2.Study on the determination of total triterpene glycosides in sea cucumbers

Objective A method was developed for determination of the total triterpene glycosides in different sea cucumbers.Methods A holostane triterpene glycoside called Echinoside A was taken as the reference standard.Triterpene glycosides were reacted with vanillin and perchloric acid.The sea cucumbers were extracted by 60% ethanol and partitioned between water and n-butanol to gain the total triterpene glycosides.The contents of total triterpene glycosides in 11 kinds of sea cucumbers were determined with the standard curve.And the relationships of the triterpene glycosides content among different sea cucumber species,growth environment,process technique,etc.were discussed by the value of TG/P(triterpene glycosides content /protein content).Results The determination wavelength was confirmed to be 560nm and the standard curve was determined as y-1.3414x-0.0077.The ab-sorbance was of a good linearity in the mass range of 0-0.5mg with R2 =0.9994.The recovery of this method was(99.01?2.82)% and the relative standard deviation was 4.68%.Conclusion The method has been proved to be convenient,reliable and suitable for the analysis of triterpene glycosides in sea cucumbers.
3.Analysis of patients with spinal cord injurythe following lower limb vein thrombus

ObjectiveTo study the features of lower limb vein thrombus patients after spinal cord injury and the effects of thrombolysis therapy.MethodsWe practiced ultrasonography on 207 patients with lower limb swelling after spinal cord injury. ResultsAmong the 207 patients, 66 had thrombus, the positive rate was 31%: acute thrombus 58 cases, subacute thrombus 1 cases and chronic thrombus 7 cases. Conclusions Ultrasonography provides an important basis for the qualitative diagnoses of lower limb vein thrombus. It has certain clinical guiding effects in choosing methods of treatment.
4.Transcriptome-based bioinformatics analysis of Arnebia euchroma ERF transcription factor family.

Twenty-seven ERF transcription factor family genes were isolated from Arnebia euchroma, with an average size of 1,010 bp, each gene encoded a 212 amino acids on average. The gene structure and expression of physicochemical properties, subcellular localization, signal peptides, senior structural domains and conservative forecasting, and analysis of A. euchroma were studied comparing with ERF gene gi261363612 of Lithospermum erythrorhizon, and phylogenetic analysis of A. euchroma ERF family was carried out. The results showed the existence of three conserved domains in this family, the senior structure based on random coil and it clustered into CBF/DREB and ERF subfamilies.

5.Effects of seed priming on physiology of seed germination and seeding growth of Marsdenia tenacissima under NaCl stress.

To offer the reference and method for salt damage in the cultivation of Marsdenia tenacissima, the seeds of M. tenacissima collected from Maguan city ( Yunnan province) were taken as the test materials to study the effects of different priming materials on improving germination and growth under high-level salt stress condition. Four different treatments, which were GA3, KNO3-KH2PO4, PEG-6000, NaCl, combined with ANOVA were applied to test the performance of germination energy, germination percentage, germination index, MDA, SOD, and CAT. The results showed that the seed germination was obviously inhibited under salt stress and the soaked seeds with different priming materials could alleviate the damage of salt stress. Under these treatments, the activities of SOD, CAT the content of soluble protein significantly increased. While the content of MDA significantly decreased. The maximum index was obtained when treated with 1.20% KNO3-KH2PO4, the germination percentage increased from 52.67% to 87.33% and the activity of SOD increased from 138.01 to 219.44 respectively. Comparing with the treatment of 1.20% KNO3-KH2PO4, the germination percentage of treating with 300 mg x L(-1) GA3 increased from 52.67% to 80.67%, while the activity of SOD increased from 138.01 to 444.61.

6.FTIR fingerprint spectrograms of traditional Chinese medicine Marsdenia tenacissima.

In this paper, Fourier transform infrared spectroscopy fingerprint analysis of Marsdenia tenacissima samples was used to develop a reliable method of tracing the geographical origins. Forty-eight samples from four provinces of China were analyzed by FTIR. We analyzed and characterized the fingerprints in both the full spectrum peaks and characteristic peaks, then the principal component analysis and the cluster analysis were carried out. The results of fingerprint analysis, correlation analysis, principal component analysis and cluster analysis can identify the geographic origins correctly, which verified and supplemented each other; the identification results and the actual location showed a high degree of consistency, namely the lower the space distance, the greater the similarity of different samples. These results revealed the obvious superiority and practical value in comparison to the more tedious and time-consuming wet chemistry method normally used. Using appropriate metrology methods can trace the geographical source correctly. The M. tenacissima materials from the region of Maguan should be considered as genuine medicinal materials taking into account the good quality.

7.NMR-based analysis of water soluble extracts of different Astragali Radix.

Water soluble extract (WSE) is an important index for the quality evaluation of Astragali Radix (AR). In this study, the WSE of the wild AR from Shanxi province (SX) and the cultivated AR from Gansu Province (GS) were compared. The WSEs of two types of AR were determined according to the appendix of Chinese pharmacopoeia. Then the WSEs were subjected to NMR analysis, and the obtained data were analyzed using HCA, PCA, OPLS-DA, microarray analysis, and Spearman rank analysis. In addition, the Pearson correlation of differential metabolites were also calculated. The results showed that the WSE content of GS-AR (37.80%) was higher than that of SX-AR (32.13%). The main constituent of WSE was sucrose, and other 18 compounds, including amino acids, organic acids, were also detected. Multivariate analysis revealed that SX-AR contained more choline, succinic acid, citric acid, glutamate, taurine and aspartate, while GS samples contained more sucrose, arginine and fumaric acid. In addition, the Pearson correlations between different metabolites of the two types of AR also showed apparent differences. The results suggested that the WSE of two types of AR differs not only in the content, but also in the chemical compositions. Thus, the cultivation way is important to the quality of AR. This study supplied a new method for the comparison of extract of herbal drugs.

8.Some Teaching Experience by Using the Mode of Learning Followed by Teaching

A new teaching method was developed in the curriculum of Harmful Microorganisms Control Technology.It is characterized by students’ self-learning followed by student’s instruction.Both students and teacher have succeeded in this model after four stages of practice,in which a pleasant learning atmosphere was created in the classroom.An effective interaction between teacher and students was achieved.Students are viewed as main objects in the classroom and they are encouraged to ask questions,to formulate their own ideas,or to find things out for themselves.Thus,students’ abilities including presentation,communication,competition,and cooperation were enhanced.By adapting their role to the new teaching method,teachers have also improved their teaching skill and strategies.
9.Blocked MyD88 signaling prevents OB after tracheal transplantation in mice

Objective To explore the role of MyD88 in heterotopic tracheal transplantation in mice and its relationship with histopathological changes.Methods The mouse model of hetemtopictracheal transplantation was used.The mice were divided into three groups:(1) tracheal isograft of C57BL/6 to C57BL/6 mice;(2) tracheal allograft of BALB/c to C57BL/6 mice;(3) tracheal graft of BALB/c to C57BL/6 mice with MyD88 inhibitor treatment.The tracheal grafts were collected at indicated time points.Histological sections were stained with hematoxylin and eosin.The pathological changes were observed and their semi-quantitative measurement was done with Image J software.Results (1) Pathological results showed that the structure of the trachea with MyD88 inhibitor treatment was clear and the loss of epithelial cells was significantly reduced as compared with the positive control group at the time of 7 and 14 days.(2) The results of semi-quantitative measurement showed that luminal occlusion rate of MyD88 inhibitor treatment group was significantly reduced as compared with the positive control group (P<0.01).However,the loss of epithelial cells was not improved 7 days after transplantation.Both of lumen occlusion rate (P<0.05) and epithelial cells loss (P<0.01) in MyD88 inhibitor treatment group were significantly reduced.Conclusion Inhibition of MyD88 molecule could significantly alleviate pathological changes of the transplanted trachea.Both of luminal occlusion rate and loss of epithelial cells were significantly ameliorated.
10.Effect of Atorvastatin on Expression of Peroxisome Proliferator-activated Receptor Beta/delta in AngiotensinⅡ-induced Hypertrophic Myocardial CellsIn Vitro

Objective To explore the effect of atorvastatin on cardiac hypertrophy and to determine the potential mechanism involved. Methods Anin vitro cardiomyocyte hypertrophy from neonatal rats was induced with angiotensinⅡ (AngⅡ) stimulation. Before AngⅡ stimulation, the cultured rat cardiac myocytes were pretreated with atorvastatin at different concentrations (0.1, 1, and 10μmol/L). The following parameters were evaluated: the myocyte surface area,3H-leucine incorporation into myocytes, mRNA expressions of atrial natriuretic peptide, brain natriuretic peptide, matrix metalloproteinase 9, matrix metalloproteinase 2, and interleukin-1β, mRNA and protein expressions of theδ/β peroxisome proliferator-activated receptor (PPAR) subtypes. Results It was shown that atorvastatin could ameliorate AngⅡ-induced neonatal cardiomyocyte hypertrophy in the area of cardiomyocytes,3H-leucine incorporation, and the expression of atrial natriuretic peptide and brain natriuretic peptide markedly. Meanwhile, atorvastatin also inhibited the augmented mRNA level of several cytokines in hypertrophic myocytes. Furthermore, the down-regulated expression of PPAR-δ/β at both the mRNA and protein levels in hypertrophic myocytes could be significantly reversed by atorvastatin treatment. Conclusions Atorvastatin could improve AngⅡ-induced cardiac hypertrophy and inhibit the expression of cytokines. Such effect might be partly achieved through activation of the PPAR-δ/β pathway.
