1.Study of pleural isolated Pseudomonas aeruginosas of the resistance mechanisms to carbapenems

Objective To analyze the antibiotic phenotype of Pseudomonas aeruginosas that were isolated from pleural samples in our hosptial and test relative genes that may cause antibiotic resistance in order to provide information for clinical treatment . Methods BioMerieux Bact/ALERT 3D 120 blood culture system was used to culture pleural samples .Simens MicroScan Walk‐Away 96 was employed for bacterial identification and susceptibility test .Imipenem‐EDTA double disk method was used for screen‐ing MBL‐producing isolates .PCR was used for testing OprD ,VIM and IMP .Results 14 of 38 Pseudomonas aeruginosas isolates showed resistance to Imipenem and 2 were with multi‐resistance .4 isolates produced MBL .Only two were found lack of OprD gene among the 14 isolates ,and IMP and VIM genes were detected in 2 isolates .Conclusion The Pseudomonas aeruginosas from pleural infection show relative higher resistant rate to Imipenem ,and the exsiting mechanisms of Imipenem resistance included MBL (VIM and IM P genes) and lack of OprD .
2.Clinical significance of urinary type Ⅰ collagen cross-linked N-telopeptides in bone metastases neoplasm

Objective To analyze the significance of urinary type Ⅰ collagen cross-linked N-telopeptides (uNTX) for the prognosis and response to therapy of bone metastatic neoplasms.Methods The subjects were divided into 3 groups: A,B and C.The levels of uNTX were measured by enzymatic immunoassay kit,and the activity of serum alkaline phosphatases(AKP) and calcium concentration were measured simultanuously.Results uNTX levels positively correlated with bone metastatic neoplasms,and were markedly high in the patients with extensive bone collagen breakdown.Conclusions uNTX is more sensitive than the activity of serum AKP and plasma calcium concentration for bone metastasis of neoplasms,so it is significant in differentiating and monitoring bone metastatic neoplasms.The measurement of uNTX is an early objective methods for evaluation of prognostic response to therapy of bone metastatic neoplasms.
3.Imbalance of MMP-9/TIMP-1 during the cutaneous "underlying disorder" in diabetic rats

Objective To investigate the change of matrix metalloproteinase-9 (MMP-9) and tissue inhibitor of metalloproteinase-1 (TIMP-1) in the skin of diabetic rats, and to explore the potential role of MMP-9/TIMP-1. Methods Diabetic rats were induced with streptozotocin (STZ). Then all rats were maintained for 6 weeks. Routine pathological examination and immnnohistochemistry were made to reveal the histological and cytological appearances. RT-PCR and Western blotting were used to detect the expression of mRNA and protein of MMP-9 and TIMP-1 in the skin. Results Six weeks after STZ treatment, examination of HE-stained skin sections from normal and diabetic animals revealed that the epidermis and dermis layers were thinner in diabetic rats than those in control rats. The skin of diabetic rats showed features of atrophy such as disorganization of connective tissue fiber bundles and enlarged space between collagen fiber bundles. In contrast, thick bundles of connective tissue were observed in the dermis of normal rat skin. In normal skin, cells had a bipolar, spindle-shaped appearance in the thick collagen bundles, while in the skin of diabetic animals the interstitial cells had a rounded, shrunken and crenated appearance. The relative values of expression of MMP-9 in diabetic group were higher than those in normal group with significant difference, however, the relative values of expression of TIMP-I in diabetic group were lower than those in control group. Conclusion The changes in cutaneous histology and cytology appear earlier than skin wound. These "underlying disorders" may be associated with the imbalance of MMP-9/TIMP-1.
4.Relationship between thyroid hormone level and urine protein quantitation in patients with severe preeclampsia

Objective To study the relationship between thyroid hormone level and 24h urine protein quanti-tation in patients with severe preeclampsia. Methods From August 2015 to October 2017,180 patients with severe preeclampsia who were treated in the Third People's Hospital of Qingdao were selected. The level of thyroid-stimula-ting hormone ( TSH) was detected by chemiluminescence particles immune method. According to the test results,the patients were divided into normal group (n=110,TSH=0. 3-3. 3mU/L) and hypothyroidism group (n=70,TSH>3. 3mU/L). Another 240 healthy pregnant women who admitted in the Third People's Hospital of Qingdao during the same period were selected as control group. The TSH,free triiodothyronine (FT3),free thyroxine (FT4) levels and 24h urine protein quantitation were compared among the three groups. The correlation between the level of thyroid hormone and 24h urine protein quantitation,the results of thyroid autoantibody in patients with severe preeclampsia were analyzed.Results The FT4,FT3 levels in the normal group were (11.92±2.54)pmol/L,(5.22 ±1.25)pmol/L, respectively,which in the hypothyroidism group were (9. 02 ± 1. 09) pmol/L,(3. 92 ± 1. 57) pmol/L,respectively, which were all lower than those in the control group (t=6. 657,16. 758,13. 221,18. 245,all P<0. 05),which in the hypothyroidism group were lower than those in the normal group (t=9. 031,6. 149,all P<0. 05). The TSH,24h urine protein quantitation levels in the normal group were (2. 56 ± 0. 86) mU/L,(66. 51 ± 18. 52) mg,respectively, which in the hypothyroidism group were (5. 87 ± 3. 02) mU/L,(79. 14 ± 12. 58) mg,respectively,which were all higher than those in the control group(t=2. 330,7. 197,16. 417,13. 335,all P<0. 05),which in the hypothyroidism group were higher than those in the normal group (t=10. 883,5. 014,all P<0. 05). There was positive correlation between serum TSH and 24h urine protein quantitation in patients with severe preeclampsia (r=0. 254,P=0. 001). There was negative correlation between FT4 and 24h urine protein quantitation (r= -0. 182,P=0. 025). There was no correlation between serum FT3 and 24h urine protein quantitation (r= -0. 080,P=0. 330). The positive rates of TPO Ab,TG Ab were 12. 86%,18. 57% in hypothyroidism group,there were no statistically significant differences compared with 10. 91%,14. 55% in the normal group (χ2 =0. 158,0. 513;P=0. 691,0. 474). Conclusion There is correlation between thyroid hormone levels and 24h urine protein quantitation in patients with severe preeclampsia. It has great significance for clinical treatment of severe preeclampsia by detecting the level of thyroid hormone.
