2.An Experimental Study on Biocompatibility of Human Hair Keratin Artificial Tendon Material

Objective To study the biocompatibility of human hair keratin artificial tendon(HHKAT) material in vitro. Methods Human embryonic tendon cell (HETC) was incubated with various concentrations (100%,50%,10%) of extracted liquid of HHKAT material for 1,3,5 and 7 days, respectively. Collagen I expression in HETC was detected by immunocytochemistry. The effects of HHKAT material on the proliferation and ultrastructure of HETC were observed. Results Various concentrations of extracted liquid of HHKAT had no effect on the morphologic, collagen I expression and ultrastructure of HETC. HETC could adhere, grow and proliferate on the surface of HHKAT material. Conclusion HHKAT is a medical biomaterial with good biocompatibility in vitro.
3.RELATIONSHIPS BETWEEN THE EXPRESSION OF cNOS mRNA IN GASTRIC CARCINOMAS AND THE INVASION,METASTASIS AND PROGNOSIS

Objective To investigate relationships between the expression of cNOS mRNA and invasion and metastasis and prognosis in primary gastric carcinomas. Methods In situ hybridization was used to determine the expression of cNOS mRNA in 119 gastric cancer and 82 follow-up data were analyzed to obtain survival figures. Results In gastric carcinoma specimens,the expression of cNOS mRNA can be observed in the tumor cells,endothelial cells and mononuclear phagocytes;the poorer the carcinoma differentiation,the higher the positive rate of cNOS mRNA in tumor cells and endothelial cells;the expression of cNOS mRNA in tumor cells and endothelial cells reveals a significant positive correlation to the depth of invasion,lymph node metastasis and TNM stage;the patient with positive expression of cNOS mRNA in endothelial cells has a poor prognosis;the expression of cNOS mRNA in mononuclear phagocytes is not related to histopathologic type,the depth of invasion,lymph node metastasis and TNM stage.Conclusion In gastric carcinoma,the expression of cNOS mRNA in tumor cells and endothelial cells might be realted to potential of invasion,metastasis,and the patient with cNOS mRNA positive expression in endothelial cells would have a poor prognosis.
4.Expression of VCA genes of Epstein-Barr virus in Pichia pastoris and clinical application

Objective To analyze the application of recombinant VCA-BALF4(S) and VCA-BFRF3 proteins in serological diagnosis of nasopharyngeal carcinoma (NPC). Methods DNA extracted from the B95-8 cells was used as the templates in a polymerase chain reaction(PCR). VCA-BALF4 1008 bp (S) (aa 287-623) and VCA-BFRF3 531 bp(177 as) were generated and inserted into pPICZaA vector.The recombinant plasmids were transformed into GS115 yeast by electroporation. The yeast transformants in-duced by methanol expressed recombinant proteins. The recombinant proteins synthesized were coated to mi-croplate for detection of EBV-IgA antibody in NPC patients by ELISA. Results We have successfully se-cretly expressed the recombinant VCA-BALF4(S) and VCA-BFRF3 protein in the Pichia pastoris. The mo-lecular weight of products were approximately 37×10~3 and 18×10~3, respectively. The recombinant proteins VCA-BALF4(S) and VCA-BFRF3 in the culture supematant showed good immunoreactivity with IgA anti-bodies to EBV by Western blot. A novel ELISA was established using Pichia pastoris-expressed VCA-BALF4 (S) and VCA-BFRF3 proteins. Serum samples were collected from patients with NPC and healthy controls and using this ELISA tested. The sensitivity of VCA-BALF4(S) and VCA-BFRF3 tests in the NPC sera were 88.7% and 71.0%, whereas the specificity of normal individuals are 96.4% and 91.8% separately. Con-clusion The recombinant proteins BALF4 (S) and BFRF3 were highly secretly expressed in Pichia pastoris,the diagnostic value of two recombinant proteins in screening for NPC patients were primary evaluated and the valuable pPICZa-BALF4(S) yeast strain was obtained.
5.Association between prodromal headache and propensity of rebleeding after aneurysmal subarachnoid hemorrhage

Objective To investigate the association between prodromal headache and the propensity of rebleeding after aneurysmal subarachnoid hemorrhage.Method Prodromal headache was defined as a sudden attack of severe headache lasting at least one hour with or without accompanying symptoms,not leading to a diagnosis of SAH 4 weeks before the confirmation of subarachnoid hemorrhage.Two hundred and nine patients with aneurymal subarachnoid hemorrhage were enrolled in this research.Age,gender,Hunt-Hess grade,D-dimer, (FBG),site and number of aneurysms and prodromal headache were recorded.Patients were divided into rebleeding group and non-rebleeding group.Comparing the above survey components between two groups was carried out by using case control study method.Age,aneurysm number,D-dimer and fibrinogen of the two groups were analyzed by t test.Prodromal headache,gender,site of aneurysms were analyzed by chi square test.Hunt- Hess grade was analyzed by rank test.Results Twenty-two(10.5%)patients had rebleeding.In the rebleeding group,10 patients(45.5%)had prodromal headache,while in the non-rebleeding group(n=187),21 patients (11.2%)had prodromal headache.Patients with prodromal headache had greater propensity of rebleeding compared with those without prodromal headache(P
6.Determination of β-sitosterol and total sterols content and antioxidant activity of oil in acai (Euterpe oleracea).

In order to establish a method for the determination of the sterols of the oil in the freeze-dried acai (Euterpe oleracea Mart.) and to evaluate its antioxidant activities, a saponification/extraction procedure and high performance liquid chromatography (HPLC) analysis method were developed and validated for the analysis of phytosterols in PEE (Petroleum ether extract). Separation was achieved on a Purosper STAR LP C18 column with a binary, gradient solvent system of acetonitrile and isopropanol. Evaporative light scattering detection (ELSD) was used to quantify β-sitosterol and the total sterols. Peak identification was verified by retention times and spikes with external standards. Standard curves were constructed (r = 0.999 2) to allow for sample quantification. Recovery of the saponification and extraction was demonstrated via analysis of spiked samples. The highest content of total sterols is β-sitosterol. The antioxidant activities of the extracts were evaluated using the total oxyradical scavenging capacity assay (TOSC assay). The result showed that the PEE exhibited significant antioxidant properties, sample concentration and the antioxidant capacity had a certain relevance.

8.EFFECT OF TRANSGENIC MT-GENE MUSHMOOM (PLEUROTUS OSTREATUS) ON ANTI-OXIDATION ABILITY OF IRRADIATED MICE

Objective: To explore the effect of transgenic metallothionein (MT) mushroom on anti-radiation ability of mice. Method: Dried power of transgenic MT mushroom was mixed into the feed of ICR mice. After being fed for 23 d, they were irradiated by 5.0Gy 60Co-?rays. Then their anti-radiation ability was determined after being fed 7 d again. Results: Transgenic MT mushroom can evidently alleviate the injury of platelet and spleen induced by radiation, and apparently increase SOD activity and reduce MDA content in irradiated mice’s serum. Conclusion: Transgenic MT mushroom can improve anti-oxidation ability of irradiated mice.
9.Chemical constituents from the linseed meal.

Ten compounds were isolated from the 70% ethanol extract of linseed meal (Linum usitatissimum L) through a combination of various chromatographic techniques, including silica gel, macroporous adsorbent resin, Sephadex LH-20, and preparative HPLC. On the basis of spectroscopic data analysis, they were elucidated as 1-methylethyl-2-O-beta-D-glucopyranosyl-(1" --> 6')-beta-D-glucopyanoside (1), linustatin (2), neolinustatin (3), lotaustralin (4), linamarin (5), deoxyguanosine (6), deoxyadenosine (7), (+)-pinoresinol-4'-O-beta-D-glucopyranoside (8), 4-O-beta-D-glucopyranosylvanillyl alcohol (9) and tachioside (10), separately. Among them, compound 1 is a new compound, and compounds 6, 8 and 10 were isolated from the linseed meal for the first time.
10.Method study for determination of hesperidin in Citri reticulatae by HPLC

AIM:To establish a quantitative method for determination of hesperidin in Citri reticulatae in order to evaluate their quality. METHODS:Sample was extracted with methanol by means of sonication for 30min. ODS column was used with methanol-water-acetic acid(35∶61∶2)as mobile phase. Detection wavelength was at 284nm. RESULTS:Hesperidin in sample solution was well separated. Linearity was good(r=0.9995)in range of 0.202~2.424?g. The average recovery was 98%, RSD of repeatability was 2.3%. CONCLUSION:This method is simple, accurate and reliable, which may well be used for the determination of hesperidin.
