1.Therapeatic effects of two different dosage of rosuvastatin on endothelial dysfunction in diabetic rats

Objective To evaluate the effects of two different dosage of rosuvastatin on endothelial dysfunction in diabetic rats. Methods The 24 diabetic rats were randomly divided into three groups (n=8,each): diabetic control group, 20 mg rosuvastatin daily (RV 20 mg group) and 10mg rosuvastatin daily for 8 weeks (RV 10 mg group) and normal control group (SD group). The levels of blood glucose, lipid, nitric oxide(NO) and endothelin-1 (ET-1) were measured before and 8 weeks after treatment. Results The levels of blood glucose were higher in all diabetic rats groups than in SD group before experiment (P<0. 01). Compared with diabetic rats control group, blood glucose was slightly lower in RV 10 mg group and RV 20 mg group at 8 weeks (P>0. 05). The plasma NO level was significantly lower in diabetic rats control group than in SD group (P<0. 05).After 8 weeks, plasma NO levels were significantly higher in RV 20 mg and RV 10 mg groups than in diabetic rats control group (P<0. 01 or P<0. 05). The plasma levels of ET-1 was significantly higher in diabetic rats control group than in SD group (P<0. 01). After 8 weeks, plasma ET-1 levels were significantly lower in RV 20 mg and RV 10 mg group than in diabetic rats control group (P<0. 01).Meanwhile, the plasma lipids were lower in RV 20 mg and RV 10 mg group than in diabetic control group (P<0. 05 or P<0. 01). Conclusions Rosuvastatin can adjust blood lipids and significantly improve endothelial function in diabetic rats by increasing plasma NO level and decreasing plasma ET-1 level.
2.The Changes of Serum Interleukin-37 Levels in Patients With Acute Coronary Syndrome

Objective: To study the serum interleukin-37 (IL-37) level changes in patients with acute coronary syndrome (ACS) and to explore the relationship between IL-37 and coronary atherosclerotic plaque.
Methods: Our research included 3 groups. ACS group, n=60, SAP (stable angina pectoris) group, n=30 and Control group, the subjects with normal coronary artery, n=15. The peripheral serum levels of IL-37 were examined by ELISA and the differences were compared among different groups.
Results: ① The serum levels of IL-37 at admission were as ACS group < SAP group < Control group, P<0.05.②Intervention could transitionally decrease IL-37 level in SAP group. With 4 weeks treatment, IL-37 levels were signiifcantly increased in both ACS group and SAP group than admission time, while they were still lower than Control group, P<0.05.③The serum level of IL-37 at admission was negatively related to IL-18 (r=-0.79, P<0.05), the ratio of IL-18/IL-37 were as ACS group>SAP group>Control group, P<0.05.④In ACS group, IL-37 level was negatively related to GRACE score (r=-0.71, P<0.05), the ratio of IL-18/IL-37 was positively related to GRACE score (r=0.73, P<0.05).⑤The diagnosis of ACS could be basically excluded if the patients with IL-37>77ug/L.
Conclusion: The serum IL-37 might be involved in the inlfammatory process in ACS patients, it could be expected as an index for ACS monitor and evaluation in clinical practice.
3.Effect of home-based early intensive behavioral intervention using VB-MAPP on verbal ability in children with autism spectrum disorder

BackgroundHome-based intervention occupies a prominent place in the treatment of autism spectrum disorder (ASD) in children, whereas previous studies on the effect of home-based early intensive behavioral intervention on verbal ability of children with ASD are somewhat inadequate. ObjectiveTo study the effects of intensive family behavioral of intervention based on Verbal Behavior Milestones Assessment and Placement Program (VB-MAPP) on the language ability of children with ASD, so as to provide references for the development of family intervention strategies for children with ASD. MethodsChildren aged 2 to 3 years old who attended the Children's Rehabilitation Department of the Second Affiliated Hospital of Shantou University Medical College from September 2020 to December 2021 and met the Diagnostic and Statistical Manual of Mental Disorders, fifth edition (DSM-5) diagnostic criteria for ASD were selected as the study objects. A non-randomized concurrent control trial was conducted to compare a study group (n=24) receiving home-based early intensive behavioral intervention using VB-MAPP with a control group (n=14) receiving training from other special institutions or early childhood education institutions. The intervention lasted for 6 months in both groups. Before and after the intervention, the VB-MAPP milestone assessment was performed in the two groups, and the VB-MAPP milestone score, mand, trac and listener responds were selected as the study indicators. Then the intervention effect was compared between two groups, and the multiple linear regression was performed to screen the related influencing factors. ResultsAfter intervention, the total milestone assessment score, mand, tact and listener responds scores of study group were higher than those of control group, with statistical difference (Z=-4.339~-2.195, P<0.05 or 0.01). Multiple linear regression analysis denoted that the average weekly hours of home-based intervention in the first three months had certain effect on listener responds (β=1.029, P<0.05). ConclusionApplication of home-based early intensive behavior intervention using VB-MAPP may contribute to the improvement of verbal abilities such as mand, tact and listener responds in children with ASD. 4.Improvement Effects of Water Extract of Glycyrrhiza uralensis on Acute Hepatic Injury Caused by Triptolide and Its Effects on the Levels of IL- 10 and TNF-α in Rats

OBJECTIVE: To observe the improvement effects of water extract of Glycyrrhiza uralensis on acute hepatic injury caused by triptolide and its effects on the levels of IL-10 and TNF-α in rats. METHODS: A total of 60 healthy male SD rats were randomized into blank control group, model group, positive control group (magnesium isoglycyrrhizinate) and G. uralensis water extract low-dose, middle-dose and high-dose groups. Blank control group and model group were given isochoric normal saline intragastrically; positive control group was given Magnesium isoglycyrrhizinate liquid intragastrically (13.5 mg/kg); G. uralensis water extract low-dose, middle-dose and high-dose groups were given G. uralensis water extract intragastrically 1 mL (120, 240, 480 mg/kg, by the amount of extract), qd, for consecutive 7 d. At 8th day of administration, model group, positive control group and G. uralensis water extract groups were given Triptolide liquid intragastrically (0.6 mg/kg) to establish acute liver injury model. HE staining was used to detect the pathological changes of liver tissue in rats. The activities of ALT and AST in serum were determined by enzyme coupling reaction. The protein expression of IL-10 and TNF-α in liver tissue were analyzed by Western blotting. RESULTS: Compared with blank control group, obvious pathological changes of liver tissue in rats were observed in model group; serum activities of ALT and AST, the protein expression level of TNF-α in liver tissue were increased significantly, while the protein expression level of IL-10 was decreased significantly (P<0.01). Compared with model group, pathological changes of liver tissue were relieved significantly. Except for the activity of AST in serum of rats in G. uralensis water extract low-dose group, the activities of ALT and AST in serum, the protein expression level of TNF-α in liver tissue were decreased significantly in G. uralensis groups, while the protein expression level of IL-10 in liver tissue was increased significantly (P<0.05 or P<0.01). CONCLUSIONS: G. uralensis water extract can relieve triptolide-induced acute liver injury in rats, the mechanism of which may be associated with expression down-regulation of pro-inflammatory factor TNF-α and expression up-regulation of anti-inflammatory factor IL-10.
