Objective To observe the preventive effects of Tripterygium hypoglaucum(Level)Hutch(THH)extract on T lymphocyte subpopulations in mice with graft-versus-host disease(GVHD)and to explore its protective mechanism.Methods 2?107 bone marrow cells mixed with 2?107 spleen cells from C57BL/6 mouse were transplanted into the myeloablative irradiated inbred BALB/c mouse.The experiment groups were designed as follows:model control group,cyclosporine A(CsA,10 mg?kg-1?d-1)group,THH(400 mg?kg-1?d-1)group,combination group(CsA 5 mg?kg-1?d-1 + THH 100 mg?kg-1?d-1).The ocurrence of GVHD was assessed by signs of weight loss,diarrhea,ruffled fur,hunched posture and histologic changes of skin,liver and small intestines.Chimerism was detected by monitoring the H-2b molecule in bone marrow cells of recipient mice with flow cytometer.The percentages of CD+3CD+4 and CD+3CD+8 T cells in peripheral blood were detected by flow cytometer.Results The survival time of CsA group,THH group and combination group were prolonged as compared with that of the model control group(P

4.Preliminary application of recombinant protein Sj-Ts4 in immunodiagnosis of Schistosomiasis japonica

Objective To evaluate the diagnostic value of the recombinant protein Sj_Ts4 in immunodiagnosis of Schistosomiasis japonica.Methods Seventy-four blood samples of schistosomiasis japonica patients(acute, chronic and advanced)were used for evaluating the sensitivity.Blood samples from 24 Clonorchiasis patients,8 patients with hookworm infections and 30 normal persons from the areas without Schistosomiasis were used ror patients.Results The positivity rates were 97.1%(33/34),100.0%(16/16),87.5%(21/24)in rSj-Ts4-ELISA and 100%(34/34),100.0%(16/16),75.0%(18/24)in SjAWA-ELISA in acute,chmnic and advanced Schistosomiasis. respectively.Statistical analysis revealed no significant difference in sensitivity(X2=1.23,P>0.05)between both recombinant and crude antigens.The false positive reaction was found to be 6.7%(2/30)in rSj-Ts4-ELISA and 3.3%(1/30)in SjAWA-ELISA when detected in 30 cases of normal control sera.but no statisticallv significant difference was noted(x2=0.35,P>0.05).Twelve point five percent(3/24),20.8%(5/24)and 12.5%(1/8),37.5% (3/8),of cross-reactions were observed between rSj-Ts4-ELISA and SjAWA-ELISA for detecting the sera of patients with clonorehiasis and hookworms.There was no significant difference of cross-reaction in two parasitic infections (x2=0.60,1.33,P>0.05)with the two tests.Conclusions The rSj-Ts4 antigen shows higher sensitivity and specificity for the diagnosis of Schistosomiasis japonica,which is helpful in the serological diagnosis of Schistosomiasis japonica in endemic areas.
5.Protein disulfide isomerase’s role in cardiovascular disease

Protein disulfide isomerase (PDI) is one of thiol-disulfide oxidoreductases that mainly located in the endo?plasmic reticulum (ER). It is generally known that PDI caralyzes the formation,rearrangement,breakage of disulfide bonds, and this enzyme is effective in regulation of protein folding. Now it is also known as a biomarker of cardiovascular disease. Protein disulfide isomerase can reduce infarct size and myocardial apoptosis in acute myocardial infarction (AMI). PDI can also improve changes of cardiac vulnerability in diabetic cardiomyopathy (DCM). Further more, it is also shown that PDI play an important role in hypertension and thrombosis. Therefore, this paper review the effects of protein disulfide isomerase in cardiovascular diseases.
6.Influence of four kinds of thread design on the primary stability of immediately loaded dental implants

Objective To discuss the effects of implants of different thread face angles on the primary stability of immediately loaded implant using the 3-dimensional finite element analysis with the models of the immediately loaded implants. Methods Using the commercial code of Pro/E software, Hypermesh software, and ABAQUS software we created 3-dimensional finite element models. The micromotions of the finite element models with different screw face angles (V-shape, buttress, square-shape and inverse buttress) were computed with the ABAQUS software. Results Concerning different thread face angles, the micromotion of buttress implant was the minimum and the micromotion of inverse buttress implant was the maximum with vertically loading; the micromotion of inverse buttress implant was the minimum and the micromotion of buttress implant was the maximum with horizontal loading. Conclusions Different screw-types have great influence on vertical interfacial micromotions but little influence on horizontal interfacial micromotions. There are two angles which are formed by top /bottom edge and the implants. The larger are the angles, the smaller are the vertical interfacial micromotions, but the weaker of the strength. Thus in designing the screw-type implants, we should consider the angles of thread faces and the strength.
7.Analysis of surveillance results on human brucellosis in 2009 and 2010 in Gansu province

Objective Through analyzing the epidemic characteristics and laws of human brucellosis in Gansu province during the past two years,to provide the basis for control of the disease. Methods Using China Information System for Diseases Control and Prevention V2.0 and survey data of human brucellosis outbreak,we calculated the incidence of brucellosis and the composition of new cases and chronic cases in 2009 and 2010, respectively, and analyzed the three distributions of the disease. Results Brucellosis incidence was 0.37 per million in 2009, and 1.7 per million in 2010. The new cases accounted for 54.6%(83/152) and 51.8%(43/83),and chronic cases were 53.3% (81/152) and 56.6% (47/83), respectively. About 1/3 of the counties (cities,districts) was found to be with the disease, and most cases were clustered in Hexi Corridor and Longdong area of Gansu province. There were three counties with outbreak. Time distribution of the disease was jagged. May, July,September and December were high, with September the highest. Sex ratio was 3 : 1, and mean age was 46 years old.67.7% (159/235) of cases were farmers, followed by retired officers and herders, with a percentage of about 7%,respectively. Conclusions Epidemic of human brucellosis shows a rapidly rising trend in Gansu province. There are new patients throughout the year, and young farmers are the main victims. A high proportion of chronic brucellosis is more harmful. Strengthen the prevention and control of the disease to increase their brucellosis protective awareness.
8.Immortalization of porcine hepatocyte lines by transfection with simian virus 40 large T antigen and human telomerase reverse transcriptase

Objective To establish normally immortalized porcine hepatocyte lines by ectopic expression of simian virus 40 large T (SV40LT) antigen and the human telomerase reverse transcriptase(hTERT). Methods Primary porcine Hepatoeyte cells were transfeeted with recombinant retrovirus containing SV40LT or hTERT respectively. Subsequently drug resistant cell clones were screened and expanded for further studies. Immortalized porcine hepatocyte was confirmed by examination. Results The morphological phenotype of the transfected cells was similar to the primary porcine hepatocyte. One clone, HepLP, has been maintained in cultue for half year, and expanded by more than 60 passages. SV40 LT and hTERT could be detected in transfected porcine hepatocyte. Pig albumin mRNA was also detected by RT-PCR. No tumor formation occurred when HepLP cells were injected into Balb/c nude mice. Conclusions The immortalized, nontumorigenic, porcine hepatoeytes maintained the properties of porcine primary hepatocytes such as the albumin secretion. This generation of immortalized porcine hepatocyte may be helpful for bioartifical liver support system, hepatocytes transplantation, drug/toxicological studies, and liver biologic studies.
9.Phylogeny relationship and molecular identification of ten Huperzia species (Huperziaceae) based on matK gene sequences.

OBJECTIVETo study the phylogeny relationship and molecular identification of 10 species from Huperzia (Huperziaceae) based on matK gene sequences data.
METHODTotal DNA of nine species from Huperzia was extracted; matK gene sequence was amplified by PCR. PCR product was directly sequenced after purification.
RESULTThe chloroplast matK gene nucleotide sequences from 9 species of Huperzia species were sequenced. The matK gene nucleotide sequences length was 1 589 bp. Analysis with Huperzia lucidula matK gene nucleotide sequences (download from GenBank) and taking Lycopodiella cernua as outgroup, Maximum Parsimony, Neighbor-Joining analyses and genetic distances were conducted using MEGA 3.1 software. 35 variable sites and 35 parsimony informative sites have been found. Pairwise genetic distances among 10 species of Huperzia was 1.59% - 0.25%.
CONCLUSIONThe results were consistent with the taxonomy in morphological of Huperzia. But H. longipetiolata and H. serrata were resolved into in different clade. There are 19 different sites of matK gene sequences between H. longipetiolata and H. serrata, the genetic distances is 0.121%. It is suggested H. longipetiolata should be as an independent species.
