3.Research progress of exosome delivery vehicles in tumor phototherapy

Exosome is a self-secreted phospholipid bilayer nanovesicles, and has shown great potential in drug delivery field due to the important advantages of low immunogenicity and homologous targeting. Phototherapy, mainly includes photodynamic therapy (PDT) and photothermal therapy (PTT), utilize light to activate photoactive drug for tumor cell killing. The advanced therapeutic strategy shows low toxic side-effect and non-invasion precise advantages, and thus has made great progress in tumor treatment over the past few years. Therefore, using exosomes as a drug delivery system to deliver phototherapeutic agents can improve therapeutic performances with a reduced side-effect, and further enhance their application potential for clinical tumor therapy. This review focus on the rising cross-subjects field involving exosomes and phototherapy, and mainly introduce the research progress and relative case of exosomes-based delivery system for cancer phototherapy. Additionally, the advantages and challenges of exosome-based phototherapy are also discussed and proposed.
4.Ecological agriculture: future of Good Agriculture Practice of Chinese materia medica.

Based on the ecological and economic problems in Good Agriculture Practice (GAP) of Chinese material medica, we introduced the origin, concept, features and operative technology of eco-agriculture worldwide, emphasizing its modes on different biological levels of landscape, ecosystem, community, population, individual and gene in China. And on this basis, we analyzed the background and current situation of eco-agriculture of Chinese materia medica, and proposed its development ideas and key tasks, including: (1) Analysis and planning of the production pattern of Chinese material medica national wide. (2) Typical features extraction of regional agriculture of Chinese materia medica. (3) Investigation of the interaction and its mechanism between typical Chinese materia medica in each region and the micro-ecology of rhizosphere soil. (4) Study on technology of eco-agriculture of Chinese materia medica. (5) Extraction and solidification of eco-agriculture modes of Chinese materia medica. (6) Study on the theory of eco-agriculture of Chinese materia medica. Also we pointed out that GAP and eco-agriculture of Chinese material medica are both different and relative, but they are not contradictory with their own features. It is an irresistible trend to promote eco-agriculture in the GAP of Chinese material medica and coordinate ecological and economic development.

6.The primary study on effects of ultrastructure of the brain of the late trimester fetal rats via transumbilical vein contrast-enhanced ultrasonography

Objective To observe the ultrastructure changes of brain of the late trimester of fetal rats via transumbilical vein injection using contrast-enhanced ultrasonography.Methods The forty pregnant rats in the late trimester were divided into four experimental groups randomly.After being successfully punctured in the umbilical vein,two fetus was randomly chosen from maternal uterus with injection contrast agent SonoVue or normal saline solution(20 fetus/each group).Group A as control group with injection of normal saline solution(120 μl/kg);group B as low dose group with injection of SonoVue 0.014 mg/kg(120μl/kg,0.118 ng/ml);group C as media dose group with injection of SonoVue 0.14 mg/kg(120 μd/kg,1.18mg/ml)and group D as high dose group with injection of SonoVue 1.4 mg/kg(120 μl/kg,11.8 mg/ml).After observing the process of every contrast-enhanced ultrasonographic examination,tissue slices of brain of these fetus were made for electronmicroscope investigation.Results ①There was no enhancement in the fetus and placenta in group A.Enhancement couldn't be clearly seen in group B because of fast wash-out contrast agent.On the contrary,the fetus and placenta enhancement in turn could be seen in group C and group D.Enhancement never be seen in maternal uterus and uterine outside.②There was no significant difference of ultrastructure changes of the fetal brain tissues in group A and group B with electronmicroscope.Different degree rough endoplasmic reticulum enlargement,mitochondria swelling,endothelial cell injury,glycogen deposition and vacuolization in the brain cells were found in group C and group D,and the value of contrast between the two groups was significantly different(P<0.05).Conclusions It is decreasd gradually effects to perform transumbilical vein contrast enhanced ultrasonography with reducing gradually dose for the late trimester of fetal rats,but enhancement has been invisible in the low dose group.
7.Effects of cinnamyl aldehyde on c-Fos and c-Myc expression in NIH3T3 cells

AIM:Cinnamyl aldehyde (CA) is one alcohol ingredient derived from Cinnamomum cassia,which is widely used in treating chronic skin wound in Chinese medicine with the curative effect of ‘rescuing YANG’. The purpose of the present study was to investigate the expression of c-Fos, c-Myc proteins at different time points in NIH3T3 treated with CA and explore the possible mechanism of promoting cell proliferation by CA. METHODS: MTT assay was used for observing cell proliferation. Expression of c-Fos and c-Myc proteins in NIH3T3 cells were assessed by immunocytochemistry assay. RESULTS: The cell proliferation was promoted obviously when CA concentration was between 8.8?10-2 ?g/L and 8.8?10 ?g/L. Objective To evaluate the efficacy and safety of diacerhein in the treatment of patients with knee osteoarthritis.Methods The efficacy and safety of diacerhein was randomly investigated in 42 pa- tients with knee osteoarthritis using parallel group methodology and a double-dummy technique to ensure dou- ble blind status with respect to diacerhein and control drugs diclofenac.Results Significant changes were ob- served in 20 meters walk pain,knee joint tenderness scale,WOMAC index scale,5F-36 health survey,knee joint swelling scale,compared with baseline(P>0.05)in both diacerhein and diclofenac group respectively.No difference was found between diacerhein group and diclofenae group.The patient global assessment and physi- cian's global assessment were similar in diacerhein group and diclofenac group(P>0.05).The side effect was similar in two groups.All of these side effects in gastrointestinal tract appeared to be transient.Conclusion Diacerhein can effectively relieve pain and swelling of knee osteoarthritis,and provides us a new effective and safe approach for treating knee osteoarthritis.
9.Effect of Yangyu Tuji on content of type Ⅰ, Ⅲ collagen and the expression of MMPs and TIMP-1 in wound caused by streptozotocin in rats

AIM: To study the effects of Yangyu Tuji (YYTJ) on delayed healing wound of diabetic rats caused by streptozotocin (STZ). METHODS: SD male rats were randomly divided into control group (control), model group (model); and 3 different dose groups of YYTJ. 55 mg/kg STZ were given by intraperitoneal injection except for control group. After 30 days, a round skin of 1.6 cm diametre was excised on all dorsal back of rats. The healing time and healing rate were observed according to re-epithelization. The content of collagenⅠ and Ⅲ was observed by Picric acid-Sirius red staining , Matrix metalloproteinase-1, 13 (MMP-1, -13), tissue inhibitor of metalloproteinases-1 (TIMP-1) by immuno-histochemistry assay. Neuritin is a new neurotrophic factor found recently. In order to identify the function of Neuritin clearly, the coding sequence of human neuritin was amplified by PCR from neuritin cDNA , this fragment digested by NocI and NotI was inserted into pET32a by T4 ligase and transformed into E. coli BL21 then the recombinant plasmid named pET32a-neuritin was constructed successfully . Neuritin was expressed distinctly after inducing by EPTG. The product was identified as neuritin by SDS-PAGE and Western blot analysis . The expression production was purified on Ni2+-NTA column.
