OBJECTIVE:To evaluate the influence of different process technics on the contents of catalpol in rehmannia root.METHODS:Dual wavelength TLC-scanning method was used to determine the contents of catalpol in different processed products of rehmannia root.RESULTS:There were significant differences in the contents of catalpol in the different processed products of rehmannia root and the crude herb.CONCLUSION:The contents of catalpol will be changed after processing with different processed methods and adding different supplementary materials.
2.A Novel Method for Assaying Enzymatic Activity of Ribosome-inactivating Protein Using Plasmid DNA as Substrate

A novel method for assaying the enzymatic activity of ribosome-inactivating proteins(RIPs) has been developed.The principle of the method is based on that RIP can remove some adenine bases from double-stranded supercoiled DNA molecules,subsequently,the deadenylated DNA was cleaved into nicked and linear form.After treatment with acidic aniline,the deadenylated DNA was degraded into many small fragments,and run out of the gel.The enzymatic activities of two RIPs(trichosanthin and cinnamomin) were tested using this method,the limit of sensitivity is about 50 ng(trichosanthin) and 5 ng(reduced cinnamomin) .It should be emphasized that the merit of this method is to avoid the preparation of ribosome.
3.Analysis of the death causes of 145 cases of renal transplant recipients

Objective To investigate whether the mortality and death causes of renal transplant recipients have changed from 1985 to 2002 in our center.Methods 145 cases of renal transplant recipients who died during 1985 to 2002 were divided into 3 groups: 1985 to 1990,1991 to 1996,1997 to 2002. The death causes and the mortality on the 1st ,2nd,3rd,4th and 5th year post transplant of each group were reviewed.Results During the three periods,the 1st,2nd,and 3rd year mortality was decreased. Infection as a cause of death fell from 31.8 % to 29.2 % and 26.7 % . Whereas death from liver disease,cancer and cardiovasvular diseases was increased.Conclusions The mortality is decreased. Infection,cardiovascular diseases,liver diseases and cancer were the main causes of death after transplantation from 1985 to 2002. It is important to prevent these diseases and treat them effectively in order to improve the recipients' survival rate.
4.Anti-oxidative and mitochondria protective effects of resveratrol on focal cerebral ischemia-reperfusion injury in rats

AIM: To investigate anti-oxidative and mitochondria protective effects of resveratrol on focal cerebral ischemia-reperfusion injury in rats. METHODS: Middle cerebral artery occlusion(MACO) was used to induce focal cerebral ischemia-reperfusion model.After 24 h reperfusion,MDA,GSH,NO,LD content and SOD activity in brain homogenate were determined,MDA,GSH content and SOD,ATPase activity in mitochondria were also determined. RESULTS: Resveratrol significantly inhibited the increase in MDA,NO,LD content and the decrease in SOD activity and GSH content in brain after cerbral ischemia-reperfusion.Resveratrol also significantly inhibited the increase in MDA and the decrease in activity of SOD,Na~+-K~+-ATPase,Ca~(2+)-Mg~(2+)-ATPase in mitochondria. CONCLUSION: Resvertrol has protective effects on focal cerebral ischemia-reperfusion injury via attenuating cerebral oxygen free radical lipid peroxidation and protecting mitochondria.
5.Effects of negative pressure wound therapy on the expression of EDA+FN in granulation tissues of human diabetic foot wounds

Objective To investigate the effects of negative pressure wound therapy (NPWT) on the expression of EDA+ FN in granulation tissues of human diabetic foot wounds.Methods Forty patients with diabetic foot wounds fitting the inclusion criteria,admitted from Jan.2014 to Jun.2016,were randomly and equally apportioned to receive either NPWT or conventional gauze therapy (control) for 14 days.Granulated tissue biopsies were collected before (0 day) and after (14 day) treatment in both groups.All biopsies were subdivided into two parts.One part was preserved in 4% paraformaldehyde for immunocytochemical staining of EDA+FN,and the other part was stored at-80 ℃ for Western blotting and PCR analysis of EDA+FN.Results The immunohistochemical analysis revealed that the mean area density of EDA+ FN increased in both NPWT group and control group at day 14 relative to day 0,but the change value of mean area density was higher in NPWT group than in control group (P<0.01).Western blotting showed that the relative protein levels of EDA+FN increased in both NPWT group and control group at day 14 relative to day 0,but the change value of relative protein levels of EDA+FN was higher in NPWT group than in control group (P<0.01).The real time PCR analysis demonstrated that the relative mRNA levels of EDA+ FN increased in both NPWT group and control group at day 14 relative to day 0,but the change value of relative mRNA levels of EDA+ FN was higher in NPWT group than in control group (P<0.01).The results demonstrated the higher protein and mRNA levels of EDA+FN in NPWT group than that in control group.Conclusion NPWT obviously enhances EDA+FN expression in granulation tissue of diabetic foot wound,as a result promotes wound healing.
6.Value of ultrasound in diagnosis and classification of pilomatricoma

Objective To explore the value of ultrasound in diagnosis and classification of pilomatricoma.Methods Ultrasonic features of 63 cases with pilomatricoma confirmed by operation and pathology were analyzed and summarized retrospectively.Results According to if there was calcification in the tumor,pilomatricoma cases were classified into two groups(calcific group and non-calcific group).In addition,there were three subgroups in the calcific group based on the size and disposition of calcification:subgroup Ⅰ,completely echogenic mass with strong posterior acoustic shadowing in the subcutaneous layer (n=17,26.98%);subgroupⅡ,hypoechoic nodules with tiny calcification diffused distributed(n=15,23.80%);subgroup Ⅲ,hypoechoic nodules with large calcification(n=8,12.70%).Twenty-three cases (36.51%)were distributed into non-calcific group.Conclusions Ultrasonography is valuable for diagnosis and differential diagnosis of pilomatricoma,and it is an essential examination method for the tumor before operation.
7.Value of ultrasound in dignosis and type of solid-pseudopapillary tumor of pancreas

Objective To study the sonographic features of solid-pseudopapillary tumors of the pancreas(SPTP) and to explore the value of ultrasound in the dignosis,differential dignosis and type of SPTP.Methods B-mode ultrasonography and color Doppler flow imaging(CDFI) of nine cases of SPTP confirmed by clinical and pathology were enrolled in the study and some cases were compared with the findings of computed tomography (CT).Results On B-mode ultrasound images,tumors showed solid,solid and cystic alternative distributions or cystic structures,presenting as round or oval shape,well-defined margins.Of these nine lesions,one was mainly composed of solid part,seven showed solid tumor with cystic component inside and one showed cystic tumor.All tumors were encapsulated.The encapsulation was completive in seven cases,incontinuous in one case and interruptive in one case.In CDFI,blood flow was found in five cases, three cases were I-brand,one case was II-brand,one case was III-brand.The peak systolic velocity ranged from 8.6 cm/s to 33.2 cm/s,three cases were less than 20 cm/s.Resistance index(RI) varied from 0.38 to 0.56 and three cases were lower than 0.5.According to the sonographic features of SPTP,combined with related reviews about the tumor,those masses were classified into three types as homogeneous solid,solid and cystic alternative distributions and cystic tumor.Conclusions The B-mode ultrasound and CDFI characteristic findings might be of great value in dignosis and differential dignosis of SPTP.

Objective\ To further clarify the biological effects of TGF?1 on blastocyst implantation. Methods We used female adult Kunming mice to mate with male for observing the distribution of TGF?1 and its receptor at different stages of implantation with immunohistochemical method. Results\ Compared with nonpregnancy,TGF?1 and its receptor immunostaining markedly increased in epithelial and glandual cells on day 4 of pregnancy.The moderate extracellular staining of TGF?1 was present around the fibroblasts.On day 5\|6 of pregnancy,strong immunostaining in intensity of TGF?1 and its receptor was noted in primary decidual zone.With blastocyst invasion,TGF?1 and its receptor immunostaining diffused in decidual cells. Conclusion\ TGF?1 might play an important role via its receptor during blastocyst implantation.\;

10.Tranexamic acid reduces blood loss in total knee arthroplasty:effectiveness and safety

BACKGROUND:Tranexamic acid is a synthetic anti-fibrinolytic drug, and can effectively control blood loss after total knee arthroplasty through vein. OBJECTIVE:To evaluate the effectiveness and safety of tranexamic acid in reducing blood loss after primary unilateral total knee arthroplasty. METHODS:From 2014 to 2015, 100 patients from the Department of Lower Limb, Sichuan Orthopaedic Hospital underwent primary unilateral total knee arthroplasty, and randomly divided into test group which used tranexamic acid and control group which used tranexamic acid, with 50 cases in each group. 1 g tranexamic acid was infused into the vein at 10 minutes before total knee arthroplasty in the test group, but tranexamic acid was not given in the control group. RESULTS AND CONCLUSION:The amount of total blood loss, postoperative wound drainage in 24 hours, and the amount of blood transfusion were significantly less in the test group than in the control group (P < 0.05). No significant differences in intraoperative blood loss and the number of patients receiving blood transfusion were found between the test and control groups. No significant difference in preoperative hemoglobin levels was detected before surgery, but hemoglobin levels were decreased after surgery between the two groups. Postoperative hemoglobin levels were apparently higher in the test group than in the control group after surgery. At 24 hours after surgery, no significant difference in coagulation was detectable between the two groups. At 6 days, double lower limb vein color Doppler ultrasound examination did not reveal deep vein thrombosis in both groups. These findings verify that treatment with tranexamic acid in the vein before primary unilateral total knee arthroplasty can evidently diminish the amount of perioperative blood loss and blood transfusion, reduce the risk and cost of blood transfusion, which is good for rehabilitation and does not increase the risk of deep vein thrombosis.
