Journal of Regional Anatomy and Operative Surgery 2014;(3):224-228
Objective To observe the effect of estrogen on BBB permeability, occludin expression after ischemia-reperfusion in rats, and to make a further investigation on the role of estrogen in cerebral ischemia. Methods Ovariectomized rats were randomly divided into sham-operated group, model group, and estrogen pretreatment group. 4 h,24 h,3 d after ischemia-reperfusion were selected as different observation points, and changes of edema, occluding expression, and blood-brain barrier permeability of the 3 points in time were observed. BBB ultra-structure electron microscope observation was made at 24 h and 3 d after ischemia-reperfusion. Changes in cerebral edema were measured by brain water content percentage;protein expressions were measured by Western blot;and BBB permeability was measured by Evans blue as-say. Results Compared with the sham group, brain tissue water content and EB content in model group both increased 4 h after ischemia-reperfusion (P<0. 05),and they keep increasing as time goes by and finally reaching the peak (P<0. 01). Compared with the model group at the same time point, brain tissue water content and EB content in estrogen pretreatment group decreased in different degrees (P<0. 05 or P<0. 01),and it was most significant at the time point of 24 h after ischemia-reperfusion (P<0. 01). Compared with model group, through electron microscope observation,ia was displayed that BBB TJ's open was reduced in estrogen pretreatment group, astrocyte foot process and capillary tube edema were lighter, and it was significant at the time point of 3 d after ischemia-reperfusion. Western blot detection of occludin protein expression found that it was lower in the model group compared with the sham group at 4 h after ischemia-reperfusion,but there was no significant difference (P>0. 05). 24 h after ischemia-reperfusion,the occludin expression were weaker than that in the sham surgery group with a significant difference (P<0. 05),and it keep weakening with a significant difference 3 d after ischemia-reperfusion (P<0. 01). In estrogen group, there was no significant difference compared with the model group in terms of occludin expression at 4 h after ischemia-reper-fusion (P>0. 05). In estrogen group, there was significant difference in occludin protein expression at 24 h and 3 d after ischemia-reperfusion compared with model group at the same time point (P<0. 05). Conclusion BBB ultrastructure disclose TJ was broke and vesicles within EC was increased and astrocyte cell foot process was swelling in MCAO model,it might be the vasogenic brain edema pathological basis for MCAO. In MCAO animal model, with ischemia-reperfusion time increasing,TJ protein occludin expression significantly decreased,it suggests that occludin plays an important role in the regulation of TJ permeability. Estrogen increases has a very close relationship with occludin ex-pression,and it may be one of the mechanisms of protecting BBB integrity and lessening cerebral edema.

Cofactor engineering, a vital part of metabolism engineering, changes the redox cofactor regeneration approach. Its main goal is to rebuild the components of metabolic products. The bioconversion of xylose for the production of ethanol is being studied intensively because ethanol is an alternative energy source and a potential liquid fuel. Saccharomyces cerevisiae has been traditionally used in producing ethanol from fermentable sugars but it cannot utilize xylose, only its isomer xylulose. Introduction of the xylose fermentation pathway from Pichia stipitis into S. cerevisiae enables xylose utilization in recombinant S. cerevisiae, but the ethanol yields of xylose fermentation with recombinant S. cerevisiae has been low and large amounts of the byproduct xylitol are produced. The major reason is that the catabolism of xylose with the fungal pathway leads an imbalance of redox cofactor. The process of the catabolism of xylose requires NADPH and NAD~+, both of which have to be regenerated in separated processes. More and more attention has therefore focused on the redox cofactor balance in S. cerevisia. The research progress of cofactor engineering to solve the imbalance of redox cofactor in xylose metabolism recombinant S. cerevisiae was introduced. This included expression of transhydrogenase, increasing the utilization of NADPH, and achieving the anaerobic reoxidation of NADH. Reversing the cofactor specificity of enzymes is another effective way.

Objective To evaluate characteristics of hematopoietic and immunologic reconstitution in mixed cord blood transplantation. Methods Single, 2 human HLA-haploidentical or HLA-mismatched cord blood units were transplanted into sublethally irradiated severe combined immunodeficiency (SCID) mice, respectively. The characteristics of engraftment, multilineage hematopoiesis and immunologic reconstitution among the three guoups were compared. Results Single or 2 cord blood units could engraft SCID mice and reconstitute human hematopoiesis and immunologic functions. No significant differences in engrafting rate were observed. Cord blood cells were apt to differentiate into NK cells and B lymphocytes. One or 2 cord blood units could be implanted in SCID mice simultaneously as shown by HLA-DQB1 gene detection. The cord blood containing more hematopoietic progenitor cells and higher colony forming potentiality was apt to engraft. Moreover, in less HLA disparity group, 2 cord blood units were prone to engraft simultaneously. Conclusions Co-transplantation of two cord blood units can reconstitute hematopoietic and immunologic functions in SCID mice. However, imbalance in multilineage differentiation existed.

[Objective]To investigate the etiology, pathological characteristic and therapeutic effect of spontaneous rupture of the finger extensors.[Method]Twelve patients were included in this research group.Eight cases were diagnosed as spontaneous rupture of the extensor pollicis long(EPL) and were repaired by transfer of the extensor indicis proprius(EIP).Three cases were spontaneous double rupture of the the ring and small finger extensors. The distal stump of the ring finger extensor was woven into the middle finger extensor and the EIP was transferred to reconstruct the extensor digiti quinti. There was one case of spontaneous triple rupture of the the middle, ring finger and small finger extensors.Transplantation of frozen allogenic extensors was utilized.[Result]The finger extensor rupture occurred after either rheumatic arthritis or distal radius fracture. The former pathological examination showed that the tenosynovitis and chronic inflammation of tendons accompanying with localized necrosis was dominant, while tendon fiber rupture played an important role in the latter. All patients achieved 100% of good results and no recurrent rupture occurred.[Conclusion]The inflammation invasion or attrition in the fracture site was a patological basis to cause spontaneous rupture of finger extensors. The EPI transposition was a reliable option to reconstruct the EPL.It is suggested that allogenic tendon transplantation be applied in the case of multiple ruptures of the extensor tendons.

Journal of Environment and Health 1993;0(03):-
ve To understand the effects of fenvalerate on the secretion of sex hormone and humoral im-mune function and to explore their mechanism. Methods 20 female SD rats aged 4 weeks were randomly divided into 2 groups: fenvalerate group treated by peritoneal injection with 1/10 LD50 of fenvalerate and control group treated with disinfected bean oil. The serum specimens were obtained from rats of 2 groups at the 4th week after the peri-toneal injection. The levels of interleukin-6 (IL-6). tumor necrosis factor-? (TNF-?) and immunoglobulin G (IgG) in serum were measured with ELISA. The levels of 17?-estradiol (E2) and testosterone (T) were measured with ra-dioimmune assay. Results The levels of IL-6, TNF-? and IgG in serum specimens of rats were 2.244 ng/ ml, 0.360 ng/ml and 4.928?g/ml for fenvalerate group, 2.805 ng/ml, 0.439 ng/ml and 3.825?g/ml for control group respec-tively. The levels of IL-6 and TNF-? in serum specimens of rats were significantly lower than those of control group (P 0.05. Conclusion Fenvalerate could effect the hu-moral immune system and the levels of sex hormones. Its passible mechanism was that fenvalerate could affect the levels of sex hormones first, and then the whole immune system further.
