1.Preconditioning effect of isoflurane on mitochondria free calcium concentrate and mitochondria permeability transition pore after liver ischemia-reperfusion injury in rat brain

Objective To investigate the protective effect and the mechanism of isoflurane preconditioning on rat brain tissue suffering from rat liver ischemia-reperfusion injury .Methods 75 SD rats were randomly divided into five groups, sham group (S group);ischemia-reperfusion group (I/R group): liver ischemia for 60 min, reperfusion for 120 min;isoflurane preconditioning group ( ISO group ): 60 min before liver I/R, ISO pretreatment for 30 min; CsA +ISO group;CsA ( MPTP specific blocker ) 50 mg/kg intravenous injection , the same as ISO group after 30 min; CsA group:CsA 50 mg/kg intravenous injection .30 min before I/R.The animals were killed 24 h after ischemia and their brain were excised for measurement of mitochondrial permeability transition pore ( MPTP) and calcium content in mitochondria .The measurement of content of S-100βprotein in serum before ischemia and 120min after reperfusion through the application of Elisa kit.Results The mitochondrial Ca2+concentration of I/R group(287.32 ±26.17)was higher than that in S group (198.54 ±21.02) and the ISO group (209.74 ±29.49) ( P <0.05), and Ca2 + concentration of CsA +ISO group (267.31 ±37.52) was higher than the ISO group (P <0.05);there was not statistical significance between I/R and CsA group(288.63 ±23.15)(P <0.05).MPTP were more opened in I/R group(△S:1.73 ±0.24)than that in S group (△S:2.36 ±0.35)and ISO group(△S:2.11 ±0.32)(P <0.05),but MPTP were more opened in CsA +ISO group than that in ISO group (P <0.05).The content of S-100βprotein in serum was significantly higher in I/R group than that in sham and ISO groups (P <0.05),and the content of S-100βprotein in serum was significantly higher in CsA +ISO group than in the ISO groups ( P <0.05 ) .Conclusion The liver ischemia-reperfusion may injury the brain of the rat and isoflurane preconditioning can protect the brain on the rat .The reduce of mitochondria calcium overload and inhibition of MPTP opening are involved in the mechanism .
2.Effect of laparoscopic surgery on the quality of life in patients with middle and low rectal cancer

Objective To study the effect of laparoscopic surgery on the quality of life in patients with middle and low rectal cancer.Methods 108 cases with middle and low rectal cancer in accordance with the number table,were randomly divided into two groups.54 patients in the observation group were operated through laparoscopic surgery and 54 patients in the control group received traditional open surgery.The patients were followed up for 6 months,the patients' quality of life were assesed.Results The operative time,blood loss and length of hospital stay in the observation group were less than the control group (P < 0.01).The lymph node dissection between the two groups had no significant difference(P >0.05).47 patients in the observation group were rectal sphincter preservation,the rate was 87.0%,37 patients in the control group with rectal sphincter preservation,the rate was 68.5 %,the difference between the two groups was significant (x2 =4.92,P < 0.05).The patients'quality of life after surgery in two groups was increased than before surgery,and the observation group increased more significantly than the control group(P < 0.05).Conclusion Laparoscopic surgery can significantly improve the quality of life in patients with middle and low rectal cancer.
3.Application of comprehensive experiment evaluation indicators to the fundamental nursing assessment

Objective To evaluate the comprehensive experiment in fundamental nursing by using the indicator system and analyze the teaching effect.Methods 450 nursing graduates of grade 2011 were divided into 56 groups and comprehensive experiments were graded by using the indicator system in each group.Results The four indexes in level one scored (18.08±1.50),(19.99±2.31),(28.18±3.35),(18.16±1.73) points and accounted for 90% 、80% 、81%、91% respectively.The total points' scorings and proportions were (84.41±5.84) points and 84% respectively.Some deficiencies were found to be improved during evaluation.Conclusions Nursing comprehensive experiment evaluation indicator system can assess comprehensive experiments well,and the related competencies of nursing graduates need to be improved.
4.Introduction and Enlightenment of the Human Subject Protection Program in U.S

5.Effect of miR-200b on intestinal epithelial tight junction via MLCK/P-MLC signaling pathway

Objective To explore the impact and mechanism of miR-200 b on intestinal epithelial tight junction. Methods The negative-lentivirus and human-miR-200 b-lentivirus were employed to infect the Caco-2 cell thus establishing two stable cell lines which were then stimulated by 10 ng/mL human tumor necrosis factor-α(TNF-α) to establish the model of the intestinal epithelial injury. Those Caco-2 cells were divided into NC, NC+TNF-α, 200b, and 200b+TNF-αgroups.The tight junction permeability was detected by transepithelial electrical resistance (TEER) and Fluorescein isothiocyanate-labeled dextran (FITC-dextran). The protein alterations myosin light chain kinase (MLCK)/phosphorylated myosin light chain (P-MLC) pathways were measured by Western blot analysis. Results Compared to NC group, NC+TNF-αgroup had lower TEER, higher FITC-dextran, and up-regulated expressions of MLCK and P-MLC proteins (P0 . 05 ). Compared to NC+TNF-αgroup, 200 b+TNF-αgroup had higher TEER, lower FITC-dextran and down-regulated expressions of MLCK and P-MLC proteins (P<0 . 05 ). Conclusion miR-200 b ameliorated TNF-α-induced intestinal epithelial tight junction disruption via regulation MLCK/P-MLC pathway.
6.Effect of Autophagy Inducer pp242 on Intestinal Barrier Dysfunction Induced by Tumor Necrosis Factor-α

Intestinal barrier dysfunction and autophagy abnormality play important roles in the mechanism of inflammatory bowel disease, however, whether autophagy has effect on intestinal barrier dysfunction has not been reported.Aims: To explore the effect of autophagy inducer pp242 on intestinal barrier dysfunction induced by tumor necrosis factor-α (TNF-α).Methods: Model of intestinal epithelial monolayer barrier was established with Caco-2 cells in Transwell chambers, and then randomly divided into four groups: control group (without any intervention), TNF-α group (10 ng/mL TNF-α), pp242 group (1 μmol/L pp242), TNF-α+pp242 group (10 ng/mL TNF-α+1 μmol/L pp242).The intestinal barrier function was evaluated by transepithelial electrical resistance (TEER) and flux of FITC-dextran.The protein expressions of autophagy related protein LC3B-Ⅱ and p62 were detected by Western blotting.Results: Compared with control group, TEER was significantly decreased (P<0.05), flux of FITC-dextran, protein expressions of LC3B-Ⅱ and p62 were significantly increased in TNF-α group (P<0.05).Compared with TNF-α group, TEER was significantly increased (P<0.05), protein expression of LC3B-Ⅱ was significantly increased (P<0.05) while protein expression of p62 was significantly decreased in TNF-α+pp242 group (P<0.05).Conclusions: Autophagy inducer pp242 relieves TNF-α-induced intestinal epithelial barrier dysfunction via activating autophagy flux.

8.Construction and identification of humerus three-dimensional finite element model in children

BACKGROUND:Mechanical experiment of finite element numerical simulation is the effective method to research the biomechanical structure of human body. OBJECTIVE:To establish the three-dimensional finite element model of a normal 6-year-old child’s humerus. METHODS:CT images of a 6-year-old child volunteer were imported to the Mimics 10.01 software. The threshold segmentation method was used to rebuild the humerus three-dimensional model. The surface optimization treatment and surface patches dicision were performed on the surface of the model with Geomagic Studio 12.0 software. Then the mesh generation was completed in the software TrueGrid. Final y, the material properties were set and the finite element model was completed. The boundary conditions and constrains were exerted to simulate the three-point-bending test of humeurs. After the simulation, the results were outputted. RESULTS AND CONCLUSION:The humerus finite element model included 3 024 nodes and 18 758 nodes-hexahedron elements. The 0.01 m/s and 3 m/s dynamic loads were loaded respectively, then the central humerus fracture occurred and the load-displacement curve was close to the cadaver test results. The simulation results show that the simulation results of children humerus finite element model are close to the cadaver’s test, and the finite element simulation method can simulate the physical properties of the human skeleton very wel .
9.Physicochemical analysis of ceftriaxone-associated urinary stone

Objective To investigate the physicochemical characteristics of urinary stone induced by ceftriaxone.Methods Two children cefriaxone-associated urinary stone samples were received for component analysis in our hospital in April 2012,of which one was from a boy whose clinical data was not available,and the other was from a boy who suffered acute lower abdominal pain and vomiting after treatment with ceftriaxone for 5 d in early April.Ultrasound demonstrated a stone in his right upper ureter.Computed tomography showed right upper ureteral stone,which was radiolucent on plain abdominal radiograph.After a conservative treatment for 3 d,the stone in right upper ureter was spontaneously passed,which was confirmed by the ultrasound and intravenous pyelography.The two received stone samples were analyzed by infrared spectroscopy,scanning electron microscopy and energy disperse spectroscopy for component analysis.Results The compositions of two stone samples were free ceftriaxone and calcium ions combined with a molar ratio of 1:1.Conclusions The ceftriaxone could induce urinary stone in children.This special stone has radiolueent imaging,and it is composed of calcium ceftriaxone salt.
10.Diagnosis and surgical treatment of pulmonary sequestration:A report of 18 cases

[Abstract ] Objective Pulmonary sequestration (PS) is a congenital development malformation of the lungs .This study aimed to explore the diagnosis and surgical treatment of PS . Methods We retrospectively analyzed the clinical data on 18 PS pa-tients (8 males and 10 females, aged 20-68 [42.34 ±14.63] years) treated in our department from September 2009 to September 2014.Medical imaging manifested tumors in 11 cases and cystic lesion in the other 7.All the patients underwent surgical resection . Results The diseased pulmonary lobes were removed for the 17 cases of the intralobar type and local resection was performed for the 1 case of the extralobar type .Fourteen of the cases were confirmed to be PS , 3 misdiagnosed, and 1 case missed at diagnosis .The pa-tients were followed up for 8-18 months after treatment , all recovered without surgery-related complications . Conclusion Pulmona-ry sequestration is a rare congenital malformation of the lungs and the lack of specific clinical manifestations makes it easy to be misdi -agnosed.Surgery is the main strategy for its treatment .
