Chinese Journal of Organ Transplantation 2005;0(07):-
Objective To compare the expression of renal transforming growth factor-?_1(TGF-?_1),its receptors type Ⅰ and Ⅱ(T?RⅠ and T?R Ⅱ) in two rat models of chronic nephropathy induced by CsA and FK506 respectively. Methods Rat models of chronic CsA-/FK506-induced nephropathy were established by administering sandimun Neoral and Prograf separately. Then their kidneys were dissected and the protein expression of TGF-?_1,T?RⅠand T?RⅡ,the mRNA levels of T?RⅠ and T?RⅡ was detected by using immunohistochemistry(IHC) and in situ hybridization(ISH) respectively. Results The IHC revealed that the integrated optical densities(IODs) of renal TGF-?_1,T?RⅠ and T?RⅡ were(605.24)?(140.24),(876.28)?(208.73) and(981.59)?(258.65) respectively in CsA-treated rats,(488.37)?(101.76),(586.63)?(131.21) and(543.34)?(105.26) respectively in FK506-treated ones. The ISH indicated that the IODs of renal T?RⅠ and T?RⅡ mRNA were(905.08)?(158.75) and(1090.92)?(242.73) respectively in CsA-treated rats,(661.37)?(205.65) and(716.27)?(195.55) respectively in FK506-treated ones. The difference of the above-mentioned five factors between the two groups was significant(P

Objective To investigate the expression of inducible nitric oxide synthase (iNOS) and p53 protein in hepatocellular carcinoma (HCC) and their relationship with angiogenesis. Methods Immunohistochemical method and image analysis technique were used to detect the expression of iNOS and p53 protein in tumor tissue sections of 59 HCC patients. Microvessel density (MVD) was counted by immunohistochemical staining with anti CD34 antibody. Results ①The expression rates of iNOS and p53 were 81.4%(48/59), 64.4%(38/59) in HCC patients, respectively. The expression intensities of iNOS and p53 were 5 635?1 287, 3 352?873 in HCC patients, respectively. ②MVD was 32.5?2.73 in the tumor tissue of HCC patients. ③The expression of iNOS was correlated with the expression of p53 and MVD in HCC patients ( P

Objective To evaluate the quality of six kinds of radix salvia miltiorrhizae tables and verify the methods of the quality evaluation . Methods Identification, detection and quantitative test method in Chinese Pharmacopoeia Part One 2005 edition and the HPLC-FPC analysis method were used to evaluate the quality of six kinds of radix salvia miltiorrhiza tables. Results The content of tanshinone IIA of six samples is not in accordance with that in the Chinese Pharmacopoeia and the content of lithospermate B of three samples is not in accordance with that in the Chinese Pharmacopoeia. Both the content of lip-soluble component and the water-soluble component of all the samples differ from those of standard radix salvia miltiorrhiza tables. Conclusion The HPLC-FPC analysis method is useful and effective to identify and detect the quality of the HPLC-FPC analysis method.More attention shall be paid to the quality of the radix salvia miltiorrhiza tables and the supervision of the radix salvia miltiorrhizae tables market should be strengthened.
8.Antitumor efficacy of lymphocytes from hepatic porta lymph node stimulated by staphylococcal enterotoxin A
Chinese Journal of General Surgery 2001;0(07):-
ObjectiveTo evaluate the antitumor effects of lymphocytes from hepatic porta lymph node co cultured with staphylococcal enterotoxin A(SEA)?MethodsLymphocytes isolated from lymph node of 5 HCC patients were co cultured with SEA. The proliferation and cytotoxicity of the lymphocytes were observed. Flow cytometer was used to detect the expression of CD3?CD4 and CD8 on the surface of the lymphocytes. TNF ? and IFN ? released into the culture medium were measured by the method of ELISA. The results were compared with that yielded from TILs of the individual patients. ResultsCultured in the presence of SEA, there was a sharp rise of T cells, especially CD8 + cells. Cytotoxic activity was increased. TNF ? and IFN ? were predominantly produced in the first week.ConclusionsLymphocytes from lymph node activated by SEA were armed with an increased antitumor efficacy.

Journal of Medical Postgraduates 2003;0(04):-
Objective: The bacterial superantigen staphylococcal enterotoxin A (SEA) has the potent ability of inducing T cell activation as well as directing activated T cells to kill tumour cells. However, its application in the tumour treatment is limited due to its systemic immune activation. In order to minimize its side effects SEA liposomes is prepared and their toxcicity in vivo is investigated . Methods: SEA liposomes were prepared by the method of reverse-phase evaporation . SEA liposomes were administered intravenously . In vivo the toxcicity of SEA liposomes was investigated by the measurement of blood pressure, colonic temperature and breath rate of New Zealand rabbits. Mice plasma TNF-? and IFN-? level were determined by ELISA . Results:After iv administration of SEA liposomes, significant reduction of mice plasma TNF-? and IFN-? level was observerd. As compared to free SEA, liposomal SEA had less effect on blood pressure, colonic temperature and breath rate of the rabbits. Conclusion:SEA Liposomes had relatively low toxicity. These advantage was probably due to its lower systemic immune activation effect and inducing consequent lower systemic TNF-? and IFN-? level.
