1.Investigation and analysis of preventive antibacterial drugs used in type Ⅰincision operation

Objective To analyze the situation and trend of the application of antibacterial drugs in the case of typeⅠincision operation in our hospital.Methods A total of 1 964 patients in our hospital from 2010 to 2014 were randomly selected,and the changes of antimicrobi-al drugs during the treatment period were analyzed.The surgical site infection probability and antimicrobial application rules in typeⅠincision surgery in 3 years,including probability,category,first time and duration of drug use,and the relationship between antibacterial drugs and typeⅠincision infection were analyzed.Results Application of antibacterial drugs in typeⅠincision operation probability decreased significant-ly,and the surgical infection probability decreased year by year,with statistically significant difference(P<0.05).The first application of an-tibacterial drug time was more reasonable,the choice of antimicrobial drugs were mainly one generation or two generation of Cephalosporin. There was a significant increase in the probability of stopping medication within 2 days after surgery,and the difference was statistically signif-icant(P<0.05).Conclusion The application of antibiotics should be strictly applied to the application of the indications for the use of anti-biotics.To understand the scope of application of antimicrobial drugs,and should try to shorten the use of time,control of infection during peri-operative period,reduce drug resistance.
3.A study on Huoxue Tongmai capsules for regulation of small molecule metabolites in plasma of patients with coronary heart disease and blood stasis syndrome

Objective To observe traditional Chinese medicine (TCM) Huoxue Tongmai capsules (HXTMC) for the regulation of small molecule metabolites in plasma of patients with coronary heart disease and TCM blood stasis syndrome. Methods A prospective study was conducted in which 136 patients with coronary heart disease and blood stasis syndrome treated in Dagang district in Tianjin from January 2009 to December 2013 were selected and randomly divided into two groups:control group (68 cases) and HXTMC group (68 cases). The control group received the conventional oral therapy with nitrates, β receptor blockers, calcium antagonists, enteric coated aspirin, etc, and the HXTMC group received the conventional therapy and additionally oral administration of HXTMC, 4 capsules each time (each capsule 0.25 g), trice a day, 30 days constituting a therapeutic course, 3 courses in total. According to the standard of curative effect evaluation of angina pectoris in coronary heart disease, the clinic total curative effects of angina pectoris and electrocardiogram (ECG) were evaluated. The concentrations of copeptin, tumor necrosis factor-α(TNF-α) and interleukin-10 (IL-10) in the plasma were tested by enzyme-linked immunosorbent assay (ELISA), and the TNF-α/IL-10 ratio was calculated. Results The total clinical effective rates of patients and the ECG in HXTMC group were significantly higher than those of the control group [clinic total effective rates:89.71%(61/68) vs. 80.88%(55/68), ECG total effective rates:57.35%(39/68) vs. 48.53%(33/68), both P<0.05]. Concentration of copeptin and TNF-α/IL-10 ratio in the plasma after treatment of the both groups were significantly lower than those before treatment, and the changes were more obvious in HXTMC group [copeptin (ng/L): 0.22±0.04 vs. 0.31±0.05, TNF-α/IL-10 ratio:0.49±0.11 vs. 0.65±0.09, both P<0.05]. Conclusion Addition of HXTMC to conventional therapy for treatment of patients with coronary heart disease accompanied by blood stasis syndrome can lower the plasma level of copeptin and TNF-α/IL-10 ratio, thus it may improve the clinical curative effects of the patients.

To sum up and analyze the representative point compatibility theory after Inner Canon of Huangdi to explore t he compatibility rules of point in acupuncture prescription.On base of references and predecessor,s experience,combined with clinical practice,compatibility rules of point can be concluded that zhu-ke-fu-ying points was the elementary compatibility,and shu-mu-qi points should be selected according to syndrome differentiation,thereby forming a traditional and orderly prescription which reflected principle-method-recipe-point.
6.Signal mining for adverse drug reactions based on healthcare big data: methodology and applications

This paper presented the conventional methods for signal detection of adverse drug reactions (ADRs) and their applications, the research progress in ADRs signal mining based on healthcare big data, and briefed the methods and uses of ADRs prediction using machine learning technology in the era of healthcare big data.The conclusion was that deep learning, as a fast growing tool in machine learning, will become hotspot of research, expected to help with ADRs signal mining and rational clinical drug use.
7.Effect of high volume mechanical ventilation on radiation-induced lung toxicity in rats

Objective To evaluate the effects of mechanical ventilation on radiation induced lung injuries of apoptosis,acute inflammation,and oxidative stress by establishing a rat mechanical ventilation model and animal model.Methods Totally 40 male Sprague-Dawley(SD) rats were randomly divided into 4 groups with 10 rats in each group:control,radiation alone,high tidal volume ventilation,and high tidal volume ventilation following by radiation.After treatment,the pathological changes in lung tissue were observed,NF-κB activity was detected by electrophoretic mobility shift assay (EMSA),the expression of NF-κB subunit P65 protein level in lung cell nucleus was detected by Western blot,and the apoptosis of lung cells was detected by terminal dexynucleotidyl transferase (TdT)-mediated dUTP nick end labeling (TUNEL) method.The wet to dry weight ratio (W/D) of lung,myeloperoxidase (MPO),malondialdehyde (MDA) and superoxide dismutase (SOD) were detected.In addition,the total protein and white blood cell number in lung lavage fluid were also measured.Results Compared to the control,the acute lung injury (ALI) score,W/D ratio,MPO activity,total protein level,white blood cell number,apoptosis index (AI),lung tissue MDA,NF-κB activity and P65 protein expression were increased significantly (q =0.000 32-0.004 81,P <0.05),while SOD values was decreased significantly (q =0.000 18-0.002 53,P <0.01),in other three groups.Compared with radiation and high tidal volume ventilation group,the above indexes were significantly higher (q =0.004 3-0.022 6,P < 0.05) but the SOD value was significantly lower (q =0.002 9-0.008 3,P < 0.05) than those in the high tidal volume ventilation plus radiation group.Conclusions High tidal volume ventilation delivered to the radiation group produced more transparent ventilator-induced lung injury (VILI) than the high tidal volume ventilation alone induced VILI including permeable pulmonary edema,acute inflammation,oxidative stress and apoptosis in lung.
8.Improvement of Quality Standard of Reduqing Oral Liquid

Objective To improve the quality standard of Reduqing Oral Liquid. Methods TLC was used to identify the featured spots of Glycyrrhizae Radix et Rhizoma, Belamcandae Rhizoma and Scutellariae Radix;HPLC method was usded to detect the contents of Baicalin in Reduqing Oral Liquid. Results TLC could identify the featured spots of Glycyrrhizae Radix et Rhizoma, Belamcandae Rhizoma and Scutellariae Radix; Baicalin was detected in range of 53.60–536.00 μg/mL with good linear relationship (r=0.999 99), the average recovery rate was 99.85%, and RSD was 0.63%(n=6). Conclusion The method is simple, accurate and reliable, which can be applied to the quality control of Reduqing Oral Liquid.
9.Dynamic expression and significance of E-selectin in rat of acute necrotizing pancreatitis induced lung injury

Objective To investigate the expression and significance of E-selectin in lung tissue of rat of acute necrotizing pancreatitis (ANP)-induced lung injury.Methods Rats were randomly divided into control and ANP groups.ANP model was induced by retrograde injection of sodium taurocholate into the biliary and pancreatic duct.The rats were sacrificed at 3,6,12 and 24 h.Serum amylase,albumin,Ca2+ and lung wet/dry (W/D) weight ratio were determined,and pancreas and lung tissues underwent routine pathological examination and histopathological scores were evaluated.The expression of E-selectin mRNA and protein in lung tissue of rat was measured by real-time fluorescence quantitative RT-PCR and immunohistochemistry.Results In ANP group at 6h,the serum level of amylase,histopathological scores of pancreas and lung tissues,lung W/D weight ratio were (3978 ± 476) U/L,8.22 ± 0.63,( 12.31 ± 1.58,5.21 ± 0.20,which were significantly higher than those in control group [ (843 ± 90)U/L,0.22 ± 0.36,0.13 ± 0.34,4.46 ±0.17,P < 0.05 or < 0.01 ],the serum level of albumin was (12.9 ± 1.0)g/L,which was significantly lower than that in control group [ ( 15.6 ± 0.7 ) g/L,P < 0.01 ],the serum level of Ca2 + at 12 h was ( 2.33 ±0.15) mmoL/L,which was significantly lower than that in control group [(2.57 ± 0.23)mmoL/L,P < 0.01 ].E-selectin was located in the cell membrane and cytoplasm of vascular endothelial cells.Expression of E-seleetin mRuNA and protein in the lung tissue at 6h was significantly higher than those in control group (3.51 ±0.45 vs 0.95 ± 0.16,0.174 ± 0.019 vs 0.060 ± 006,P < 0.01 ).Conclusions Pulmonary expression of E-selectin of vascular endothelial cell is significantly up-regulated in a time dependant manner when ANPinduced ALI occurs; and it is involved in leukocyte mural and extravasation,and can promote lung injury.
