1.Role of NF-κB siRNA in MCP-1 levels induced by IL-17 stimulation in cardiac myocytes

Objective:To investigate the effect of NF-κB p65siRNA of IL-17 inducing the expression of MCP-1 in the primary cultured cardiac myocytes.Methods:The cardiac myocytes were isolated from neonatal mice by different adhesion method.NF-κB P65 siRNA was transfected into cardiac myocytes and the rates of transcription and translation of MCP-1 were detected by RT-PCR and enzyme-linked immunosorbent assay( ELISA) ,the rates of transcription and translation of P-P65 and P65 were detected by RT-PCR and Western blot.Results:Compared with negative siRNA group,the expression of the MCP-1 at mRNA and protein levels were increased in negative siRNA +IL-17 group(P<0.05).Compared with negative siRNA group,the expression of the MCP-1 at mRNA and protein levels were decreased in NF-κB P65 siRNA group( P<0.05).Compared with negative siRNA+IL-17 group,the expression of the MCP-1 at mRNA and protein levels were decreased in NF-κB P65 siRNA+IL-17 group(P<0.05).The expression of the NF-κB P65 at mRNA and protein levels in cardiac myocytes were specifically and extensively suppressed by NF-κB P65 siRNA(P<0.05).After stimulated by IL-17,the amount of P-P65 in the cardiac myocytes was significantly increased in time-dependent manner compared with that of black group( P<0.05) , but the levels of P65 changed little ( P>0.05 ) .Conclusion: IL-17 stimulates MCP-1 expression in cardiac myocytes via NF-κB activation,and NF-κB P65 siRNA can effectively inhibit the upregulation of IL-17 on MCP-1.
2.Preliminary development of college students' physical exercise behavior scale

Objective The college and university students' physical exercise behavior scale has been de-veloped. Methods Baaed on the theory of planning behavior,after investigation and analysis,a questionnaire was made to test 1180 students from some colleges and universities in Changjiang Delta area. Its reliability and validity was tested,and confirmatory factor analysis was carried out. Results The scale consists of 4 factors: behavioral intention,subjective criterion,cognitive behavior,and integrative attitude. Its retest reliability was 0.920, correla-tion coefficient of the factors between 0.732 and 0.854 ,and the total scale Cronbach α 0.931. The correlated co-efficients between subseales and total scales range 0.455~0.785 ;the IFI, CFI,and NFI fit indices of the four di-mensions were over 0.9 ;RMSEA 0.042. Conclusion The reliability and validity of the scale are suitable as an effective tool to test university students' sports exercise behavior.
3.Experimental Study on Effect of Salvia Miltiorrhiza and Shengmai Injection in Treating Systemic Inflammatory Response Syndrome

Objective To investigate the effect of Salvia Miltiorrhiza (SM) and Shengmai injection (SI) in treating systemic inflammatory response syndrome (SIRS) and their mechanism. Methods The animal model of SIRS was established by injecting lipopolysaccharide(LPS, 1 mg/kg)intraperitoneally. Forty Wistar rats were randomly divided into four groups: control group, SM group, SI group and combined treatment group (SM+SI group), which were treated with normal saline(5 ml/kg) plus LPS(1 mg/kg), SM(5 ml/kg)plus LPS(1 mg/kg), (SI(5 ml/kg))plus LPS(1 mg/kg), SM(2.5 ml/kg) plus SI(2.5 ml/kg) and LPS(1 mg/kg) respectively. Six rats of each group were sacrificed for sample collection of blood, liver, lung and kidney 8 hours after LPS injection. Blood routine, serum TNF-? and IL-6 were measured. Specimen of organs were fixed in formalin and sent for routine pathological examination. The survival of other 4 rats of each group were observed untill 48 hours after LPS injection. SPSS 10.0 was used in statistical analysis. Results Two rats in control group died 13 hours and 22 hours after LPS injection respectively, the remaining 2 rats in this group and the rats in other 3 groups survived 48 hours after LPS injection. The white blood cell count of control group was significantly higher than that of other groups. The serum TNF-? and IL-6 of control group were significantly more than those of other groups. Pathological damages were found in all groups, and the most severe ones were in control group. SM and SI could decrease the level of serum TNF-? and IL-6 in the process of LPS-stimulated SIRS, down-regulate the severe inflammatory response, attenuate organ damages of the liver, lung and kidney, and increase forty-eihgt-hour survival rate obviously. Conclusion The experiment provides a theoretical base for clinical use of SM and SI in treatment of SIRS.
4.Study on relationship between physical activity and characteristics of exercise behaviour among college students

Objective To explore the relationship between physical activity ( PA) in college students and self?efficacy,and provide guarantee for improving health education in colleges. Methods International physical ac?tivity questionnaire( IPAQ) ,scales of physical behavior,scales of stage of change in physical exercise behavior,and scales of self?efficacy were used to test 504 students from some of colleges in Zhejiang province. Results Signifi?cantly more males (38.52%) than females (27.27%) were in the high activity tertile (P<0.01); the amount of PA per week in males and females were (4608±872.07)MET and(4361±674.03)MET(P<0.01); the scores of males and females were (3.32±0.44) and(3.04±0.36),(P<0.01). The number of males (35.5%) in maintenance period and period of action is greater than that of females (28.9%). Males had higher self?efficacy and more scores of behavior than females (P<0.05) . There were significant characteristics of PA in their self?efficacy,score of be?havior and stages of change( F=18.38, F=15.08, F=34.71, P<0.01),and a significant correlation between PA and self?efficacy,stages of change and physical exercise behavior ( r=0.336, r=0.36, r=0.46, P<0.01).Conclu?sion Self?efficacy may have important effect on the level of PA. Stages of change and scores of behavior may ex?plain difference of PA between males and females.
5.Study of the self life of three kinds of experimental dentin self-etching primers

Objective:To observe the effect of storage period of three experimental dentin self-etching primers on the shear bond strength.Methods:Three experimental primers with 2-hydroxyethyl methacrylate (HEMA)or N-methacryloyl glycine (NMGly)as a hydrophilic monomer and 1 0-methacryloxydecyl dihydrogen phosphate(MDP)or N-methacryloyl-2-aminoethylphosphonic acid (NMEP) as an acid monomer,were designed.Immediately after preparation of the samples of MDP-HEMA,MDP-NMGly and NMEP-NMGly, the primers were stored at 40 ℃for 0,6 and 1 4 weeks.At the end of each storage period,the shear bond strength of resin to dentin(n=1 4)and the pH value of the primers were measured.Results:After storage at 40 ℃ for 1 4 weeks,the shear bond strength of MDP-HEMA decreased from (1 7.61 ±1 .56)MPa to (7.53 ±1 .76)MPa;however the bond strenth of MDP-NMGly and NMEP-NMGly did not decrease;most of the specimens of MDP-HEMA and MDP-NMGly exhibited an interfacial failure of the resin from the dentin sur-face;however NMEP-NMGly exhibited a cohesional failure of the dentin and/or resin.The SEMimages showed a separation of resin and dentin of MDP-HEMA and MDP-NMGly had storaged at 40 ℃for 1 4 weeks;while the interface of NMEP-NMGly did not change.Con-clusion:The NMEP-NMGly primer has noticeably higher hydrolytic stability than the MDP-HEMA and MDP-NMGly primers.
6.Application research of micro-course based on WeChat public platform in emergency training

The article analyzed the problems in Emergency Medicine and illuminated the character-istics of the teaching model based on micro-course , and pointed out that the introduction of the micro course into the emergency training was the inevitable trend of the development of teaching. Combined with the real case of emergency training, the article described the micro teaching structure and implementation plan in detail, namely with the WeChat public number as the platform for teaching, setting up the curricu-lum according to the differences of the training staff, setting up the interactive communication area, taking into account the basic theory and expanding the study, to achieve the repeatability, individuation, interac-tivity and convenience of the teaching.
7.Determination of Berberine Hydrochloride in Xiao A Lotion by HPLC

Objective: To develop a method to determine the content of a berberine hydrochloride in Xiao A Lotion. Methods: HPLC was used to determine the content of berberine hydrochloride in Xiao A Lotion. The separation was performed on Inertsil ODS 3 column with 0.05mol/L NaH 2PO 4 (pH3.0) acetonitrile (70∶30) as a mobile phase and the wavelength of UV detector was 270nm. Results: The Linearity of this method was well with the average recovery of 101.4%. Conclusion: The method is simple, reliable, and sensitive. It also shows good separating degree. It can be used in quality control of Xiao A Lotion.

10.Effect of propofol on autophagy during oxygen-glucose deprivation/restoration in human liver cells

Objective To evaluate the effect of propofol on autophagy during oxygen-glucose deprivation and restoration (OGD/R) in human liver cells.Methods Human hepatic HL-7702 cells at the logarithmic growth phase were seeded into culture plates and randomly divided into 3 groups (n =12 each) using a random number table:control group (group C),OGD/R group,and propofol + OGD/R group (group P+OGD/R).The cells were cultured in normal culture medium in group C.In OGD/R and P+OGD/R groups,the cells were subjected to O2-glucose deprivation for 6 h followed by restoration of O2-glucose supply for 12 h.Propofol with a final concentration of 50 mmol/L was added at 10 min before oxygen-glucose deprivation.The cell viability was detected by MTT assay.The expression of autophagy-related proteins such as microtubule-associated protein light chain 3 (LC3) and Beclin-1 was evaluated by Western blot.Immunofluorescence was used to determine the number and distribution of autophagosomes.Results Compared with group C,the cell viability was significantly decreased,the expression of LC3 and Beclin-1 was significantly up-regulated (P<0.05),and the number of autophagosomes was significantly increased in OGD/R and P+OGD/R groups.Compared with group OGD/R,the cell viability was significantly increased,the expression of LC3 and Beclin-1 was significantly down-regulated (P<0.05),and the number of autophagosomes was significantly decreased in group P+OGD/R.Conclusion The mechanism by which propofol reduces OGD/R injury is probably related to inhibition of autophagy in human liver cells.
