chinese Pharmaceutical Association.

Objective To describe the anxiety of caregivers and condition management of preschool asthma children ,and analyze the relation?ship between anxiety and disease management. Methods A total of 208 caregivers of asthma children from two hospitals in Shanghai were select?ed using convenient sampling, and surveyed by self?rating anxiety scale(SAS),condition management ability scale and condition management diffi?culty scale. Results The score of SAS were negatively correlated with the condition management ability and positively correlated with the condi?tion management difficulty. Hierarchical regression analysis showed that the model explained 26.6%amount and 35.1%amount of variation respec?tively. Conclusion The anxiety of caregiver of children with asthma may affect the disease management. Future interventions should be strength?ened on the caregivers'psychological problems,so as to improve disease management and control asthma more effectively.

Objective To observe the clinical efficacy of centro-square needling at Shangxing (GV23) (so called Ghost Hall point) in treating anxiety after cerebral stroke as a major treatment.Method Sixty patients were randomized into a treatment group and a control group. The two groups were both given Flupentixol and Melitracen tablets (Deanxit). In addition, the treatment group received centro-square needling as the major treatment, and the control group received ordinary needling. Hamilton Anxiety Scale (HAMA), Zung's Self-rating Anxiety Scale (SAS), and Barthel Index (BI) of Activities of Daily Living (ADL) were observed before and after the treatment for evaluating the therapeutic efficacy.Result There was a significant difference in comparing the HAMA score between the treatment group and control group after the treatment (P<0.05). There were no significant differences in comparing the total effective rate and recovery rate between the two groups (P>0.05).Conclusion The treatment group shows a significant efficacy in improving the anxiety symptoms; the effect in improving the anxiety symptoms is insignificant in the control group; the recoveryrate and total effective rate are equivalent between the two groups.

OBJECTIVETo study the blood enriching effects of Paeoniae Radix Rubra and Paeoniae Radix Alba, paeoniflorin and albiflorin on mouse model of blood deficiency caused by γ-ray radiation.
METHODBuild mouse model of blood deficiency induced by γ-ray radiation. Paeoniae Radix Rubra and Paeoniae Radix Alba were given during modeling. The amount of WBC was detected af- ter the treatment. Based on the result of WBC and paeoniflorin content, albiflorin content in Paeoniae Radix Rubra and Paeoniae Radix Alba, the same model and the same method were used to comparatively study the effect of blood enriching of paeoniflorin and albiflorin.
RESULTOn the 7th day, the amount of WBC in model mice treated with 2 g x kg(-1) Paeoniae Radix Alba and 2 g x kg(-1) Paeoniae Radix Rubra significantly increased compared with that of model group (P < 0.05). In another experiment with the same model, the amount of WBC in model mice treated with 120 mg x kg(-1) paeoflorin and 120 mg x kg(-1) albiflorin significantly increased (P < 0.05) compared with that of model group on the 7th day. On the 10th day, the amount of WBC in rats treated with 120 mg x kg(-1) paeoflorin increased significantly (P < 0.05) compared with that of model group. Compared with the same dose of paeoniflorin, the amount of WBC in mice treated with albiflorin had no significant difference.
CONCLUSIONAll Paeoniae Radix Alba, Paeoniae Radix Rubra, paeoniflorin and al- biflorin can raise the amount of WBC and have the effect of enriching blood induced by radiation, while paeoniflorin and albiflorin have a similar result in this model. The result indicated that both paeoniflorin and albiflorin are effective constituents in Paeoniae Radix Alba, and paeoniflorin work as the common effective constituent in both Paeoniae Radix Rubra and Paeoniae Radix Alba.
