In order to establish the quality classification criteria of Paeonia suffruticosa seeds, thirty-one batches of P. suffruticosa seeds from different provenances were selected. The seed rooting rate, seed germination rate, seed purity, seed viability, 1,000-seed weight and moisture content were determined and analyzed through SPSS 20.0 software. Seed rooting rate, seed germination rate and seed purity were selected as the main index for classification, while 1,000-seed weight, seed viability and moisture content could be used as important references. The seed quality grading of P. suffruticosa was set as three grades. The seed quality of each grade should meet following requirements: For the first grade seeds, seed rooting rate ≥ 80%, seed germination rate ≥ 80%, seed purity ≥ 90%, seed viability ≥ 80%, 1,000-seed weight ≥ 250 g, moisture content, ≤ 10. For the second grade seeds, seed rooting rate ≥ 50%, seed germination rate ≥ 60%, seed purity ≥ 70%, seed viability ≥ 75%, 1,000-seed weight ≥ 225 g, moisture content ≤ 10. For the third grade seeds, seed rooting rate ≥ 20%, seed germination rate ≥ 45%, seed purity ≥ 60%, seed viability ≥ 45%, 1,000-seed weight ≥ 205 g, moisture content ≤ 10. The quality classification criteria of P. suffruticosa seeds have been initially established.

In order to optimize the testing methods for Paeonia suffruticosa seed quality, and provide basis for establishing seed testing rules and seed quality standard of P. suffruticosa. The seed quality of P. suffruticosa from different producing areas was measured based on the related seed testing regulations. The seed testing methods for quality items of P. suffruticosa was established preliminarily. The samples weight of P. suffruticosa was at least 7 000 g for purity analysis and was at least 700 g for test. The phenotypic observation and size measurement were used for authenticity testing. The 1 000-seed weight was determined by 100-seed method, and the water content was carried out by low temperature drying method (10 hours). After soaking in distilled water for 24 h, the seeds was treated with different temperature stratifications of day and night (25 degrees C/20 degrees C, day/night) in the dark for 60 d. After soaking in the liquor of GA3 300 mg x L(-1) for 24 h, the P. suffruticos seeds were cultured in wet sand at 15 degrees C for 12-60 days for germination testing. Seed viability was tested by TlC method.

OBJECTIVETo investigate drug resistance status in patients with highly active antiretroviral therapy (HAART) in Shandong province.
METHODSA total of 758 patients were separated from the anticoagulatory whole blood during May and October in 2011. The entire protease gene and part of the reverse transcriptase gene were amplified by RT-PCR and nest-PCR in the samples with viral load larger than 1000 copies/ml, then sequenced the gene fragments. Mutation of drug resistant gene and drug susceptibility was analyzed by the online tool HIV db program developed by Stanford University.
RESULTSThe rate of virologic failure in patients was 9.1% (69/758). A total of 53 gene sequences that acquired were used for genotypic resistance analysis. A total of 23 patients were indicated drug resistance with the total of 3.1% (23/742). Drug resistance rates of nucleotide reverse transcriptase inhibitor (NRTI) and non-NRTI(NNRTI) were 2.4% (18/742) and 3.0% (22/742), respectively, and the primary mutation types of drug resistance were M184V and Y181C for NRTI and NNRTI, with no resistance to protease inhibitor (PI). In the 23 patients indicated drug resistance, 78.3% (18/23) were NRTI resistance, 95.7% (22/23) were NNRTI resistance and 73.9% (17/23) dual NRTI and NNRTI resistance.
CONCLUSIONThe presence of drug resistant gene in HIV strains among AIDS patients with HAART in Shandong province was at low level, but mutation diversity was found in drug resistant gene.

Objective To investigate the clinical and pathological features of pediatric CNS atypical teratoid/rhabdoid tumor(AT/RT)and evaluate the role of INI1 antibody on the differential diagnosis and prognosis of AT/RT.Methods The clinical data and pathological features of 12-year-old boy with AT/RT were concluded. The protein expression of INI1 was detected by immunohistochemistry;follow-up was performed and relative documents of that disease were compared;the above data were retrospectively analyzed. Results Mioroanatomy showed typical teratoid cells and primitive mesenchymal cells in the tumor. Both of these 2 tumor components displayed positive reactions of vimentin and CD99.The SMA and EMA positive expressions were noted in the teratoid cells.Immunohistochemical staining demonstrated that INI1 antibody was nuclear negative in tumor cells, but positive within endothelial cells of the blood.Follow-up information showed the patient was dead 2 months after the operation. Conclusions CNS atypical AT/RT is highly aggressive tumor. The appearance of rhabdoid cells is its important morphologic features and its main differential diagnosis evidence.Mesenchymal components and the various results of immunohistochemical staining indicate that AT/RT tumor cells, having the character of the embryonal tumors, have various differentiation potential. The expression of INI1 antibody suggests that a mutation or deletion in the INI1 gene occurs in the tumor, which has important value in diagnosing and differentiating the tumor.

Human xanthine oxidase is considered to be a target for therapy of hyperuricemia. Cichorium intybus is a Chinese plant medicine which widely used in Xinjiang against various diseases. In order to screen the inhibitors of xanthine oxidase from C. intybus and to explore main pharmacological actions of cichory a compound collection of C. intybus was built via consulting related references about chemical research on cichory. The three-dimensional crystal structure of xanthine oxidase (PDB code: 1N5X) from Protein Data Bank was downloaded.. Autodock 4.2 was employed to screen the inhibitors of xanthine oxidase from cichory 70 compounds were found to possess quite low binding free energy comparing with TEI (febuxostat). C. intybus contains constituents possessing potential inhibitive activity against xanthine oxidase. It can explain the main pharmacological actions of cichory which can significantly lower the level of serum uric acid.

Journal of Third Military Medical University 2001;23(4):440-442
Objective To explore the relationship between hemolymph phenol oxidase and the melanization of Plasmodium yoelii oocysts in Anopheles dirus. Methods An Anopheles dirus-Plasmodium yoelii system was used Anopheles dirus were divided into 3 groups, that is, non-blood-fedding (N), normal-blood-fedding (B) and infected-blood-fedding (I). The activities of MPO and o-DPO in hemolymph from 3 groups were determined with native polyacrylamide gel electrophoresis (PAGE) and density scanning at 5, 7, 11 and 15 d after blood feeding. Results Both MPO and o-DPO activity were significantly higher in group I than group N and B (P<0.05). But with the melanization of Plasmodium yoelii oocysts, both MPO and o-DPO activity in group I were decreased in comparison with group N, especially on the 15 th day after infected-blood feeding. MPO and o-DPO activity in group B were significantly stronger than those of group N. Conclusion Blood feeding and infection of Plasmodium yoelii both can activate the cascade. The heamolymph phenol oxidase may play an important role in the melanization of Plasmodium yoelii oocysts in Anopheles dirus.
